Attune™ NxT No-Wash No-Lyse Filter Kit

The Attune® NxT No-Wash No-Lyse Filter Kit enables configuration of the Attune® NxT Acoustic Focusing Cytometer with an additional side scatter channel off of the violet laser, allowing no-lyse no-wash immunophenotyping of whole blood. The Attune® NxT cytometer is quickly and easily configured to collect violet side scatter by changing dichroic and emission blades.

Enables no-wash/no-lyse application
Immunophenotyping of whole blood using traditional flow cytometry is typically performed with lysis of the red blood cells (RBCs) followed by a wash step. These steps can result in significant cell loss and damage.

While a no-lyse, no-wash method for immunophenotyping of whole blood has been previously described, the method is of limited utility in traditional flow cytometry, which use hydrodynamic forces to orient cells in the flow stream. Moreover, significant sample dilution is required in no-lyse, no-wash methods to achieve low coincidence with red blood cells, necessitating extremely long acquisition times.

With the Attune® NxT cytometer you can acquire data quickly, even from dilute samples, using either a fluorescent reagent as a trigger to eliminate red blood cells or a differential side scatter using both blue and violet scatter. This kit enables the side scatter approach.

Attune™ NxT Automation Bundle, stacks/hotel (Invitrogen™)

The Attune NxT Automation Bundle consists of an Orbitor RS2 Microplate Mover and a barcode reader for quick and easy plate identification. This Orbitor RS2 Microplate Mover is an automation robot designed for use with the Attune NxT Acoustic Focusing Cytometer that enables flow cytometry in automated environments for increased sample throughput. It is offered in three different configurations: one with sequential access stacks for plate storage and handling (Cat. No. A33007), one with random access hotels (24 plates/3 hotels) (Cat. A33008), and one with two sequential access stacks and one random access hotel (this page).

Extensive vertical reach allows the Orbitor RS2 Microplate Mover to access virtually any microplate instrument location, including stacked instruments, and its telescoping arm can reach a variety of distances. The ingenious arm travels through the base, resulting for rapid and efficient movements. This robot has a full 360°workspace with unlimited base rotation; this eliminates the need for hard stops and reverse movements.

Advanced closed-loop motion control provides collision detection and position monitoring, allowing the Orbitor RS2 Microplate Mover to quickly resume without the need to re-home. With a wide range of grip-control functionality, this robot combines performance with innovation. From variable force control to plate detection, the feature set is designed to impress. The result is remarkable speed, precise motion, and consistent, accurate plate placement.

The Orbitor RS2 Microplate Mover is powered by Thermo Scientific Momentum Integration Software with features such as Momentum scheduling software, error detection and recovery, and a dynamic scheduler that allows changes while running.

Attune™ NxT Violet Laser Upgrade Kit

The Attune® NxT Violet Laser Upgrade Kit is designed to expand the 405 nm excitation capabilities of the Attune® NxT Acoustic Focusing Cytometer. Given the large array of labels, dyes, and applications to choose from, the ability to upgrade your instrument is key. The Violet Laser upgrade kit enables the detection of many stem cell side populations using Vybrant® DyeCycle™ violet, Pacific™ Blue, green, and orange dyes, as well as many Qdot® Nanocrystals and violet excitable polymer dyes.

This is a refurbished Attune NxT Acoustic Focusing Cytometer, which is an easy-to-use instrument designed to detect cell size and granularity, as well as fluorescence expression of labeled antibodies, proteins, and functional reagents. The instrument's modular design accommodates the most common fluorophores and fluorescent proteins used in flow cytometry enabling alignment with previous experimental results and accommodating future needs through simple upgrades. Acoustic focusing improves efficiencies compared to traditional flow cytometers
Traditional flow cytometers rely on a large volume of fluid to focus cells prior to analysis, resulting in extensive sample preparation prior to analysis and introducing the possibility of error due to artifacts created during the sample preparation process. With the Attune NxT Acoustic Focusing Cytometer, blood samples can be run without being manipulated using the No-Wash No-Lyse blood cell assay. The instrument also offers flexibility in the sample concentration that can be analyzed.

Attune™ NxT Automation SmartStor Sample Management System (Invitrogen™)

The Attune NxT Automation SmartStor Sample Management System is an accessory optimized for use in an automation line with the Attune NxT Acoustic Focusing Cytometer, autosampler, and Orbitor microplate mover. The SmartStor system’s presentation nest on the left-hand side allows the Orbitor arm to pick and load plates onto the autosampler. The SmartStor system maintains temperature from 4 to 40°C and holds up to 20 standard-height microplates. A 9-position hotel for deep-well plates is available for separate purchase (Cat. No. A38244). The SmartStor system comes with a 20-plate stack that is easily removed and replaced with the 9-position hotel if storage of deep well plates is needed.
