Pierce™ Protein A/G IgG Binding Buffer (Thermo Scientific™)

Thermo Scientific Pierce IgG Binding and Elution Buffers ensure high-yield, nondenaturing antibody purification with IgG affinity purification supports, such as Protein A and Protein G agarose beads. The Protein A/G IgG Binding Buffer is a phosphate-based buffer containing EDTA, formulated for antibody purification using either Protein A or Protein G.

These products include IgG Binding Buffers specific for each immunoglobulin-binding protein (Protein A, Protein G, Protein A/G). The Pierce IgG Elution Buffer provides efficient dissociation and elution of antibodies from all three IgG-binding proteins, including Protein L. This low-pH elution buffer is also effective for most antibody-antigen or protein-protein affinity purification systems.

The Gentle Ag/Ab Binding and Elution Buffer system uses near-neutral, high-salt conditions for elution, enabling even sensitive and labile antibodies (or other proteins) to be eluted from an affinity system without denaturation and inactivation. The phosphate-free binding buffer in this system is a necessary substitute for the Protein A binding buffer (pH 8), which precipitates with the Gentle Ag/Ab Elution Buffer.

Pierce™ Saturated Ammonium Sulfate Solution (Thermo Scientific™)

Thermo Scientific Pierce Saturated Ammonium Sulfate Solution is formulated and ready to use for simple immunoglobulin, antibody, or other protein fractionation applications based on selective precipitation, which is also called salting out.

Features of Saturated Ammonium Sulfate Solution:

Convenient—solution is ready to use; no preparation required
Concentrates—recover antibody in 100 mL from more than 1L of cell culture supernatant
Improves purity—partial-purification of antibodies by ammonium sulfate precipitation greatly improves the final purity that can be obtained with affinity purification
Extends column life—using pre-purified antibody samples instead of crude samples enables affinity columns to be used for many more cycles
Compatible—ammonium sulfate is easily removed by dialysis or desalting columns, eliminating any obstacles to downstream methods
Versatile—use for precipitating immunoglobulins or "salting out" fractions of other sample types

The saturated ammonium sulfate solution (4.32M) enables removal of abundant proteins from serum, cell culture supernatant and ascites fluid. Slowly adding the solution to a protein sample results in decreased protein solubility and eventual protein precipitation as the charged ammonium sulfate ions compete with protein for water interaction. High molecular weight proteins, such as antibodies, are among the first to precipitate. When used in combination with antibody purification methods such as affinity chromatography, ammonium sulfate precipitation is ideal for achieving highly purified antibody samples.
