Mixtures of body fluids and/or tissues from a male and female are frequently encountered with sexual assault casework and other violent crimes. These mixed samples often contain trace quantities of male DNA present in a large background of female DNA. Typical samples can include vaginal swabs containing a small number of sperm cells, body swabs from female victims of oral assaults, and fingernail scrapings/clippings from female victims of violent assaults. Standard autosomal STR analysis may be unable to detect the minor male contributor in these types of samples, while the Yfiler® kit can routinely detect a minor male contributor present at less than 1:1000.

The AmpFLSTR® Huaxia™ PCR Direct Amplification Kit employs a validated "direct" amplification chemistry designed specifically for the China forensic community to use with DNA database samples and other single source DNA samples. Optimized for casework, the AmpFLSTR® Identifiler® Plus PCR Amplification Kit enables forensic analysts to recover more interpretable results from a wider range of DNA evidence samples with increased confidence. The new kit includes the same primers and allelic ladder as the widely used Identifiler® Kit and harnesses next-generation PCR amplification technology to help provide enhanced sensitivity, a cleaner baseline, improved performance on mixtures, and the ability to overcome high levels of PCR inhibition.

Features of the AmpFLSTR® Identifiler® Plus PCR Amplification Kit include:

Table 1: As part of its rigorous development, the Identifiler® Direct Kit was field tested by several medium to high-throughput DNA databasing laboratories to assess "first pass" success rate. Combined results from external test site laboratories are shown below. 1.2 mm punches from blood and buccal samples on FTA® Cards and Buccal DNA Collectors™ were amplified with the Identifiler® Direct Kit, and the resulting data was analyzed using GeneMapper® ID-X, at two different minimum peak amplitude thresholds.

Configured for Success
Utilizing our expertise in 5-dye and mobility modifier technology, 17 loci are accommodated in a single amplification while maintaining optimum spacing (Figure 2). This permits unambiguous allele designation. All amplicons fall below 450 base pairs (bp) to maximize performance across a wide range of forensic samples. The majority of loci are concentrated below 200 bp, enabling recovery of a greater number of alleles from challenging degraded samples.
