In the context of immunohistochemistry (IHC), biotin is conjugated to antibodies or to the enzyme reporters used to detect target antigens.

Restore PLUS Buffer is an alternative formulation of the original Thermo Scientific Restore Western Blot Stripping Buffer. Restore PLUS Stripping Buffer was designed for use with antibodies that are difficult to remove from western blots and require longer incubation times or incubation temperatures greater than 22°C with gentler formulations. High-affinity antibodies can be quickly and effectively stripped from western blots at room temperature without removing transferred proteins, thereby allowing multiple reprobes of the target.

Protocol Summary:
• Wash blot to remove chemiluminescent substrate
• Incubate blot in Restore Western Blot Stripping Buffer for 5 to 15 minutes at room temperature
(or incubate at 37°C for high affinity antibodies)
• Remove blot and wash in Wash Buffer
• Block membrane
• Test for sufficient removal of antibodies
• Perform next immunoblot experiment

SureLock™ Tandem Midi Blot Module Invitrogen™

The Invitrogen SureLock Tandem Midi Blot Module is a wet transfer device used exclusively with the SureLock Tandem Midi Gel Tank. The tank accommodates one blot module per chamber, or two blot modules total. The SureLock Tandem Midi Blot Module is compatible with all Invitrogen midi gels (blot size: 8.5 cm x 14 cm). The blot module accommodates one midi gel per wet transfer. The unique design make the module easy to use, while the efficient size of the inner core uses less transfer buffer per transfer (only ~300 mL per transfer). Since transfer buffer contains methanol, this also results in less hazardous waste requiring special disposal. At the recommended conditions and constant voltage, proteins can be transferred to nitrocellulose or PVDF membranes in 30 minutes and at room temperature.

ProLong™ Live Antifade Reagent, for live cell imaging Invitrogen™

ProLong® Live Antifade Reagent helps prevent the loss of fluorescent signal due to photobleaching during live cell imaging. When dyes are illuminated, they can degrade (usually permanently), reducing the sample’s fluorescent intensity and leading to the creation of free radical singlet oxygen, an element that can further degrade neighboring dye molecules. ProLong® Live Antifade Reagent metabolizes this element, increasing the signal intensity and duration while keeping the background signal low. ProLong® Live reagent has little-to-no effect on cellular viability, proliferation, or other cellular functions in experiments up to 48 hours.

ProLong® Live Antifade Reagent contains enzymes from the plasma membrane of naturally occurring E. coli. These enzymes metabolize the elements that can cause photobleaching. They work in the extracellular space and are not known to enter cells, so intracellular functions are minimally affected by ProLong® Live Antifade Reagent. As shown in figure below, cells grown in the presence of ProLong® Live Antifade Reagent display minimal effects on cellular viability or proliferation. ProLong® Live Antifade Reagent provides photobleach protection for most fluorophores frequently used in live cell fluorescent imaging, such as GFP, RFP, Hoechst 33342, MitoTracker®, LysoTracker® and CellTracker® dyes. ProLong® Live Antifade Reagent can be added to any cell culture media or buffer that is suitable for fluorescent imaging.
