Human Skeletal Myoblasts (Gibco™)

Primary Normal Human Skeletal Myoblasts (HSkM) are designed to undergo highly efficient differentiation directly following plating of cryopreserved cells. Each vial contains cells that have been cryopreserved in a medium containing 10% DMSO and will differentiate within 48 hours after plating. These cells do not require expansion prior to differentiation. Cell viability is >70% post thaw with a ≥50% myogenic index after 48 hours. Intended for use by researchers investigating the molecular and biochemical bases of various normal and disease processes. They are compatible with Bacmam delivery technologies for various downstream applications. Large size contains 4.5 x 10^6 ≤ N ≤ 7.5 x 10^6 cells per vial and small size contains 1.1 x 10^6 ≤ N ≤ 1.9 x 10^6 cells per vial.

C57BL/6 Mouse Embryonic Fibroblasts, irradiated (Gibco™)

Irradiated Gibco C57BL/6 Mouse Embryonic Fibroblasts (MEFs) are feeder cells that support the culture of healthy undifferentiated embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs), especially mouse ESCs/iPSCs. Manufactured by MTI-GlobalStem and equivalent to MTI-GlobalStem Cat. Nos. Use these feeder cells to support undifferentiated ESC or iPSC culture while performing G418 selection.

Human Suspension Hepatocytes, Polymorphic Donors, (9-12 x 106 cells) (Gibco™)

Each lot of cryopreserved human hepatocytes is suitable for use in suspension metabolism applications and has been pharmacogenetically characterized as an homozygous null allele (e.g. CYP2D6*4⁄*4) via genotyping analysis (TaqMan®). The commonly tested Phase I (P450) and Phase II (UGT, SULT) drug metabolizing enzymes are analyzed for metabolic activity in each lot. Metabolite formation is measured by standard biochemical assays using LC-MS/MS or HPLC.

Kupffer Cells for Improved Physiological Modeling

Growing evidence shows that under both normal and pathological conditions, many hepatocyte functions are regulated by substances released from neighboring non-parenchymal cells (NPC). These cells, particularly Kupffer cells, play an important role in the modulation of xenobiotic metabolism in the liver. Kupffer cells secrete potent mediators of the inflammatory response that controls liver inflammation. These cytokine mediators control hepatocyte metabolic rates through direct interactions with phase I and phase II enzymes. Kupffer cell and hepatocyte co-cultures can self-assemble within 72 hours of treatment with pro-inflammatory cytokines or LPS, and can function by effectively modulating P450 expression, thus giving a more physiologically relevant result. Studies indicate that co-culture of hepatocytes with NPCs would better represent both normal liver physiology as well as disease states.
