RPMI 1640 Medium, no glutamine Gibco™

RPMI 1640 Medium was originally developed to culture human leukemic cells in suspension and as a monolayer. Roswell Park Memorial Institute (RPMI) 1640 Medium has since been found suitable for a variety of mammalian cells, including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. Using RPMI
RPMI 1640 Medium is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 Medium contains biotin, vitamin B12, and PABA, which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations. RPMI 1640 Medium contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 Medium requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 Medium uses a sodium bicarbonate buffer system (2.0 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

DMEM, high glucose, no glutamine, no calcium Gibco™

Dulbecco's Modified Eagle Medium (DMEM) is a widely used basal medium for supporting the growth of many different mammalian cells. Cells successfully cultured in DMEM include primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells, as well as cell lines such as HeLa, 293, Cos-7, and PC-12. DMEM is unique from other media as it contains 4 times the concentration of amino acids and vitamins than the original Eagle's Minimal Essential Medium. DMEM was originally formulated with low glucose (1 g/L) and sodium pyruvate, but is often used with higher glucose levels, with or without sodium pyruvate.

Using RPMI 1640 Medium
RPMI 1640 medium is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 medium with GlutaMAX™ supplement minimizes toxic ammonia build-up and improves cell viability and growth in an easy-to-use format. RPMI 1640 medium contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 medium requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 medium uses a sodium bicarbonate buffer system (2.0 g/L) and therefore requires a 5–10% CO2 environment to maintain physiological pH.

LV-MAX™ Production Medium Gibco™

Gibco LV-MAX Production Medium is a chemically defined, serum-free, protein-free medium, specially developed for high-density growth and transfection of Gibco Viral Production Cells (suspension-adapted HEK 293-derived cells) promoting high-titer lentiviral production.

• Supports growth of suspension Viral Production Cells to densities of over 1.0 x 107 cells/mL

CD FortiCHO™ Medium Gibco™

CD FortiCHO™ medium is specifically designed to offer the highest batch and fed-batch culture performance and yield with recombinant CHO cells in a chemically defined environment. The protein-free, animal origin component-free formulation provides consistency and helps reduce the need to screen for adventitious agents. With CD FortiCHO™ medium cell cultures last 2+ days longer than competitive alternatives, which means decreased HCP (Host Cell Protein) concentration for removal in downstream purification.

CD FortiCHO™ medium is fortified to support maximal recombinant CHO cell growth and protein expression in suspension batch culture, offering 120% greater growth and production than select competitive media with simple glucose supplementation. CD FortiCHO™ medium can provide up to 75% greater fed-batch growth and production over glucose-only fed-batch culture for selected cell lines and applications when used with CD EfficientFeed™ C nutrient supplement.

When to Use CD FortiCHO™ Medium
• The cell line you are using is a transfected CHO K1, GS CHO or CHO-S™ cell line
• Maximum batch culture cell densities and protein titers are needed
• You have time to adapt cells into a new medium in an effort to maximize titers
• Cell health at harvest is a priority for downstream processing

When to Consider Another Gibco® Medium

Consider CD OptiCHO™ medium when:
• You are using a transfected CHO cell line other than CHO K1, GS CHO or CHO-S™ cell lines
• Optimization of feeding strategies will be part of your base medium selection testing in an effort to maximize productivity
• Your CHO cell line is “finicky," i.e. hard to grow or adapt

Consider CD CHO medium when:
• Your GS CHO cells are recommended to be grown in CD CHO medium
• Your cells have been successfully cultured in CD CHO medium in the past and your process is nearing a regulatory filing (DMF available)

Consider CD DG44 when:
• You use parental DG44 or DXB11 CHO cells (dhfr-)

Leibovitz's L-15 Medium supports the growth of HEP-2 monkey kidney cells and primary explants of embryonic and adult human tissue. L-15 is buffered by phosphates and free base amino acids instead of sodium bicarbonate. This medium is designed for supporting cell growth in environments without CO2 equilibration.

Minimum Essential Medium (MEM) α is widely used for mammalian cell culture as well as selection for transfected DHFR negative cells. MEM α can be used with a variety of suspension and adherent mammalian cells, including keratinocytes, primary rat astrocytes, and human melanoma cells. Gibco® MEM α is a modification of Minimum Essential Medium (MEM) that contains non-essential amino acids, sodium pyruvate, lipoic acid, vitamin B12, biotin, and ascorbic acid. MEM α is available without nucleosides for use as a selection medium for DG44 and other DHFR negative cells. This product is made with Earle’s salts.

For supply chain continuity, we manufacture Gibco® MEM α at two separate facilities located in Grand Island, NY and Scotland, UK. Both sites are compliant with cGMP manufacturing requirements, are certified to ISO 13485, and are registered with the FDA as medical device manufacturers.

MEM α contains no proteins, lipids, or growth factors. Therefore, MEM α requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). MEM α uses a sodium bicarbonate buffer system (2.2 g/L) and therefore requires a 5-10% CO2 environment to maintain physiological pH. Powder forms of Gibco® cell culture medium require sodium bicarbonate supplementation, pH adjustment, and filtration at the time of preparation (see protocol for details).

PBS Tablets Gibco™

Phosphate-buffered saline (PBS) is a balanced salt formulation used for a variety of cell culture applications, such as washing cells before dissociation, transporting cells or tissue, diluting cells for counting, and preparing reagents. PBS is formulated without calcium and magnesium for rinsing chelators from the culture before cell dissociation. Each 5 g Gibco® PBS Tablet should be dissolved in 500 mL of distilled water. The pH will be 7.45 and requires no adjustment.

Collagenase, Type I, powder Gibco™

Collagenase is a protease that cleaves the bond between a neutral amino acid (X) and glycine in the sequence Pro-X-Gly-Pro, which is found with high frequency in collagen. Collagenase is unique among proteases in its ability to degrade the triple-helical native collagen fibrils commonly found in connective tissues such as skin, tendon, blood vessels, and bone. Collagenase disaggregation is suitable for the culture of human tumors, mouse kidney, human adult and fetal brain, and many other tissues including epithelium. Collagenase is relatively gentle, dissociates well at physiological temperature and pH, and requires no mechanical agitation or special equipment.

Gibco® Collagenase Type I is isolated from Clostridium histolyticum and packaged as a lyophilized, non-sterile powder for research use in cell or tissue dissociation, and organ perfusions. Gibco® Collagenase Type I activity is guaranteed to be greater than 125 units/mg. Compared to other collagenase preparations, Gibco® Collagenase Type I has average levels of collagenase, caseinase, clostripain, and tryptic activities, and is well-suited for the digestion of fat, adrenal, and liver cells or tissues.

Advanced RPMI 1640 Medium Gibco™

Advanced RPMI (Roswell Park Memorial Institute) 1640 is a widely used basal medium that allows the culture of mammalian cells with reduced Fetal Bovine Serum (FBS) supplementation. Compared to classic RPMI 1640, serum supplementation can be reduced by 50-90% with no change in growth rate or morphology.i Cells successfully cultured in Advanced RPMI 1640, with no adaptation, include Sp2, Vero, Raji, Daudi, and Jurkat.

Gibco® Advanced RPMI 1640 is unique from other media due to addition of the following ingredients to allow for serum reduction: ethanolamine, glutathione, ascorbic acid, insulin, transferrin, AlbuMAX® I lipid-rich bovine serum albumin for cell culture, and the trace elements sodium selenite, ammonium metavanadate, cupric sulfate, and manganous chloride.

Gibco® Advanced RPMI 1640 is manufactured at a cGMP compliant facility located in Grand Island, New York. The facility is registered with the FDA as a medical device manufacturer and is certified to the ISO 13485 standards.

Advanced RPMI 1640 requires supplementation with 1-5% Fetal Bovine Serum and 4mM L-glutamine or GlutaMAX™ supplement. Many cell lines do not require adaptation to this media. The FBS concentration must be optimized for each cell line to obtain maximum serum reduction. Advanced RPMI 1640 uses a sodium bicarbonate buffer system (3.7 g/L) and therefore requires a 5-10% CO2 environment to maintain physiological pH.

CD BHK-21 Production Medium Gibco™

Gibco® CD BHK-21 Production Medium is a chemically defined, protein-free cell culture medium. It is designed to support the growth of BHK-21 cells in serum-free suspension culture. This medium is suitable for large-scale manufacturing applications.

Gibco® CD BHK-21 Production Medium is the first cell culture medium supporting complete serum-free and protein-free processes for BHK-21 cells. The dry powder format is easy to reconstitute, since it has no hydrolysates in the formulation. Adapting cells from serum-containing media is a rapid and simple procedure. BHK-21 cells can achieve high cell densities and maintain high cell viability in this medium.

Gibco® CD BHK-21 Production Medium is suitable for large-scale suspension culture. Because it does not require supplementation with serum or other undefined components, Gibco® CD BHK-21 Production Medium eliminates the risks associated with serum, while allowing you to achieve consistent virus titers and simplifying downstream purification.
