CTS RevitaCell Supplement (100X) is a cGMP-manufactured, animal origin-free, chemically defined supplement that is used with pluripotent stem cells (PSCs), either as a post-thaw recovery solution to improve cell viability or in combination with CTS Essential 8 Medium to enhance single cell passaging applications. Low Serum Growth Supplement Kit (LSGS Kit) Gibco™

The LSGS Kit provides, in separate vials, all the components of complete LSGS (S-003-10): fetal bovine serum, hydrocortisone, human epidermal growth factor, and basic fibroblast growth factor/heparin (stabilized with BSA). A vial of gentamycin/amphotericin solution (GA) is also included. Use of GA is optional. When a 500 ml bottle of medium is supplemented with LSGS or the components of the LSGS Kit, the final concentrations of the components in the supplemented medium are:

Geltrex™ LDEV-Free Reduced Growth Factor Basement Membrane Matrix Gibco™

Geltrex™ LDEV-Free Reduced Growth Factor Basement Membrane Matrix is a soluble form of basement membrane extracted from murine Engelbreth-Holm-Swarm (EHS) tumors and is free of viruses, including lactose dehydrogenase elevating virus (LDEV), sometimes found in this type of preparation. The major components of Geltrex™ matrix include laminin, collagen IV, entactin, and heparin sulfate proteoglycans. It is ideal for the promotion and maintenance of differentiated phenotypes in a variety of cell cultures including primary epithelial cells, endothelial cells, smooth muscle cells, and human induced pluripotent stem cells (iPSC). Geltrex™ matrix offers, on average, a 2-fold reduction in several growth factors. As a result, analysis of specific growth factor effects in applications requiring basement membrane extract can be carried out in a controlled and reproducible format.

Human Corneal Growth Supplement (HCGS) is a sterile, concentrated (100×) solution intended for use with EpiLife® medium to culture human corneal epithelial cells. HCGS is an ionically-balanced supplement containing all of the growth factors, hormones, and tissue extracts necessary for the culture of human corneal epithelial cells, and is provided in the correct amount to supplement a 500 ml bottle of medium.

This pre-diluted version of the matrix can be taken directly from the refrigerator and dispensed into cell culture plates without further handling. Geltrex hESC-Qualified Ready-To-Use basement membrane matrix is optimized for stem cell culture applications using StemPro hESC SFM. For stem cell culture using the StemFlex Medium or Essential 8 Media System, use Geltrex LDEV-Free, hESC-Qualified, Reduced Growth Factor Basement Membrane Matrix.

Microvascular Growth Supplement (MVGS) Gibco™

A sterile, concentrated (20X) solution intended for use with Medium 131 to culture human microvascular endothelial cells. Each 25 ml bottle of MVGS contains all of the growth factors, hormones, and tissue extracts necessary for the culture of human microvascular endothelial cells, and is the correct amount of supplement for a 500 ml bottle of Medium 131. When a 500 ml bottle of medium is supplemented with MVGS, the final concentrations of the components in the supplemented medium are:

• hydrocortisone (1 µg/ml)

Human Melanocyte Growth Supplement-2 (HMGS-2), PMA-free, is a sterile, concentrated (100X) solution intended for use with Medium 254 or Medium 254CF to culture human epidermal melanocytes. Each 5-mL bottle of HMGS-2 contains all of the growth factors, hormones, and tissue extracts necessary for the culture of human epidermal melanocytes and is the correct amount of supplement for a 500-mL bottle of medium.

When a 500-mL bottle of medium is supplemented with HMGS-2, the final concentrations of the components in the supplemented medium are:

• Fetal bovine serum: 0.5%
• Basic fibroblast growth factor: 3 ng/mL

Low Serum Growth Supplement (LSGS) is a sterile, concentrated (50X) solution intended for use as one component in a complete culture environment for the growth of human dermal fibroblasts and endothelial cells derived from large vessels. LSGS is intended for use with Medium 106 (fibroblasts) or Medium 200 (endothelial cells).

LSGS contains: fetal bovine serum, basic fibroblast growth factor, heparin, hydrocortisone, and epidermal growth factor.

When a 500-mL bottle of medium is supplemented with LSGS or the components of the LSGS Kit, the final concentrations of the components in the supplemented medium are:
• Fetal bovine serum: 2% v/v
• Hydrocortisone: 1 µg/mL
• Human epidermal growth factor: 10 ng/mL
• Basic fibroblast growth factor: 3 ng/mL

HuMEC Supplement Kit Gibco™

HuMEC Supplement Kit contains growth factors and other components and is designed to work with HuMEC Basal Serum-Free Medium (12753018). Together, these products are engineered specifically for the culture of human mammary epithelial cells (A10565). This translates to greater experimental validity, more reproducible results, and reduction of lot-to-lot variability and aberrations that can occur with the addition of serum. Human mammary epithelial cells cultured in HuMEC Medium have gene expression profiles consistent with normal mammary epithelial cells, as tested on a cDNA microarray.

Applications:
HuMEC Medium is optimized specifically for the growth of human mammary epithelial cells, and has been performance tested in a growth assay using normal human mammary epithelial cells. HuMEC Medium promotes superior growth of normal human mammary epithelial cells, making it ideal for researchers in breast cancer, drug discovery, and other aspects of breast biology.

Contents and Storage:
The HuMEC Supplement Kit includes 5 ml of a supplement mix containing epidermal growth factor, hydrocortisone, isoproterenol, transferrin, and insulin, and 25 mg of bovine pituitary extract. Store the supplement mix at -5°C to -20°C.

RevitaCell™ Supplement (100X) Gibco™

RevitaCell™ Supplement (100X) has been optimized for use with pluripotent stem cells (PSCs), either as a post-thaw recovery solution to improve cell viability or in combination with Essential 8™ Medium to enhance single cell passaging applications. It can also be used for recovery of primary neurons, traditionally a challenging cell to efficiently recover. The significant improvement seen in these applications is due to the presence of a more specific ROCK inhibitor coupled with molecules containing antioxidant and free radical scavenger properties that minimize the impact of stress on the cells.

• Enabling—provides maximum cell viability when performing single cell passaging
• Rapid cell recovery—high post-thaw cell viability and growth of pluripotent stem cells (passaged as clumps or single cells) and neurons
• Consistent—formulated using animal origin-free and chemically defined materials for quality and confidence

Single cell passaging
Use of the RevitaCellTM Supplement for 18-24 hours post-single cell passaging provides vast improvement in cell survival. Cells are shown to reach passaging confluency within 3-4 days post-seeding.

Higher specificity
Kinase profiling of pro-survival small molecules in RevitaCell™ Supplement shows higher specificity in comparison to traditional ROCK inhibitors (i.e., Y-27632 and Thiazovivin).

Quicker post-thaw recovery
PSCs recovered at 40,000 viable cells/cm2 using media supplemented with RevitaCell™ Supplement for the first 24 hours post-thaw recovery followed by feeding with unsupplemented Essential 8™ Medium will typically reach 80% confluency in <5 days, significantly reducing the average post-thaw recovery time of most cryopreservation solutions. Neurite outgrowth of neurons is also significantly improved using RevitaCell™ Supplement.

Mammary Epithelial Growth Supplement (MEGS) Gibco™

A sterile, concentrated (100X) solution intended for use as one component in a complete culture environment for the growth of human mammary epithelial cells. Each 5 ml bottle of MEGS contains all of the growth factors, hormones, and tissue extracts necessary for the culture of mammary epithelial cells and is the correct amount of supplement for a 500 ml bottle of Medium 171.

When a 500 ml bottle of medium is supplemented with MEGS, the final concentrations of the components in the supplemented medium are:

• Bovine pituitary extract (BPE) (0.4% v/v)
• Recombinant human insulin-like growth factor-I (0.01 µg/mL)
• Hydrocortisone (0.5 µg/ml)

Human Melanocyte Growth Supplement (HMGS) is a sterile, concentrated (100X) solution intended for use with Medium 254 or Medium 254CF to culture human epidermal melanocytes.

Each 5-mL bottle of HMGS contains all of the growth factors, hormones, and tissue extracts necessary for the culture of human epidermal melanocytes and is the correct amount of supplement for a 500-mL bottle of basal Medium 254 or Medium 254CF.

A defined, sterile, concentrated (100X) solution intended for use with EpiLife® medium to culture human epidermal keratinocytes. EDGS is not intended for use with Medium 154. EDGS contains all of the growth factors and hormones necessary for the growth of human epidermal keratinocytes. Each bottle contains the correct amount of supplement for a 500 ml bottle of EpiLife® medium. EDGS contains:
• purified bovine serum albumin
• purified bovine transferrin
• hydrocortisone
• recombinant human insulin-like growth factor type-1 (rhIGF-1)
• prostaglandin E2 (PGE2)
• recombinant human epidermal growth factor (rhEGF) The bovine products in EDGS have been isolated from animals of North American origin. The isolation process for these components includes steps that inactivate viruses. All components of EDGS are greater than 95% pure.
