Dynabeads™ Mouse T-Activator CD3/CD28 for T-Cell Expansion and Activation (Gibco™)

Dynabeads® Mouse T-Activator CD3/CD28 are 4.5 µm superparamagnetic beads with anti-CD3 and anti-CD28 antibodies covalently bound to the bead surface. Dynabeads® Mouse T-Activator CD3/CD28 is designed for the activation and expansion of mouse T cells for research purposes.

Simple method for T cell activation
Dynabeads® Mouse T-Activator CD3/CD28 offers a simple method for activation and expansion of T cells that does not require feeder cells (antigen-presenting cells) or antigen. The 4.5 µm diameter, inert, superparamagnetic beads are similar in size to antigen-presenting cells, and are covalently coupled to anti-CD3 and anti-CD28 antibodies. These two antibodies provide primary and co-stimulatory signals, optimized for efficient T cell activation and expansion. Expansion of the T cell population can be stimulated using recombinant IL-2, and after activation or expansion, the magnetic beads can be easily removed using a DynaMag™ magnet.

Starting samples
Start with single-cell suspensions from spleen or lymph nodes, mononuclear cells (MNCs), whole blood, or T cell subsets including CD3+ T cells, CD4+ or CD8+ T cells, or regulatory T cells. Learn more about all of our Dynabeads® cell isolation products.

Downstream applications
The activated T cells can be analyzed shortly after activation (for transfection/transduction or to study TCR signaling, proteomics, gene expression, etc.). T cells can be left in culture to differentiate into T helper cell subsets or expansion of polyclonal/antigen-specific T cells.

About Dynabeads® Human T-Activator CD3/CD28
Dynabeads® Human T-Activator CD3/CD28 offer a simple method for activation and expansion of T cells that does not require feeder cells (antigen-presenting cells) or antigen. The uniform 4.5 µm diameter, inert, superparamagnetic beads are similar in size to antigen-presenting cells, and are covalently coupled to anti-CD3 and anti-CD28 antibodies. These two antibodies provide primary and co-stimulatory signals, optimized for efficient T cell activation and expansion. Expansion of the T cell population can be stimulated using recombinant IL-2, and after activation or expansion, the magnetic beads can be easily removed using a DynaMag™ magnet. For expansion of antigen-specific T cells from T cell clones or T cell lines, we recommend using Dynabeads® Human T-Activator CD3/CD28/CD137.

CTS™ (Cell Therapy Systems) Dynabeads™ CD3/CD28 (Gibco™)

CTS™ Dynabeads™ CD3/CD28 are intended for ex vivo isolation, activation, and expansion of human T-cells in translational research. The technology—previously known as Xcyte™ Dynabeads™ or Dynabeads™ ClinExVivo™ CD3/CD28—has been used in a number of clinical studies. By combining anti-CD3 and anti-CD28 antibodies on Dynabeads, the beads provide both the primary and co-stimulatory signals that are required for activation and expansion of T cells. CTS Dynabeads CD3/CD28 are manufactured and controlled according to ISO 9001:2000 and ISO 13485:2012 requirements. A Device Master File is held with the FDA for cross-referencing in IND applications, and a CoA is available on request. Dynabeads™ Human T-Activator CD3/CD28/CD137 (Gibco™)

Dynabeads® Human T-Activator are coupled with anti-CD3, anti-CD28, and anti-CD137 antibodies. CD137 (4-1BB) is a member of the tumor necrosis factor family. Agonistic anti-CD137 antibody acts as an activating costimulatory molecule especially important for effector/memory T cells and promotes the survival and proliferation of T lymphocytes.

Dynabeads® Human Treg Expander is intended for activation and expansion of human regulatory T-cells (Treg cells) after isolation with the Dynabeads® Regulatory CD4+CD25+ T Cell Kit (Cat. No. 11363D).

Dynabeads® Human Treg Expander offers a simple method for activation and expansion of Treg cells that does not require antigen presenting cells or antigen. Dynabeads® Human Treg Expander are uniform 4.5 µm, superparamagnetic beads coupled with an optimized mixture of anti-CD3 and anti-CD28 monoclonal antibodies. By combining anti-CD3 and anti-CD28 antibodies on Dynabeads®, the beads will provide both the primary and co-stimulatory signals that are required for activation and expansion of the Treg cells.

Starting Sample
Human regulatory T-cells isolated with the Dynabeads® Regulatory CD4+CD25+ T Cell Kit (Cat. No. 11363D).

CTS™ Dynabeads™ Treg Xpander (Gibco™)

Optimized for the ex vivo activation and expansion of regulatory T cells (Tregs), CTS Dynabeads Treg Xpander provides more specific and higher fold expansion compared to our CTS Dynabeads CD3/CD28. Tregs activated and expanded with this product show suppressive activity and display the characteristic Treg cytokine profile and phenotype. Furthermore, high stability of the expanded Tregs has been demonstrated through analysis of the Treg-specific demethylated region.

Regulatory T cells (CD4+ CD25+ FOXP3+) are a suppressive subset of CD4+ T helper cells and are important for the regulation of immune responses. Due to a relative frequency of 1–2% of the total lymphocyte population, ex vivo expansion of Tregs prior to adoptive transfer is expected for most clinical applications.
