Microsatellites, also referred to as short tandem repeats (STRs), are commonly used for parentage testing, identification, and forensics due to their informativeness and Mendelian inheritance,. The Canine ISAG STR Parentage Kit (2014) provides all of the necessary reagents for simultaneous amplification of the 22 ISAG-recommended microsatellite loci in a single multiplex PCR reaction. Amplicons are sized using capillary electrophoresis on Applied Biosystems genetic analyzers with GeneMapper software to determine each animal’s unique genetic profile. Canine control DNA is provided to verify acceptable PCR and electrophoresis conditions.

The kit reagents and protocols have been optimized to deliver similar peak heights (amplification yields) for each loci and sufficient size and/or fluorescent separation between markers to ensure accurate marker discrimination (Figure 1). Furthermore, allele callings obtained with this kit represent the true alleles of an individual, instead of the 'plus-A peaks' or 'split peaks' typically interpreted when using a conventional TaqDNA polymerase, due to the incorporation of the Phusion Hot Start DNA proofreading polymerase. The StockMarks® for Dogs Canine Genotyping Kit is part of a complete solution for animal parentage typing and genotyping employing short tandem repeats (STRs), also referred to as microsatellites. The CLA (Cell Line Authentication) IdentiFiler Direct PCR Amplification Kit simplifies the amplification step during authentication of cell lines. The kit enables direct amplification of the same 16 loci included in our AmpFLSTR Identifiler Direct kit, eliminating the need to perform DNA extraction or purification steps for DNA database and other single-source reference samples. Blood or buccal samples on FTA cards can be punched into PCR plates or tubes, and taken directly to PCR amplification, without reduction in data quality. Samples collected using a Buccal DNA Collector or other non-FTA substrate can be processed without heat incubation or additional workflow steps using Prep-n-Go Buffer (available separately).
