Image-iT™ FX Signal Enhancer Invitrogen™

Image-iT® FX signal enhancer is a liquid that is applied directly to slides or coverslips containing fixed and permeabilized cell or tissue samples prior to staining with fluorescent probes. When Image-iT® FX signal enhancer is used, nonspecific fluorescence (background) commonly seen with the application of fluorescent conjugates of streptavidin, goat anti-mouse, or goat anti-rabbit IgG is largely eliminated.

Image-iT Plant Tissue Hard-Set Mountant is used for imaging, storage, and archiving of plant tissues optically cleared with Image-iT Plant Tissue Clearing Reagent. ProLong™ Gold Antifade Mountant

ProLong Gold Antifade Mountant is a liquid mountant applied directly to fluorescently labeled cell or tissue samples on microscope slides. It is formulated without DNA stain or with the blue DNA stain DAPI or with the deep red DNA stain SYTOX Deep Red. ProLong Gold mountant contains chemical components designed to protect fluorescent dyes from fading (photobleaching) during fluorescence microscopy experiments, and delivers this protection without significantly quenching the initial fluorescence signal. It is a curing mountant that forms a near-perfect optical path (1.47 RI) and allows longer-term storage of the sample. It comes ready-to-use (room temperatures storage and no mixing required)—just apply a drop to the sample, add a coverslip, cure, and image.

ProLong Gold Antifade Mountant is not recommended for mounting samples containing fluorescent proteins, such as GFP. For superior antifade protection of fluorescent proteins and fluorescent dyes, ProLong Diamond Antifade Mountant is recommended. ProLong Gold Antifade Mountant comes already mixed and ready to use in a dropper bottle or 10-mL bottle with or without DNA stain. DAPI stain is excited by UV light at 360 nm when bound to DNA, with an emission maximum at 460 nm, and is detected using a DAPI traditional filter. SYTOX Deep Red stain is excited by red light at 660 nm when bound to DNA, with an emission maximum at 682 nm, and is detected using a Cy5/deep red traditional filter.

Cell imaging applications
Image-iT® FX Signal Enhancer reagent is a highly effective product for blocking background staining that results from the nonspecific interactions of a wide variety of fluorescent dyes with cell and tissue constituents. Background staining seen with fluorescent conjugates of streptavidin and antibody conjugates are largely eliminated when the Image-iT® FX Signal Enhancer reagent is applied to fixed and permeabilized cells prior to staining.

Suggestions for use
• Apply Image-iT® FX Signal Enhancer reagent directly to slides or coverslips containing cell or tissue samples prior to immune-staining.
• After the fix/perm step, simply wash and add enough drops to cover your sample, then incubate for 30 min and proceed with your normal staining protocol.
• Additional blocking steps may be performed subsequent to blocking with the Image-iT™ Signal Enhancer ReadyProbes® Reagent, as needed

Image-iT™ Fixative Solution (4% formaldehyde, methanol-free) Invitrogen™

Image-iT™ Fixative Solution is a ready-to-use neutral PBS solution containing 4% formaldehyde (w/v) that is free of methanol. Its unique formulation makes it ideal for use with fluorescent proteins, GFP, RFP, and other small dye fluorophores. eBioscience™ Intracellular Fixation & Permeabilization Buffer Set Invitrogen™

The Intracellular Fixation & Permeabilization Buffer Set is designed for use in intracellular staining and flow cytometric analysis and has been specially formulated to reduce non-specific staining of fluorochrome-labelled antibodies and increase fluorescence signal to noise ratios. In the first step live cells can be “fixed” with the Fixation Buffer, which cross- links proteins. The second step, using the Permeabilization Buffer, creates holes in the membrane thereby allowing the intracellular staining antibodies to enter the cell effectively. Subsequent washing steps, antibody additions, and incubations after cell permeabilization should be performed using the Permeabilization Buffer. The final washing step and cell resuspension prior to running the sample is done using Flow Cytometry Staining Buffer (cat. 00-4222). This kit is commonly used for staining both cytoplasmic proteins as well as proteins in the secretory pathway such as chemokines and cytokines. It is not recommended for intranuclear staining. Instead, please refer to the Foxp3 Transcription Factor Buffer Set (cat. 00-5523) for nuclear staining.

Before the fixation step, it is recommended to label the live cells using one of the Fixable Viability Dyes (eFluor™ 450, 506, 660, 780, 520, or 455UV) and then proceed with fixation.
