Thermo Scientific Pierce Nickel Coated Black 96-Well Plates provide a simple format to bind His-tagged fusion proteins for ELISA and other plate-based assays involving protein interactions of expressed recombinant proteins. These black plates can be used with fluorescent detection methods.

Features of Pierce Nickel Coated Plates:

• Ni2+ activated surface enables metal-chelate binding of His-tagged proteins
• Detergents used to lyse cells don't inhibit binding to activated plates as they do with plain polystyrene
• Better binding for sensitive assays compared to other commercially available nickel-activated plates
• Detection limit: 1ng of polyhistidine fusion protein
• Binding capacity: approx. 9 pmol His-tagged protein (27kDa) per well

These Ni(2+) chelate-coated plates are ideal for analyzing polyhistidine-tagged fusion proteins by ELISA-based methods. Proteins that contain a succession of several histidine residues at the amino or carboxyl terminus have a strong binding affinity for metal. Bacterial lysates containing polyhistidine-tagged fusion proteins can be added directly to the plates without the need for blocking.

These Thermo Scientific Pierce Maleimide Activated Black, 96-Well Plates are ideal for binding sulfhydryl-containing molecules that are difficult to coat onto polystyrene plates, such as peptides that contain a terminal cysteine. Our coated plates are an especially useful tool for assessing specific anti-hapten antibody titers during antibody production.

Features of Maleimide Activated Black, 96-Well Plates:

• Black, for fluorescent assays
• Pre-blocked to reduce nonspecific binding
• Easy (spontaneous) immobilization of peptides derivatized with a terminal cysteine and proteins with free sulfhydryl
• Binding Capacity: 100 to 150 pmol of a sulfhydryl-containing peptide (307 Da) per well
• Activation Level: 100 µL

Maleimides react with free sulfhydryl group(s) at pH of 6.5-7.5, forming stable thioether linkages. Reaction with amines becomes significant at pH > 7.5. Some sulfhydryl-containing peptides and proteins may oxidize in solution and form disulfide bonds, which cannot react with maleimides. Disulfide bonds can be reduced to produce free sulfhydryls. The TCEP Disulfide Reducing Gel enables peptide or protein reduction while recovering the sample in the absence of reducing agents. Sulfhydryls can be introduced via amine modification using N-succinimidyl S-acetylthioacetate (SATA) or 2-iminothiolane-HCl (Traut's Reagent).

NeutrAvidin Protein has the additional advantage of lacking the bacterial RYD sequence found on streptavidin, which eliminates off-target binding to the RGD binding domain of adhesion receptors present in a variety of cells.

Thermo Scientific Pierce Glutathione Coated Plates are clear, 96-well polystyrene strip plates whose wells have been coated with intact glutathione (GSH) and pre-blocked for use in capture and detection assays involving glutathione-S-transferase (GST) fusion proteins. These glutathione coated plates can be used in the analysis of cell lysates to determine the presence and concentration of the GST-fusion protein produced in an expression system. Alternatively, the plates can be used to capture and coat fusion proteins via their GST tag for subsequent binding assays involving the recombinant protein (either protein interactions or a specific antibody). Because the plates are pre-blocked, initial purification of the cell lysates is not necessary.

Thermo Scientific Pierce Protein A Coated Plates provide an alternative to direct, passive adsorption methods for immobilizing antibodies for ELISA and other plate-based assay techniques.

These plates are uniformly and stably coated with Protein A, and enable antibodies or pre-established antibody-antigen complexes to be captured to wells of the polystyrene plate for subsequent detection in plate readers. The strategy is useful for a variety of assay situations, but it is generally not suitable for sandwich-type ELISA.
