Image-iT Plant Tissue Clearing Reagent is used to render whole-plant tissue optically clear for imaging. Image-iT Plant Tissue Hard-Set Mountant can then be used to mount and preserve the optically cleared whole-plant tissue for imaging, storage, and archiving. These reagents are compatible with both fluorescence-based and colorimetric-based applications. Image-iT Plant Tissue Clearing Reagent is an effective replacement for chloral hydrate, traditionally used to clear plant tissues in botanical microscopy.

Plant tissues often have structures with different refractive indexes, causing light scattering and spherical aberration, resulting in opaqueness. Image-iT Plant Tissue Clearing Reagent, with a high refractive index of 1.45, allows light to pass through the plant tissues with minimal light scattering, resulting in optical clarity and the ability to image deeper within the tissue.

Traditionally, researchers used chloral hydrate to clear plant specimens, but under current regulations it is a Federally Regulated Schedule IV substance (1), making it harder to acquire and use. Image-iT Plant Tissue Clearing Reagent is an effective replacement for chloral hydrate in almost all applications and tissue types where optical clearing is required.

Image-iT Plant Tissue Clearing Reagent and Image-iT Plant Tissue Hard-Set Mountant are based on Visikol™ for Plant Biology™ and Visikol™ Mount. Visikol for Plant Biology has been used in various fluorescent and colorimetric studies with plants (2,3,4) and has proved an effective substitute for chloral hydrate (1,5).

CytoVista™ 2 mm Sagittal Mouse Brain Slicer Invitrogen™

This 2-mm sagittal mouse brain slicer allows you to easily cut a mouse brain into smaller pieces for easier imaging with objectives that have a limited working distance. This slicer is 3D-printed out of resin and can be easily washed and cleaned between uses. All you need is a standard razor blade to start slicing your tissues.

• Rapid clearing of fluorescently labeled tissue for 3D imaging
• Minimal shrinkage, expansion, or other morphological changes to tissue

The CytoVista tissue and 3D cell culture clearing workflow clears tissue or 3D cultured cells with minimal changes to morphology or the sensitivity of detection with almost any fluorophore. With an easy-to-use protocol, whole mouse brain (8-mm section) can be cleared in 24 hours, or a 1-mm section in 2 hours, without the use of any special instruments or equipment.

The CytoVista clearing workflow is compatible with most fluorophores, including fluorescent proteins that are detected with common fluorescent imaging instruments, such as wide-field microscopes, high content analyzers, confocal microscopes, and lightsheet microscopes. For first time users, we recommend the CytoVista Tissue Clearing/Staining Kit (Cat No V11324) or CytoVista 3D Cell Culture Clearing/Staining Kit (Cat No. V11325). These kits contain all necessary reagents for both clearing and IHC staining.

CytoVista clearing technology sufficiently clears tissue for 3D fluorescent imaging, but unlike other clearing techniques does not significantly changes the tissue morphology. Minimal morphological changes, such as shrinkage and contraction, have been observed. For precious samples, the clearing can be reversed, and the tissue can be processed for histology studies, like H&E staining.

For tissues that are easier to clear, like brain, lung, intestine, skin, and tissues of less than 250-µm thickness, treatment with CytoVista Tissue Clearing Reagent is sufficient for 3D imaging. However, tissues of more than 250-µm thickness or that are hard to clear like kidney, liver, heart, and placenta will require an initial treatment with CytoVista Tissue Clearing Reagent followed by treatment with CytoVista Tissue Clearing Enhancer.

CytoVista 3D Cell Culture Clearing Reagent is recommended for spheroids or organoids up to 1 mm in thickness.

For hard tissues like bone, cartilage, and connective tissue, CytoVista Tissue Clearing Reagent is not recommended.

ProLong™ Gold Antifade Mountant with DAPI

ProLong Gold Antifade Mountant is a liquid mountant applied directly to fluorescently labeled cell or tissue samples on microscope slides. It is formulated without DNA stain or with the blue DNA stain DAPI or with the deep red DNA stain SYTOX Deep Red. ProLong Gold mountant contains chemical components designed to protect fluorescent dyes from fading (photobleaching) during fluorescence microscopy experiments, and delivers this protection without significantly quenching the initial fluorescence signal. It is a curing mountant that forms a near-perfect optical path (1.47 RI) and allows longer-term storage of the sample. It comes ready-to-use (room temperatures storage and no mixing required)—just apply a drop to the sample, add a coverslip, cure, and image.

ProLong Gold Antifade Mountant is not recommended for mounting samples containing fluorescent proteins, such as GFP.
