5500 W Buffer G is needed for ligation sequencing on the 5500 W Series genetic analyzers. Supplemental buffers like this one can be used to replace components of the main buffer kit. 5500 W Buffer G contains sufficient reagents to run up to 432 cycles, or 72 cycles per lane for 6 lanes.

SOLiD™ Buffer E, Double (Applied Biosystems™)

The SOLiD™ Buffers are needed for ligation sequencing on the 5500⁄5500xl Genetic Analyzer. SOLiD™ Buffer E, Double (PN 4463016) consists of SOLiD™ Buffer E Part 1 and SOLiD™ Buffer E Part 2, which need to be mixed prior to running on the instrument. SOLiD™ Buffer E, Double contains sufficient reagents to run 950 to 1632 cycles, or up to 136 cycles per lane.

These kits are individually orderable components of the SOLiD™ Run Cycle Buffer Kit (PN 4461840). Supplemental buffers can be used to replace components of the main buffer kit. Each double size has sufficient buffer for covering two FlowChips.

SequenceRx Enhancer Solution A (Invitrogen™)

The SequenceRx Enhancer System is designed to improve the length and quality of sequencing data from difficult sequencing templates. It is a panel of seven co-solvents which are added individually to automated fluorescent DNA sequencing reactions. This system is not designed for use on long stretches of poly A.

The SequenceRx Enhancer System:

• Is useful on templates containing regions with secondary structure such as high GC-content, small nucleotide repeats, hairpins, direct repeats, and inverted repeats
• Requires no change in protocols-the SequenceRx Solution replaces part of the water added to the reaction mixture
• Is compatible with all sequencing chemistries and platforms
• Uses one reaction to determine the sequence for all four nucleotides (100 reactions equal 100 templates)

SOLiD™ FlowChip Deposition Buffer 1 (Applied Biosystems™)

The SOLiD™ FlowChip Deposition Buffer 1 is required for depositing templated beads onto a SOLiD™ FlowChip for sequencing on the 5500⁄5500xl Genetic Analyzer.

Store the SOLiD™ FlowChip Deposition Buffer 1 at 2°C to 8°C until ready for use.

SOLiD™ Buffers are used for ligation sequencing on the 5500⁄5500xl Genetic Analyzers. The SOLiD™ Run Cycle Buffer Kit (Double) contains a set of six buffers required during all types of sequencing runs for ligation sequencing in the forward and reverse directions. The kit consists of SOLiD™ Buffers A, B, C, E, F, and G.

SOLiD™ Buffers A, B, and E are provided in two parts, and components must be mixed before use. SOLiD™ Buffer F is provided as a 10X solution and requires dilution to 1X before pouring into its reagent tank. SOLiD™ Buffer G is poured into its reagent tank before use.

The kit contains sufficient reagents to run 950 to 1632 cycles, or up to 136 cycles per lane.
