Triggering Receptor Expressed On Myeloid Cells 2 (TREM2) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in analysis of protein structure. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant human protein is expressed from a DNA sequence encoding the extracellular domain of human TREM2 (NP_061838.1) (Met 1-Ser 174) fused to a polyhistidine tag at the C-terminus.

Activity: The activity of this protein has not been tested.

Formulation: lyophilized in PBS, pH7.4, 5% mannitol, 5% trehalose, 0.02% Tween®-80.

Reconstitution: dissolve the protein in sterile double-distilled water to a concentration of 0.2 mg/mL or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Background
Triggering receptor expressed on myeloid cells 2 ( TREM2 ) is a single Ig domain receptor. It is expressed on macrophages and dendritic cells but not on granulocytes or monocytes. TREM2 may play a role in chronic inflammations and may stimulate production of constitutive rather than inflammatory chemokines and cytokines. TREM2 forms a receptor signaling complex with TYROBP and triggers activation of the immune responses in macrophages and dendritic cells. It also associates with the signal adapter protein, DAP12, which has a cytoplasmic ITAM, leading to the subsequent activation of cytoplasmic tyrosine kinases. TREM2 is both required and sufficient for competent uptake of apoptotic neuronal cells. TREM2 and TREM2-L form a receptor-ligand pair connecting microglia with apoptotic neurons, directing removal of damaged cells to allow repair. Deficiency of the adapter protein DAP12 or its associated receptor TREM2 is associated with abnormal osteoclast development in humans. Defects in TREM2 are causes of PLOSL, also known as NHD. In addition, TREM2 signaling is also an important pathway to promote healing.

CTLA4 Recombinant Mouse Protein, His Tag

Cytotoxic T-Lymphocyte-Associated 4 (CTLA4), recombinant mouse protein is supplied as a lyophilized powder. It is suitable for use in analysis of protein structure. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant protein was expressed from a DNA sequence encoding the extracellular domain of mouse CTLA4 (NP_033973.2) (Met 1-Phe 162) fused to a polyhistidine tag at the C-terminus.

Activity: Measured by its binding ability in a functional ELISA.
1. Immobilized recombinant mouse CTLA4-His (Cat:50503-M08H) at 10 µg/ml (100 µl/well) can mouse CD80-Fc (cat:50446-M02H) with a linear range of 0.78-62.5 ng/ml.
2. Immobilized recombinant mouse CTLA4-His (Cat:50503-M08H) at 10 µg/ml (100 µl/well) can human B7-1-Fc (Cat:10698-H03H) with a linear range of 0.78-125 ng/ml..
