Features of Novex Tris-Glycine gels, WedgeWell format, include:
Improved shelf life—store gels for up to 6 months at 4°C
Diversity—use for native and denaturing protein assays
Wedge-shaped wells—easily load up to two times more sample volume
Fast run conditions—quickly separate your proteins using constant voltage in less than 60 minutes

Choose the right gel format for your experiments
Novex Tris-Glycine gels, WedgeWell format, come in a variety of fixed concentrations from 6% to 18%, as well as gradients with ranges of 4–12%, 4–20%, 8–16%, and 10–20%. You can select from our many well formats, including 10-, 12-, and 15-well, and different pack sizes.

Run your proteins in native or denatured form
Novex Tris-Glycine gels, WedgeWell format, do not contain SDS and can be used to run your proteins in native or in denatured form. For denatured proteins, we recommend using Novex Tris-Glycine SDS Sample Buffer and Novex Tris-Glycine SDS Running Buffer. For native proteins, we recommend using Novex Tris-Glycine Native Sample Buffer and Novex Tris-Glycine Native Running Buffer.

Convenient to use
Novex Tris-Glycine gels, WedgeWell format, have innovative wedge-shaped wells that allow up to two times more sample load volume than other 1.0 mm-thick gels. The wedge-shaped wells also have larger openings, making sample loading easier.

For transferring your proteins to a membrane, we recommend using Novex Tris-Glycine Transfer Buffer when preforming traditional wet transfer using the Invitrogen Mini Blot Module. Transfers can also be performed with rapid semi-dry transfer using the Invitrogen Power Blotter or rapid dry transfer using the iBlot 2 Gel Transfer Device.

Pierce™ Silver Stain for Mass Spectrometry Thermo Scientific™

Pierce Silver Stain for Mass Spectrometry is a complete kit for rapid and ultra-sensitive silver staining of proteins in polyacrylamide gels and efficient destaining of excised gel pieces for mass spectrometry analysis. This kit enables both first-time and experienced users to achieve consistent and reliable staining using high-, low-, and gradient-percentage gels in 1D and 2D formats. The optimized staining method ensures extremely sensitive staining while minimizing covalent crosslinking of protein to the gel matrix, which can inhibit protein recovery. The destaining reagents facilitate complete removal of silver from stained protein bands and maximum protein recovery for subsequent mass spectrometry analysis.

Compare all silver stains ›

Features:
Sensitivity—0.25 ng per spot in 30 minutes after fixing
MS compatible—reagents and protocol are optimized to provide excellent performance in mass spectrometry following extraction of peptides from stained 1D or 2D gels (SDS-PAGE)
Flexible staining protocol—fix gel for 15 to 30 minutes or overnight without any effect on staining performance; stain for 1 to 30 minutes (typically 2 to 3 minutes)
Compatibility—effective silver stain for even difficult-to-stain basic proteins, such as lysozyme (pI 10) and chymotrypsinogen A (pI 9.2), which are detectable at 0.2 ng and 0.5 ng, respectively
Complete kit—contains all reagents necessary for staining and destaining before in-gel proteolysis and peptide recovery for analysis by mass spectrometry

Silver staining methods generally use either glutaraldehyde or formaldehyde, which cause some covalent crosslinking of protein to each other and the gel matrix. To the extent that this crosslinking occurs, extraction or elution of protein from the gel will be inhibited. Pierce Silver Stain uses formaldehyde in the stain and developer working solutions. However, the procedure accompanying the kit for mass spectrometry is optimized to maximize polypeptide recovery without greatly sacrificing sensitivity.

Choose the right NuPAGE Bis-Tris gel for your protein separation
Obtain optimal separation of your proteins by choosing the right combination of gel and running buffer. NuPAGE Bis-Tris protein gels come in four polyacrylamide concentrations: 8%, 10%, 12%, and a 4–12% gradient. Gels come in two sizes: mini (8 cm x 8 cm) or midi (8.7 cm x 13.3 cm) and either 1.0 mm (mini and midi gels) or 1.5 mm (mini gel format only) in thickness. NuPAGE Bis-Tris gels also come in multiple well formats.

NuPAGE Bis-Tris gels are formulated for denaturing gel electrophoresis applications. For optimal sample preparation, use the NuPAGE LDS Sample Buffer and NuPAGE Sample Reducing Agent. Use NuPAGE Antioxidant in the running buffer to maintain the reduced state of the proteins during the run and to allow maximum band sharpness. The gels can be run using NuPAGE MES SDS Running Buffer to better resolve small proteins or NuPAGE MOPS SDS Running Buffer to resolve medium- to large-size proteins.

We also provide NuPAGE Tris-Acetate gels for separating larger proteins. For classic Laemmli-based Tris-glycine electrophoresis, we provide Novex Tris-Glycine gels.

For transfer of proteins to a membrane, we recommend using NuPAGE Transfer Buffer. Rapid semi-dry transfer can be done using the Pierce Power Blotter or rapid dry transfer using the iBlot 2 Gel Transfer Device. Alternatively, traditional wet transfer can be performed using the XCell II Blot Module or the Mini Blot Module.
