CellMask™ Deep Red Plasma membrane Stain (Invitrogen™)

The CellMask™ Plasma Membrane Stains allow fast and uniform labeling of the plasma membrane without the cell-type differences exhibited by lectins. These stains may be used as a segmentation tool for HCS (high-content screening), as well as to stain cellular plasma membranes for standard fluorescence microscopy. The stain survives fixation, but not permeabilization, so it is not suitable for experiments that also involve probing internal targets via antibodies.

LysoSensor™ Yellow/Blue dextran, 10,000 MW, Anionic, Fixable (Invitrogen™)

LysoSensor Yellow/Blue dextran is a ratiometric probe that can be used to measure the pH of acidic organelles, such as lysosomes. This dextran conjugate is taken up by the cells through endocytosis. The LysoSensor dye produces blue fluorescence in neutral environments, but changes to yellow fluorescence in more acidic environments. The pKa of LysoSensor Yellow/Blue dextran is ~3.9.

MitoTracker™ Orange CM-H2TMRos - Special Packaging (Invitrogen™)

MitoTracker® Orange CM-H2TMRos is a reduced, nonfluorescent version of MitoTracker Orange (M-7510) that fluoresces upon oxidation. This dye also stains mitochondria in live cells and its accumulation is dependent upon membrane potential. The dye is well-retained after aldehyde fixation.

LysoSensor™ Green DND-189 - Special Packaging (Invitrogen™)

LysoSensor™ Green DND-189 can be used to measure the pH of acidic organelles, such as lysosomes. The LysoSensor dye becomes more fluorescent in acidic environments. The pKa of LysoSensor™ Green DND-189 is ~5.2.

ER-Tracker™ Blue-White DPX, for live-cell imaging (Invitrogen™)

ER-Tracker Blue-White DPX dye is a photostable probe that is selective for the endoplasmic reticulum (ER) in live cells. The cells can be aldehyde-fixed after staining with the dye but significant fluorescence signal will be lost. With excitation at ~374 nm, the environmentally-sensitive probe exhibits emission between 430 and 640 nm. A long-pass DAPI filter works best.

Yeast Mitochondrial Stain Sampler Kit (Invitrogen™)

The Yeast Mitochondrial Stain Sampler Kit contains sample quantities of five different cell-permeant probes that have been found to label mitochondria in live yeast. In addition to the well characterized mitochondrial stains rhodamine 123, and DiOC6(3), this kit includes rhodamine B hexyl ester, MitoTracker® Green FM and SYTO® 18 yeast mitochondrial stain, a proprietary green fluorescent nucleic acid stain. Each of these probes is sequestered in functioning mitochondria.

LysoTracker™ Deep Red (Invitrogen™)

LysoTracker® Deep Red is a deep red-fluorescent dye for labeling and tracking acidic organelles in live cells.

Features of all LysoTracker® probes include:

• Effective labeling of live cells at nanomolar concentrations
• Live-cell imaging
• With excitation/emission of 647/668 nm, the same filter set can be used as used for Cy5 or MitoTracker® Deep Red

Simple, Highly Specific, One-Step Staining and Tracking of Acidic Organelles
The LysoTracker® probes consist of a fluorophore linked to a weak base that is only partially protonated at neutral pH. This allows LysoTracker® probes to freely permeate cell membranes enabling them to label live cells. LysoTracker® probes are highly selective for acidic organelles, and unlike dinitrophenyl (DNP), LysoTracker® probes do not require use of a second antibody for detection.

LysoTracker® probes are available in a variety of fluorescent colors.

Image-iT™ TMRM Reagent (mitochondrial membrane potential indicator) (Invitrogen™)

Healthy mitochondrial membranes maintain a difference in electrical potential between the interior and exterior of the organelle, referred to as a membrane potential. Tetramethylrhodamine, methyl ester (TMRM) is a cell-permeant dye that accumulates in active mitochondria with intact membrane potentials. If the cells are healthy and have functioning mitochondria, the signal will be bright. Upon loss of the mitochondrial membrane potential, TMRM accumulation will cease and the signal will dim or disappear. TMRM signal can be detected with fluorescence microscopy, flow cytometry, cell sorting, high-throughput screening, and high content analysis.

LysoTracker™ Yellow HCK-123 - Special Packaging (Invitrogen™)

LysoTracker® Yellow HCK-123 is a yellow fluorescent dye that stains acidic compartments in live cells with excitation/emission maxima ~465/535 nm.

Yeast Vacuole Membrane Marker MDY-64 (Invitrogen™)

MDY-64 is a green-fluorescent yeast vacuole membrane stain.

MitoTracker™ Green FM - Special Packaging (Invitrogen™)

MitoTracker Green FM is green-fluorescent mitochondrial stain which appears to localize to mitochondria regardless of mitochondrial membrane potential. The dye will stain live cells but is not well-retained after aldehyde fixation.
