RPMI 1640 Medium, powder Gibco™

RPMI 1640 Medium was originally developed to culture human leukemic cells in suspension and as a monolayer. Roswell Park Memorial Institute (RPMI) 1640 Medium has since been found suitable for a variety of mammalian cells, including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. We offer a variety of RPMI 1640 Medium modifications for a range of cell culture applications. Find the right formulation using the media selector tool.

This RPMI is modified as follows:
WithWithout
• L-glutamine• HEPES
• Phenol Red• Sodium Bicarbonate

The complete formulation is available.

Using RPMI 1640
RPMI 1640 Medium is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 Medium contains biotin, vitamin B12, and PABA, which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations. RPMI 1640 Medium contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 Medium requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 Medium uses a sodium bicarbonate buffer system (2.0 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH. Powder forms of Gibco® cell culture media require sodium bicarbonate supplementation, pH adjustment, and filtration at the time of preparation (see protocol for details).

Product use
For in vitro diagnostic use. CAUTION: Not for human or animal therapeutic use. Uses other than the intended use may be a violation of local law. Customers using Gibco® RPMI 1640 in a manufacturing process, who have a submission with the FDA, may request a letter of authorization from us to reference our Type II Drug Master File (DMF).

cGMP manufacturing and quality system
RPMI 1640 Medium is manufactured at a cGMP-compliant facility located in Grand Island, New York. The facility is registered with the FDA as a medical device manufacturer and is certified to ISO 13485 standards. For supply chain continuity, we offer an identical RPMI 1640 product made in our Scotland facility (51800-043). This facility is registered with the FDA as a medical device manufacturer and is certified to the ISO 13485 standard.

Gibco® FreeStyle™ F17 Medium provides equal or better performance compared to Gibco® FreeStyle™ 293 Expression Medium and Gibco® FreeStyle™ CHO Medium. Gibco® FreeStyle™ F17 Medium supports 293-F cells at a peak density of 6–7 × 106 viable cells per mL, and supports CHO-S cells at a peak density of 8–9 × 106 viable cells per mL. Cells cultured in Gibco® FreeStyle™ F17 can be transfected using a wide variety of transfection reagents, including lipid-based transfection reagents, such as 293fectin™ and Freestyle™ MAX Reagent. Protein expression typically exceeds 75–150 µg/mL IgG in 293-F cells.

Animal origin–free, protein-free, and chemically defined formulation
Gibco® FreeStyle™ F17 Medium is animal origin–free, protein-free, and chemically defined, allowing for easier purification of your protein of interest. Gibco® chemically defined media contain no proteins, hydrolysates, or components of unknown composition.

Scalability in spinner flasks and bioreactors
Protein production using Gibco® FreeStyle™ F17 Medium can be scaled up in spinner flasks or bioreactors. The appropriate spinner or impeller speed and seeding density should be optimized for each system. Depending on the impeller design and speed, it may be necessary to supplement Gibco® FreeStyle™ F17 Medium with additional Pluronic® F-68 to avoid sheer stress in the culture.

cGMP manufacturing and quality system
Gibco® FreeStyle™ F17 Medium is manufactured at a cGMP-compliant facility located in Grand Island, New York. The facility is registered with the FDA as a medical device manufacturer and is certified to ISO 13485 standards.

Human Endothelial SFM Gibco™

Human Endothelial Serum Free Medium (SFM) has been developed to support the long-term propagation of human umbilical vein endothelial cells (HUVEC). Complete Human Endothelial SFM has been demonstrated to successfully support primary isolation and subsequent secondary growth of HUVEC for ≤15 passages. HUVEC cultured in complete Human Endothelial SFM exhibit the histotypic 'cobblestone' morphology and retain endothelial specific markers including: expression of Factor VIII-related antigen, UEA-1 lectin binding, uptake of DiI-acetylated LDL, vimentin, and IL-1a-induced ICAM-1 expression. Additionally, HUVEC cultured in Human Endothelial SFM have been shown to maintain cAMP and prostacyclin signal transduction systems. Human Endothelial SFM also supports the growth and retention of physiological markers for human umbilical arterial and dermal microvascular endothelial cells.

Human Endothelial SFM is a basal serum-free growth medium and requires supplementation with bFGF (20 ng/mL), EGF (10 ng/mL), and human plasma fibronectin (10 µg/mL).
