Human Plateable Hepatocytes, 5-Donor (Gibco™)

These "5-Donor" Human Plateable Hepatocytes are a pooled population of primary hepatocytes produced by combining cells from 5 individual donors in a single vial. Pooling donors helps to minimize lot-to-lot variation and improves predictability of drug metabolism studies and other biological assays.

CellStream Isolation Technology
HEP50 Pooled Human Hepatocytes are produced using our novel proprietary CellStream Isolation Technology. This process eliminates the disadvantages of density gradient fractionation prior to cryopreservation of the pool. Instead, we gently isolate and purify the pre-screened, high-viability, and metabolically-active hepatocytes from other cells and debris. The result is a remarkably pure pooled hepatocyte preparation with high viability for use in drug metabolism studies and other biological assays.

Human Plateable Hepatocytes, Transporter Qualified (Gibco™)

Plateable cryopreserved human hepatocytes assessed for transporter function using uptake transporter assays at both 4°C and 37°C.

Cells per vial: 4-8 million

Hepatocyte cultures were incubated with radiolabeled taurocholate, digoxin, and estradiol 17β glucuronide (E2-17G), substrates of the uptake and efflux transporters NTCP and BSEP, OCT1 and MDR1, and OATPs and MRP2, respectively. Accumulation rates and biliary excretion index (BEI%) were calculated.

Human Suspension Hepatocytes, Polymorphic Donors, (9-12 x 106 cells) (Gibco™)

Each lot of cryopreserved human hepatocytes is suitable for use in suspension metabolism applications and has been pharmacogenetically characterized as an homozygous null allele (e.g. CYP2D6*4⁄*4) via genotyping analysis (TaqMan®). The commonly tested Phase I (P450) and Phase II (UGT, SULT) drug metabolizing enzymes are analyzed for metabolic activity in each lot. Metabolite formation is measured by standard biochemical assays using LC-MS/MS or HPLC.

Kupffer Cells Provide for Improved Physiological Modeling
Growing evidence shows that under both normal and pathological conditions, many hepatocyte functions are regulated by substances released from neighboring non-parenchymal cells (NPC). These cells, particularly Kupffer cells, play an important role in the modulation of xenobiotic metabolism in the liver. Kupffer cells secrete potent mediators of the inflammatory response that controls liver inflammation. These cytokine mediators control hepatocyte metabolic rates through direct interactions with phase I and phase II enzymes. Kupffer cell and hepatocyte co-cultures can self-assemble within 72 hours of treatment with pro-inflammatory cytokines or LPS, and can function by effectively modulating P450 expression, thus giving a more physiologically relevant result. Studies indicate that co-culture of hepatocytes with NPCs better represent both normal liver physiology as well as disease states.

C57BL/6 Mouse Embryonic Fibroblasts, irradiated (Gibco™)

Irradiated Gibco C57BL/6 Mouse Embryonic Fibroblasts (MEFs) are feeder cells that support the culture of healthy undifferentiated embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs), especially mouse ESCs/iPSCs. Manufactured by MTI-GlobalStem and equivalent to MTI-GlobalStem Cat. Nos. GSC-6002G and GSC-6202G, these MEF cells are isolated from inbred C57BL/6 mice, mitotically arrested by irradiation, and tested to help ensure performance and minimize the risk of contamination. Cryopreserved human hepatocytes for use in transporter-mediated uptake applications.

Prototypical hepatic uptake transporter substrates taurocholate and estradiol-17β-glucuronide have been assessed to qualify the transport function of hepatic NTCP (SLC10A1) and OATP1B14/3 (SLCO1B1/3), respectively. The transport of each substrate (radiolabeled) was monitored by scintillation analysis and active transport values (37°C - 4°C) determined by linear regression.
