The Bolt® Western Pack (PVDF) includes two Mini Blot Modules and all of the buffers and pre-cut membrane/filter paper sandwiches you will need for the wet transfer of proteins from Bolt® Bis-Tris Plus gels to PVDF membranes. Each pack contains:

About the Mini Blot Module
The Mini Blot Module is a wet transfer device used exclusively with the Mini Gel Tank. The tank accommodates one blot module per chamber, or two blot modules total with the side-by-side layout. The Mini Blot Module is compatible with Bolt® Bis-Tris Plus gels and accommodates one gel per wet transfer. The universal connection and molded gasket make the blot module easy to use, while the inner core of the blot module allows for use of less methanol-based transfer buffer per wet transfer. At the recommended conditions and constant voltage, proteins can be transferred to nitrocellulose or PVDF membranes typically in 30 to 60 minutes.

Power Blotter Pre-cut Membranes and Filters, nitrocellulose, regular size Invitrogen™

These Invitrogen Power Blotter Pre-cut Membranes and Filters are designed for transfer of proteins from two mini-size or one midi-size gel using the Power Blotter System. The pre-cut membranes and filters include 20 sheets of nitrocellulose and 80 sheets of absorbent blotting paper (0.85 mm thickness). Power Blotter Pre-cut Membranes and Filters combined with Power Blotter 1-Step Transfer Buffer allow for high-efficiency semi-dry transfer of proteins in less than 10 minutes.

Features of nitrocellulose membranes:
High-quality—pure 100% nitrocellulose membranes with high surface area and excellent uniformity
High-binding affinity—provides excellent protein binding (209 µg/cm2), blocks easily, and produces very low background in chemiluminescent and fluorescent western blotting

The nitrocellulose membrane (0.2 µM pore size) is composed of 100% pure nitrocellulose to provide high-quality transfers. Nitrocellulose is a popular binding matrix for western blotting because of its high affinity for proteins and compatibility with a variety of detection methods (e.g., chemiluminescence, chromogenic, fluorescence). The pre-cut membrane/filter paper sandwiches fit two mini-size or one midi-size gel.

Biodyne B Nylon Membrane is manufactured with a matrix pore size of 0.4 micron, and is supplied in packages of 8 x 12 cm sheets. Biodyne B Membrane contains exclusively positive charges, providing the highest possible binding capacity for negatively charged molecules like DNA and RNA. The membrane is resistant to heat and solvents and will not shrink, crack or tear.

About the Mini Gel Tank
The Mini Gel Tank is compatible with a variety of Novex® gels, NuPAGE® gels, and Bolt® Bis-Tris Plus gels. Each Mini Gel Tank can accommodate up to two gels per run. The unique tank design enables convenient side-by-side gel loading and enhanced viewing during use. Optimized conditions using constant voltage allow for Bolt® Bis-Tris Plus gels to be run in approximately 35 minutes. Run times may vary depending on buffer conditions and power supplies being used.

About Bolt® Bis-Tris Plus Gels
Bolt® Bis-Tris Plus gels are precast polyacrylamide gels designed for optimal separation of your small- to medium-sized proteins under denaturing conditions. Similar to NuPAGE® Bis-Tris gels, Bolt® Bis-Tris Plus gels are designed to deliver consistent gel performance and provide a neutral pH environment that minimizes protein modifications. Bolt® gels are ideal for western blot transfer and analysis, and any other techniques where protein integrity is crucial. Also use Bolt® gels to obtain optimal results for your day-to-day protein separation needs. Bolt® Bis-Tris Plus gels come in four g types and multiple well formats. Each box contains 10 gels.

About the iBlot® 2 Gel Transfer Device
The iBlot® 2 Gel Transfer Device is our second-generation dry-transfer device, providing the same speed and convenience as the original iBlot® device, but with many new features. The iBlot® 2 Gel Transfer Device performs western blotting transfer simply, efficiently, and reliably, within seven minutes or less, and without the need for liquid buffers. Invitrogen SureLock Tandem Midi Membranes and Filters, 0.45 μm, PVDF, have excellent binding properties for western blotting, dot-blot assays, and protein sequencing. The 0.45-µm pore size makes these membranes ideal for protein transfer for proteins >10 kDa. The dimensions (8.6 x 14 cm) of these PVDF membrane and filter paper sandwiches are ideal for wet-transfer of Invitrogen midi gels using the SureLock Tandem Midi Blot Module. The 0.45-µm pore size membrane and filter paper sandwiches are conveniently supplied as pack of 20.

Each Power Blotter Select Transfer Stack contains filter paper, membrane, and the appropriate cathode and anode buffers incorporated into a solid gel matrix to allow fast, reliable transfer of proteins.

PVDF membrane
The PVDF membrane (0.2 µm pore size, low fluorescence) in the stack has higher binding capacity than nitrocellulose. It is pre-activated and ready for use without any pretreatment with alcohols. The membrane is compatible with commonly used detection methods such as staining, immunodetection, fluorescence, and radiolabeling. The proteins bind to the membrane through hydrophobic interactions. The protein-binding capacity is 240 µg/cm2.

Mini transfer stacks
The mini-size (8 cm x 8 cm) Power Blotter Select Stacks can be used with all Invitrogen Novex, Bolt, and NuPAGE mini-size gels or equivalent.
