CSM Media for Mav203 Yeast Cells Invitrogen™

The CSM Media for Mav203 Yeast Cells is a specifically designed media for making agar plates that enable the growth of Saccharomyces cerevisae MaV203 cells. This product comes with enough reagents to prepare 2 liters of solid media with the following components per liter:

• CSM (without tryptophan): 0.74 g
• Yeast Nitrogen Base: 6.66 g
• Ammonium Sulfate: 5 g
• Glucose: 20 g
• Agar: 20 g

Compatible with All MaV203 Applications
The CSM Media is optimized for the GeneArt® High-Order Genetic Assembly System, including the recovery for transformations of vectors once they have been adapted to yeast using the GeneArt® High-Order Vector Conversion Cassette. The media can also be used to grow MaV203 competent yeast cells, both library and sub-cloning scale (cat# 11281-011 and 11445-012).

Plate Preparation Protocol
For each liter, use one pouch of CSM -Trp Agar Yeast Media minus Glucose and 1 bottle of 20% Glucose. Dissolve the contents of the pouch in 900ml of distilled water and autoclave for 20 minutes on liquid cycle. Allow the solution to cool to 55°C and then add one bottle of 20% Glucose (100ml). Mix well and pour plates.

AmnioMAX™ C-100 Basal Medium Gibco™

Gibco® AmnioMAX™ C-100 Basal Medium is part of a kit used to prepare a fully-supplemented Gibco® AmnioMAX™ C-100 Complete Medium developed for the short term culture of human amniotic fluid cells for cytogenetic studies and in vitro diagnostic procedures.

Easy-to-use format
Gibco® AmnioMAX™ C-100 Complete Medium is a two-part kit for preparation of a complete medium that requires no additional supplementation. The complete medium can be stored at 2–8°C for up to ten days.

Quality and performance testing
Every lot of Gibco® AmnioMAX™ C-100 Complete Medium is performance tested by a certified US reference cytogenetics laboratory to ensure consistently superior performance. Pooled primary human amniotic fluid samples are cultured for six days in Gibco® AmnioMAX™ C-100 Complete Medium before measuring the total number of colonies and total number of mitotic colonies. In addition, each lot is tested for sterility, pH and osmolality.

Unique, optimized formulation
Gibco® AmnioMAX™ C-100 Complete Medium contains Fetal Bovine Serum (FBS), gentamicin, and L-glutamine to maximize cell attachment and growth. This optimized medium also has an enhanced buffering system that provides greater pH stability during culture manipulations.

ExpiFectamine™ CHO Transfection Kit Gibco™

The ExpiFectamine™ CHO Transfection Kit is a core component of the ExpiCHO™ Expression System Kit (Cat. No. A29133). It is designed for transient transfection of high-density cultures of Chinese hamster ovary (CHO) cells. The high-efficiency, cationic transfection reagent, coupled with an optimized transfection enhancer and culture feed, will power the highest possible level of protein expression from ExpiCHO-S™ Cells (Cat. No. A29127) cultured in ExpiCHO™ Expression Medium (Cat. No. A2910001).

The ExpiFectamine CHO Transfection Kit:
• Achieves greater than 90% transfection efficiency of high-density suspension CHO cell culture, with matching transfection enhancer and culture feed to boost transfection performance and protein expression
• Requires one-third to one-half less plasmid DNA than other transfection reagents while achieving superior expression performance

The ExpiFectamine CHO Transfection Kit is specifically designed to work with ExpiCHO-S Cells in the ExpiCHO™ Expression System Kit. The transfection kit supplies the cationic ExpiFectamine™ CHO Transfection Reagent, ExpiFectamine™ CHO Transfection Enhancer, and ExpiCHO Feed.

Collagenase, Type I, powder Gibco™

Collagenase is a protease that cleaves the bond between a neutral amino acid (X) and glycine in the sequence Pro-X-Gly-Pro, which is found with high frequency in collagen. Collagenase is unique among proteases in its ability to degrade the triple-helical native collagen fibrils commonly found in connective tissues such as skin, tendon, blood vessels, and bone. Collagenase disaggregation is suitable for the culture of human tumors, mouse kidney, human adult and fetal brain, and many other tissues including epithelium. Collagenase is relatively gentle, dissociates well at physiological temperature and pH, and requires no mechanical agitation or special equipment.

Gibco® Collagenase Type I is isolated from Clostridium histolyticum and packaged as a lyophilized, non-sterile powder for research use in cell or tissue dissociation, and organ perfusions. Gibco® Collagenase Type I activity is guaranteed to be greater than 125 units/mg. Compared to other collagenase preparations, Gibco® Collagenase Type I has average levels of collagenase, caseinase, clostripain, and tryptic activities, and is well-suited for the digestion of fat, adrenal, and liver cells or tissues.

ExpiCHO-S™ Cells, 6 Vial Cell Bank Pack Gibco™

ExpiCHO-S™ Cells are derived from a non-engineered subclone that has been screened and isolated from CHO-S Chinese hamster ovary (CHO) cells (1). They are a core component of the ExpiCHO™ Expression System Kit (Cat. No. A29133). ExpiCHO-S Cells are maintained in suspension culture, have minimal tendency to clump, and grow to high density in ExpiCHO™ Expression Medium (Cat. No. A2910001). As part of the ExpiCHO Expression System Kit, the cells generate superior protein yields by transient transfection compared to standard CHO, 293, and high-density Expi293F™ cells.

ExpiCHO-S™ Cells:
• Allow you to attain yields of up to 3 g of protein per L of transfected culture as part of the ExpiCHO expression system
• Enable rapid, high-yield transient protein production, allowing labs developing protein biotherapeutics to work with CHO-expressed proteins from start to finish in the drug development process
• Are adapted to high-density suspension growth in ExpiCHO Expression Medium
• Exhibit rapid doubling times of approximately 17 hours
• Enable consistent transient expression performance for over 30 passages after thaw
• Maintain high cell viability and productivity for greater than 10 days post-transfection as part of the ExpiCHO expression system

Frozen cells are supplied in a vial containing 1 mL of cells at 1 x 107 viable cells per mL in ExpiCHO Expression Medium and 10% DMSO. The cells should be thawed directly into ExpiCHO Expression Medium. The 6-vial multipack eliminates the need to expand and bank the cells in the lab.

Cells are typically grown in flasks on a shaker platform in a humidified, CO2 cell culture incubator. However, cultures are scalable from volumes of <1 mL in multi-well plates to >10 L in bioreactors. Protein harvest can be performed at 5 to 14 days post-transfection, depending on the protein expressed.

RPMI 1640 Medium (Dutch modification) Gibco™

Roswell Park Memorial Institute (RPMI) 1640 medium was originally developed to culture human leukemic cells in suspension and as a monolayer. RPMI 1640 has since been found suitable for a variety of mammalian cells including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. We offer a variety of Gibco® RPMI 1640 modifications for a range of cell culture applications. Find the right formulation using the media selector tool.

Gibco® RPMI 1640 is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 contains biotin, vitamin B12 and PABA which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations. This RPMI formulation has the Dutch modification (addition of HEPES, with sodium bicarbonate lowered to 1 g/L).

RPMI 1640 contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 uses a sodium bicarbonate buffer system (1.0 g/L) and therefore requires a 5-7% CO2 environment to maintain physiological pH.

CTS™ OpTmizer™ T-Cell Expansion SFM, no phenol red, bottle format Gibco™

Gibco CTS OpTmizer T-Cell Expansion SFM supports the growth and expansion of human T lymphocytes. It is a complete serum-free, xeno-free medium consisting of CTS OpTmizer T-Cell Expansion Basal Medium and CTS OpTmizer T-Cell Expansion Supplement, which are mixed together prior to use.

CTS OpTmizer T-Cell Expansion SFM is available with and without phenol red. This version of CTS OpTmizer SFM lacks phenol red, and is available in both bottle and bag format.

Features of CTS OpTmizer T-Cell Expansion SFM include:
• Supports high-density T cell culture (>4 x 106 CD3+ T-cells/mL)
• Supports T-cell activation using Dynabeads magnetic beads, soluble antibodies, and stimulatory antibody-presenting cell protocols

Demonstrated functional performance
Cells grown in CTS OpTmizer T-Cell Expansion SFM with or without human serum demonstrate good cytokine production profiles in polyclonally activated T cells. CTS OpTmizer T-Cell Expansion SFM maintains the desired the CD62L+ central memory T-cell phenotype.

GMP manufactured
The primary manufacturing site for CTS OpTmizer T-Cell Expansion SFM is FDA-registered and has an ISO13485-certified quality management system. The methods and controls used for the manufacturing are in conformity with current Good Manufacturing Practices for medical devices, 21 CFR Part 820, of the regulation. Alternate manufacturing sites are available, providing risk mitigation by helping to ensure security of supply.

Versatility
CTS OpTmizer T-Cell Expansion SFM has been widely adopted in clinical T-cell research applications including the culture of CAR T-cells. It works with multiple culture formats, including culture flasks, culture bags, and rocking bioreactor systems. CTS OpTmizer T-Cell Expansion SFM was designed to support expansion of cultured human T-cells without additional serum supplementation. Increased performance is generally observed when supplemented with CTS Immune Cell Serum Replacement.

B-27 Supplement XenoFree Minus Insulin is based on the classic B-27 Supplement formulation and contains no animal-derived components at the primary component level. The absence of insulin makes this formulation ideal for the growth of hippocampal and other CNS neurons in applications where the presence of insulin causes interference or in the derivation of cardiomyocytes and mesodermal cells from stem cells.

Gibco® CD BHK-21 Production Medium is the first cell culture medium supporting complete serum-free and protein-free processes for BHK-21 cells. BHK-21 cells can achieve high cell densities and maintain high cell viability in this medium.

Gibco® CD BHK-21 Production Medium is suitable for large-scale suspension culture. DMEM, high glucose, GlutaMAX™ Supplement Gibco™

DMEM (Dulbecco's Modified Eagle Medium) is a widely used basal medium for supporting the growth of many different mammalian cells. Cells successfully cultured in DMEM include primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells, as well as cell lines such as HeLa, 293, Cos-7, and PC-12. Using DMEM
DMEM is unique from other media as it contains 4 times the concentration of amino acids and vitamins than the original Eagle's Minimal Essential Medium. DMEM was originally formulated with low glucose (1 g/L) and sodium pyruvate, but is often used with higher glucose levels, with or without sodium pyruvate. DMEM with GlutaMAX™ supplement minimizes toxic ammonia build-up and improves cell viability and growth in an easy-to-use format. DMEM contains no proteins, lipids, or growth factors. Therefore, DMEM requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). DMEM uses a sodium bicarbonate buffer system (3.7 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.
