ELF™ 97 Phosphatase Substrate (ELF™ 97 Phosphate), 0.2 µm Filtered (Invitrogen™)

The ELF® 97 phosphatase substrate upon hydrolysis produces a bright and photostable yellow-green fluorescent precipitate at the site of enzymatic activity. This fluorescent precipitate has several unique spectral characteristics, including an extremely large Stokes shift that makes it easily distinguishable from endogenous fluorescence.

GFP-EIF2A

EIF2A has been expressed as a fusion with GFP (Green Fluorescent Protein) to act as a physiological substrate for the EIF2AK family of kinases including PERK (EIF2AK3) and PKR (EIF2AK2).

Fluorescein-MAP2K1

Inactive MAP2K1 (MEK1) has been labeled with fluorescein, and is a substrate for MAPK family members.

For use in combination with the LanthaSreen™ Tb-MAP2K1 [pSer217/221] Antibody (PV4813) to evaluate inhibitors in medium to high throughput screening applications.

PI(3,4)P2 Lipid Kinase Substrate (Invitrogen™)

The PI(3,4)P2 Lipid Kinase Substrate has been specifically optimized for use with Type I PIP kinases (e.g., PIP5K1A, PIP5K1B, PIP5K1C). The substrate consists of solubilized phosphatidylinositol 3,4-bisphosphate (PI(3,4)P2) in an aqueous buffer, and is used in combination with our Adapta™ Universal Kinase Assay to help identify unique kinase inhibitors.

Fluorescein-IKK Substrate

A fluorescein-labeled peptide substrate that is primarily used with the IkappaB kinase (IKK) family.

For use in combination with the LanthaScreen® Tb-IkappaBalpha [pSer32] antibody (PV3562) to evaluate inhibitors in medium to high throughput screening applications.

CHKtide

A peptide substrate based upon CDC25C that is phosphorylated by CHEK1 and other kinases. For use with the Adapta™ Universal Kinase Assay (PV5099).

DiFMUP (6,8-Difluoro-4-Methylumbelliferyl Phosphate) (Invitrogen™)

DiFMUP is a fluorinated 4-methylumbelliferyl phosphate (MUP) derivative that has a lower pKa than that of MUP, making DiFMUP an excellent substrate for continuously assaying acid phosphatases at low pH. The reaction product of DiFMUP has excitation/emission maxima of ~358/450 nm. DiFMUP is also available packaged in ten vials of 10 mg each (D-22065).

Fluorescein-Poly GT

A general fluorescein peptide substrate consisting of a polymer of glutamate, and tyrosine that is used to monitor tyrosine kinase activity.

For use in combination with the LanthaScreen® Tb-pY20 Antibody (PV3552) to evaluate inhibitors in medium to high throughput screening applications.

Alexa Fluor™ 647-Poly-GT Peptide Substrate

A general AlexaFluor647-labeled peptide substrate consisting of a polymer of glutamate, and tyrosine that is used to monitor tyrosine kinase acitivty.

For use in combination with the LanthaScreen® Eu-PY20 Antibody (PV5696) to evaluate inhibitors in medium to high throughput screening applications.

PI:PS Lipid Kinase Substrate

Specifically optimized for use with the Class III PI3 kinase (PIK3C3 (hVPS34)) and Type III PI4 lipid kinases, this substrate can also be used as a general substrate for the Class I lipid kinases. The lipid vesicles consist of a 1:9 molar ratio of phosphatidylinositol (PI) to phosphoserine (PS).

For use with the Adapta™ Universal Kinase Assay (PV5099).

Fluorescein-ERM (LRRKtide)

A fluorescein-labeled peptide substrate based upon ezrin/radixin/moesin (ERM) that is phosphorylated by LRRK2 and LRRK2 mutants.

For use in combination with the LanthaScreen® Tb-anti-ERM (pLRRKtide) Antibody (PV4899) to evalute inhibitors in medium to high throughput screening applications.

Fluorescein-CREBtide Substrate

A fluorescein-labeled peptide substrate that is based upon the CREB ( cAMP responsive element binding) protein. The peptide substrate is phosphorylated by a wide range of Ser/Thr kinases.

For use in combination with the LanthaScreen® Tb-CREB [pSer133] antibody (PV3542) to evaluate inhibitors in medium to high throughput screening applications.

ZIPtide Substrate

A peptide substrate for the Ca/Calmodulin-dependent protein kinase (CaM) family including CAMK1 and DAPK1. For use with the Adapta™ Universal Kinase Assay (PV5099).

GFP-ATF2 (19-96)

A truncated version (residues 19-96) of ATF2 has been expressed as a fusion with GFP (Green Fluorescent Protein) to act as a substrate for the JNK and p38 families of kinases.

PI Lipid Kinase Substrate

Specifically optimized for use with the Class II (PIK3C2A & PIK3C2B) PI3 kinases. These vesicles consist of 100% phosphatidylinositol (PI). For use with the Adapta™ Universal Kinase Assay (PV5099).

Fluorescein-Poly GAT

A general fluorescein peptide substrate consisting of a polymer of glutamate, alanine, and tyrosine that is used to monitor tyrosine kinase activity.

Fluorescein Diphosphate, Tetraammonium Salt (FDP) (Invitrogen™)

FDP is a colorless and nonfluorescent substrate for alkaline phosphatases. Sequential alkaline phosphatase mediated hydrolysis of its two phosphate substituents yields weakly fluorescent fluorescein monophosphate followed by strongly fluorescent fluorescein (excitation/emission ~490/514 nm).

Fluorescein-P53 (Ser15) Peptide Substrate

A fluorescein-labeled peptide substrate based upon p53 containing Ser15. p53 is phosphorylated at Ser15 by a wide range of kinases including PI3K/AKT pathway kinases such DNA-PK, ATM, and ATR.

For use in combination with the LanthaScreen® Tb-anti-phospho-p53 [pSer15] antibody (PV5130) to evaluate inhibitors in medium to high throughput screening applications.

Fluorescein-PKC Substrate

A fluorescein-labeled peptide substrate for the Protein Kinase C (PKC) family.

GFP-4E-BP1

4E-BP1 expressed as a fusion with GFP (Green Fluorescent Protein) to act as a physiological substrate for FRAP1 (mTor).

PIP2:PS Lipid Kinase Substrate

Specifically optimized for use with the Class I PI3 kinases (PIK3CA, PIK3CB, PIK3CD, and PIK3CG), this substrate consists of lipid vesicles of a 1:19 molar ratio of phosphoinositol-4,5-bisphosphate (PIP2) with phosphoserine (PS).

For use with the Adapta™ Universal Kinase Assay (PV5099).

ERM (LRRKtide) Substrate

A peptide substrate based upon ezrin/radixin/moesin (ERM) that is phosphorylated by LRRK2 and LRRK2 mutants. For use with the Adapta™ Universal Kinase Assay (PV5099).

Fluorescein-Crosstide Substrate

A general substrate for Ser/Thr kinases, the Crosstide peptide substrate is based upon GSK-3alpha/beta.

MUP (4-Methylumbelliferyl Phosphate, Free Acid) (Invitrogen™)

MUP is a fluorescent substrate for alkaline phosphatases. Alkaline phosphatase mediated hydrolysis of its phosphate substituent yields the blue-fluorescent 4-methylumbelliferyl (excitation/emission ~386/448 nm).

DDAO Phosphate (9H-(1,3-Dichloro-9,9-Dimethylacridin-2-One-7-yl) Phosphate, Diammonium Salt) (Invitrogen™)

The phosphatase substrate DDAO phosphate [9H-(1,3-dichloro-9,9-dimethylacridin-2-one-7-yl) phosphate, diammonium salt] yields a hydrolysis product that can be excited with the 633 nm laser (excitation/emission maxima ~645/660). Although the substrate itself is fluorescent (excitation/emission maxima ~460/610 nm), the difference between the substrate's excitation maximum and that of its hydrolysis product is over 200 nm, allowing the two species to be easily distinguished.

PI(5)P Lipid Kinase Substrate (Invitrogen™)

The PI(5)P Lipid Kinase Substrate has been specifically optimized for use with Type II PIP kinases (e.g., PIP4K2A). The substrate consists of solubilized phosphatidylinositol 5-phosphate (PI(5)P) in an aqueous buffer, and is used in combination with our Adapta™ Universal Kinase Assay to help identify unique kinase inhibitors.

A truncated version (residues 1-179) of c-Jun has been expressed as a fusion with GFP (Green Fluorescent Protein) to act as a substrate for the JNK family of kinases.
