Vacuum Hydrolysis Tubes are precision-designed for use in protein hydrolysis and subsequent analysis of protein and peptide amino acid compositions. The heavy-walled borosilicate glass construction ensures complete compatibility with all commonly used acid hydrolysis reagents. Acid hydrolysis at elevated temperatures, in vacuo, will hydrolyze most proteins and peptides to their amino acid components without significant decomposition.

Vacuum hydrolysis methods commonly use 6N constant boiling hydrochloric acid (HCl), a mixture of HCl and propionic acid, 3N mercaptoethanesulfonic acid or 4N methanesulfonic acid. The constant boiling HCl method uses standard protein hydrolysis at 105 to 110°C for 16 to 24 hours. Using the HCl/propionic acid method, hydrolysis times can be shortened to 15 minutes at 160°C. Tryptophan is easily degraded by acid hydrolysis in 6N HCl. The presence of oxygen, heavy metals and carbohydrates are contributing factors to this degradation. A 22-hour hydrolysis method was developed to improve tryptophan recovery from protein hydrolysis samples by using 3N mercaptoethanesulfonic acid. A second method to improve tryptophan recovery involves hydrolysis at 115°C using 4N methanesulfonic acid.

PureLink™ Viral Collection Tubes (Invitrogen™)

The PureLink® Viral RNA/DNA Mini Kit is a nucleic acid purification system designed for fast and easy isolation of viral RNA or DNA from cell-free samples, such as serum, plasma, and cerebrospinal fluid. Starting sample volumes can be as high as 500 µl and elution volumes as low as 10 µl, allowing for a 50-fold concentration of nucleic acids. The higher target concentration obtained using the PureLink® Viral RNA/ DNA Mini Kit provides greater sensitivity and accuracy in downstream detection procedures such as real-time PCR or endpoint analysis. Specially formulated buffers enable the purification of RNA or DNA, allowing one system to be used for all your virus extraction samples. In addition, the kit provides sufficient quantities of lysis buffer to address large starting sample volumes.

Ct values were consistently lower for samples prepared with the PureLink® Viral RNA/DNA Mini Kit (Figure 2), indicating greater sensitivity.

In addition, the quantitation was accurate down to 20 pfu compared to samples prepared with the kit from supplier Q, which leveled off at 200 pfu. Results indicate an improvement in the recovery of viral RNA as well as greater overall consistency of quantitative amplification.

Lower Ct values were obtained with DNA prepared with the PureLink ™ Viral RNA/DNA Mini Kit as compared to those obtained using the kit from supplier Q (Figure 3), indicating greater sensitivity of detection of viral DNA. The data demonstrate that the PureLink® Viral RNA/DNA Mini Kit can recover DNA down to 20 pfu at significantly lower Ct values compared to the competitor’s kit.

The PureLink® Viral RNA/DNA Mini Kit yielded target molecules of consistently lower Ct values compared to the EZ1 Kit; in some cases as much as 4 cycles earlier (Figure 4). The arrows indicate samples that failed to produce a Ct value. In total, 14 out of 48 samples failed to produce a Ct when processed on the BioRobot EZ1 (29%), while the PureLink® Viral RNA/DNA Kit failed to provide a Ct value in only 3 samples (6%), of which both replicates extracted with the EZ1 Kit also failed to produce a Ct value.

For enhanced viral RNA or DNA detection, the PureLink® Viral RNA/DNA Mini Kit has been demonstrated to give lower Ct values and increased nucleic acid recovery when compared to competitor products using volumes recommended by the supplier. The flexibility of the PureLink® Viral RNA/DNA Mini Kit with respect to sample volume, elution volume, and virus type makes it the best solution for purifying viral RNA or DNA.

Contents and storage:
The PureLink® Viral RNA/DNA Mini Kit contains Mini Spin Columns in Collection Tubes, Collection Tubes, Recovery Tubes, Lyophilized Carrier RNA, Proteinase K, Lysis Buffer, 5X Wash Buffer II, and RNase-free Water. Each kit contains sufficient material for 50 purifications. The kit is shipped at room temperature; upon receipt, store the lyophilized carrier RNA at -20°C and the proteinase K at 4°C. Stored as directed, the kit is stable for 1 year.

Tempus™ Blood RNA Tube (Applied Biosystems™)

How it works
Each Tempus™ Blood RNA Tube contains 6 mL Stabilizing Reagent. When the blood (3 mL) is drawn into the tube and mixed with this reagent, lysis occurs almost immediately. The stabilizing reagent inactivates cellular RNases and selectively precipitates RNA; genomic DNA (gDNA) while proteins remain in solution. Blood drawn into the Tempus™ Blood RNA Tube is stable for up to five days at room temperature, or minimally seven days at 4°C.

Accessory product
When the Tempus™ Blood RNA Tubes are used with the Tempus™ 12-Port RNA Isolation Kit (Cat. No. 4378672), RNAs with greater stability and higher yields of RNA are produced (see figure).

Phasemaker™ Tubes (Invitrogen™)

Invitrogen™ Phasemaker™ Tubes contain a thick liquid polymer that separates and completely isolates the upper aqueous phase of a TRIzol® Reagent mix from the organic phase underneath, greatly simplifying removal of the upper phase containing the RNA.

The polymer in Phasemaker Tubes is heavier than the aqueous phase of the TRIzol Reagent mix, but lighter than the organic phase, so after centrifugation it positions itself between these two layers. As a thick polymer, it creates a tight seal between the two phases, easing removal of the aqueous phase and helping to reduce the risk of carrying over contaminants from the organic layer, thereby increasing RNA recovery up to 30%.

The polymer in the Phasemaker Tubes is inert and does not interfere with standard RNA applications. Isolated RNA can be used in RT-PCR, Northern blot analysis, dot blot hybridization, poly(A)+ selection, in vitro translation, RNase protection assay, and molecular cloning.

Phasemaker Tubes accommodate RNA extraction using TRIzol Reagent, TRIzol LS Reagent, and TRIzol Plus Purification Kit only. They are not recommended for use with other organic reagents as the density of those reagents might not be suitable. Phasemaker Tubes are not for use in DNA or protein isolation, but are intended for RNA extraction only.

Phasemaker Tubes work with fresh or frozen samples, including tissue and blood. They also work with samples previously treated with RNAlater™ RNA stabilization solution. Phasemaker Tubes are already pre-dispensed; the tube size is 2 mL.
