TaqMan™ TAMRA Probe (Applied Biosystems™)

Applied Biosystems TaqMan TAMRA probes are dual-labeled probes used for real-time PCR applications using TaqMan chemistry. TaqMan TAMRA Probes feature a 5’ fluorescent reporter dye (FAM, VIC, or TET) and 3’ fluorescent quencher (TAMRA dye). TaqMan TAMRA probes, which were some of the first TaqMan probes to be developed, can be used for a variety of applications.

TAMRA is a fluorescent dye and can be multiplexed with up to three dyes. (If multiplexing higher than triplex, TaqMan QSY probes are recommended.) TAMRA probes are typically longer (30 to >40 bases). For enhanced specificity, shorter TaqMan MGB nonfluorescent quencher (NFQ) probes (generally 15 to 20 bases) are also available.

All TaqMan TAMRA probes are HPLC-purified and verified by mass spectrometry to help ensure lot-to-lot reproducibility between batches. TaqMan™ MGB Probe (Applied Biosystems™)

Applied Biosystems TaqMan MGB (minor groove binder) probes are dual-labeled probes used for real-time PCR applications using TaqMan chemistry. TaqMan MGB Probes incorporate a 5' fluorescent reporter dye and a 3' nonfluorescent quencher (NFQ). The NFQ offers the advantage of lower background signal, which results in better precision in quantitation. Custom TaqMan MGB probes are HPLC-purified and available with FAM, VIC, TET, and NED reporter dyes.

Shorter, more specific probes
TaqMan MGB probes include a MGB moiety at the 3’ end that increases the melting temperature (Tm) of the probe and stabilizes probe/target hybrids. This means that TaqMan MGB probes can be significantly shorter than traditional probes, providing better sequence discrimination and flexibility to accommodate more targets.

Nonfluorescent quencher (NFQ) maximizes sensitivity
TaqMan MGB probes incorporate an NFQ to absorb (quench) signal from the fluorescent dye label at the other end of the probe. The properties of the NFQ combined with the short length of the MGB probe result in lower background signal than with non-MGB NFQ probes. Lower background means increased sensitivity and precision in your data.

TaqMan™ Copy Number Assay (Applied Biosystems™)

Applied Biosystems TaqMan Copy Number Assays use gold-standard TaqMan MGB probe chemistry to evaluate the copy number of genomic DNA targets using Applied Biosystems real-time PCR instruments and software. TaqMan Copy Number Assays are run together with a TaqMan Copy Number Reference Assay in a duplex qPCR reaction; the copy number assay detects the target sequence, and the reference assay detects a sequence that is known to be present in two copies in the diploid genome. The results are then analyzed by the relative quantitation method using Applied Biosystems CopyCaller Software.

The predesigned TaqMan Copy Number Assay collection consists of human and mouse assay libraries for copy number variation (CNV) analysis. The human assay collection includes over 1.6 million assays targeting gene exons and introns, extragenic regions, and CNV sequences from the Database of Genomic Variants (DGV). TaqMan™ MicroRNA Assay (Applied Biosystems™)

Applied Biosystems TaqMan MicroRNA Assays quantitate miRNAs with the specificity and sensitivity of TaqMan Assay chemistry. A simple, two-step protocol requires only reverse transcription with an miRNA-specific primer, followed by real-time PCR with TaqMan probes. TaqMan MicroRNA Assays are:
