Collagenase, Type I, powder Gibco™

Collagenase is a protease that cleaves the bond between a neutral amino acid (X) and glycine in the sequence Pro-X-Gly-Pro, which is found with high frequency in collagen. Collagenase is unique among proteases in its ability to degrade the triple-helical native collagen fibrils commonly found in connective tissues such as skin, tendon, blood vessels, and bone. Collagenase disaggregation is suitable for the culture of human tumors, mouse kidney, human adult and fetal brain, and many other tissues including epithelium. Collagenase is relatively gentle, dissociates well at physiological temperature and pH, and requires no mechanical agitation or special equipment.

Gibco® Collagenase Type I is isolated from Clostridium histolyticum and packaged as a lyophilized, non-sterile powder for research use in cell or tissue dissociation, and organ perfusions. Gibco® Collagenase Type I activity is guaranteed to be greater than 125 units/mg. Compared to other collagenase preparations, Gibco® Collagenase Type I has average levels of collagenase, caseinase, clostripain, and tryptic activities, and is well-suited for the digestion of fat, adrenal, and liver cells or tissues.

ExpiCHO™ Stable Production Medium Gibco™

Gibco ExpiCHO Stable Production Medium (SPM) is specifically designed to simplify your cell line development process into production with minimal effort and a high degree of confidence. This chemically defined, protein-free, animal origin component-free formulation is fully optimized for large-scale fed-batch culture with ExpiCHO-S cells. ExpiCHO SPM supports an easy transition from transient to stable production, provides high titers without the need for additional medium optimization (see figures below), and works well with existing EfficientFeed C+ supplement.

ExpiCHO SPM is part of the ExpiCHO family, an end-to-end solution that empowers you to carry out your entire workflow from transient to stable expression without having to screen or optimize media and feeds.

The power to streamline transfer to manufacturing
ExpiCHO SPM liquid format and AGT dry format demonstrate comparable cell growth and titer performance (see figure below). Therefore, scale-up and manufacturing using AGT dry format is simplified by knowing that the reconstituted AGT dry format will behave similarly to the liquid format used prior to scale-up. The AGT dry format is easy to reconstitute; just add water and it will automatically adjust pH and osmolality. This can help provide efficiencies in supply chain complexity, reconstitution complexity, and, therefore, overall expense.

When to use ExpiCHO Stable Production Medium:
• When you are using the ExpiCHO-S cell line for transient expression and want to scale up
• After you have completed the stable transfection of ExpiCHO-S cells in ExpiCHO Expression Medium. (Follow our recommended protocol.)

MarrowMAX™ Bone Marrow Medium Gibco™

Gibco® MarrowMAX™ Bone Marrow Medium is a fully-supplemented medium developed for the short term culture of bone marrow and other hematopoietic cells for cytogenetic studies and in vitro diagnostic procedures.

Superior performance
Gibco® MarrowMAX™ Bone Marrow Medium outperforms other commercially available media containing Giant Cell Tumor (GCT) conditioned media, providing consistent performance with a higher mitotic index and superior chromosomal morphology (learn more about the superior performance of MarrowMAX™ medium here).

Ready-to-use format
Gibco® MarrowMAX™ Bone Marrow Medium is a frozen, 1X medium, ready to use upon thawing. No supplementation is required. Thawed medium can be stored at 2–8°C for up to 60 days.

Quality and performance testing
Every lot of Gibco® MarrowMAX™ Bone Marrow Medium is performance tested by a certified US reference cytogenetics laboratory to ensure consistently superior performance. Bone marrow cells from the aspirate of a normal adult donor are cultured for 24 hours in Gibco® MarrowMAX™ Bone Marrow Medium before measuring the mitotic index and chromosome banding resolution. In addition, each lot is tested for sterility, pH, osmolality, mycoplasma and endotoxin.

Unique, optimized formulation
Gibco® MarrowMAX™ Bone Marrow Medium is an optimized RPMI 1640 medium, supplemented with Fetal Bovine Serum (FBS), gentamicin, and L-glutamine. It also contains a novel human stromal cell conditioned medium that consists of a unique blend of hematopoietic growth factors. Gibco® MarrowMAX™ Bone Marrow Medium does not contain GCT conditioned medium.

Versene Solution Gibco™

Versene is an EDTA solution for use as a gentle non-enzymatic cell dissociation reagent. Gibco® Versene Solution (0.48 mM) is formulated as 0.2 g EDTA(Na4) per liter of Phosphate Buffered Saline (PBS).

Expi293F™ Inducible GnTI- Cells Gibco™

The Expi293F Inducible GnTI- cell line is derived from engineered Expi293F cells that do not have N-acetylglucosaminyltransferase I (GnTI) activity, therefore lacking complex N-glycans, and stably express the tetracycline repressor (TetR) protein, thus enabling tetracycline-regulated protein expression. The cell line was established by the selective knockout of the GnTI gene and stable integration of the regulatory plasmid pcDNA6/TR, which encodes the TetR gene under the control of the human CMV promoter. It is a powerful tool for tetracycline-inducible high-yield expression of homogeneously glycosylated recombinant proteins.

Features of Expi293F Inducible GnTI- cells include:
• Suspension, high-density culture in Expi293 Expression Medium
• Homogeneous N-glycosylation of expressed proteins
• Tetracycline-inducible protein expression with extremely low basal expression levels
• Equivalent protein yields to parental Expi293F cells—up to 1 g/L

For inducible expression of your protein of interest, pcDNA5/TO mammalian expression vector or an equivalent inducible vector is required for tetracycline-regulated gene expression.

Frozen cells are supplied in a vial containing 1 mL of cells at 1 x 107 viable cells/mL in Expi293 Expression Medium and 10% DMSO. The cells should be thawed directly into Expi293 Expression Medium.

Hibernate™-E Medium Gibco™

Hibernate Medium is a CO2-independent nutrient medium for the maintenance of neural cells, tissue, and tissue slices. Hibernate-E Medium is optimized for embryonic cells or tissue in ambient carbon dioxide levels.

Hibernate Medium enables neurons to be maintained in ambient CO2 levels for up to 48 hours. This provides scientists with greater convenience and control while running bench-top live cell experiments, such as microscopy, flow cytometry, and other physiology studies. Hibernate Medium also allows for the preservation of viable brain tissue before processing for up to a month with refrigeration and maintenance of tissue slices.

Hibernate Medium may be combined with B-27 Supplements to yield a complete medium.

Synth-a-Freeze™ Cryopreservation Medium Gibco™

Synth-a-Freeze Cryopreservation Medium is a defined, ready-to-use, liquid cryopreservation medium that can be used with a broad pectrum of primary cells, as well as cell lines, and demonstrates improved cell viability and recovery after thawing. Synth-a-Freeze medium contains 10% dimethylsulfoxide (DMSO) without antibiotics, antimycotics, hormones, growth factors, serum, or protein and is buffered with HEPES and sodium bicarbonate.

Synth-a-Freeze medium is intended for freezing and storing a variety of mammalian cell types including human keratinocytes, embryonic stem cells, neural stem cells, and mesenchymal stem cells. It should not be used for melanocytes.

DMEM, high glucose, GlutaMAX™ Supplement, HEPES Gibco™

DMEM (Dulbecco's Modified Eagle Medium) is a widely used basal medium for supporting the growth of many different mammalian cells. Cells successfully cultured in DMEM include primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells, as well as cell lines such as HeLa, 293, Cos-7, and PC-12. Using DMEM
DMEM is unique from other media as it contains 4 times the concentration of amino acids and vitamins than the original Eagle's Minimal Essential Medium. DMEM was originally formulated with low glucose (1 g/L) and sodium pyruvate, but is often used with higher glucose levels, with or without sodium pyruvate. DMEM with GlutaMAX™ supplement minimizes toxic ammonia build-up and improves cell viability and growth in an easy-to-use format. DMEM contains no proteins, lipids, or growth factors. Therefore, DMEM requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). DMEM uses a sodium bicarbonate buffer system (3.7 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

MEM Gibco™

MEM (Minimum Essential Medium) is one of the most commonly used of all cell culture media. MEM can be used with a variety of suspension and adherent mammalian cells, including HeLa, BHK-21, 293, HEP-2, HT-1080, MCF-7, fibroblasts, and primary rat astrocytes. Using MEM
MEM was developed by Harry Eagle, based on his earlier formulation of Basal Medium Eagle (BME). Many other modifications of MEM followed, including Glasgow’s MEM, MEM α, DMEM, and Temin’s Modification. MEM is available with Earle’s salts for use in a CO2 incubator, or with Hanks' salts for use without CO2. This product is made with Earle’s salts. MEM contains no proteins, lipids, or growth factors. Therefore, MEM requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). MEM uses a sodium bicarbonate buffer system (2.2 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

CD OptiCHO™ Medium Gibco™

CD OptiCHO™ medium is specifically designed to offer high performance and yield with recombinant CHO cells in a chemically defined fed-batch environment. The protein-free, animal origin free formulation is designed to provide consistency and reduces the need to screen for adventitious agents. CD OptiCHO™ medium builds on Gibco® CD CHO’s legacy of consistent cell growth and titers, while offering up to 40% better titer and cell growth over select competitors. Thanks to its low osmolality and advanced CHO CD EfficientFeed™ Liquid Nutrient Supplement Kit feeding options, CD OptiCHO™ medium is capable of gaining 2-5 fold increases in fed-batch productivity over batch culture.

When to Use CD OptiCHO™ Medium

• You are using a transfected CHO cell line other than CHO K1, GS CHO or CHO-S™ cell lines
• Optimization of feeding strategies will be part of your base medium selection testing in an effort to maximize productivity
• Your CHO cell line is “finicky," i.e. hard to grow or adapt

When to Consider Another Gibco® Medium

Consider CD FortiCHO™ medium when:
• The cell line you are using is a transfected CHO K1, GS CHO or CHO-S™ cell line
• Maximum batch culture cell densities and protein titers are needed
• You have time to adapt cells into a new medium in an effort to maximize titers
• Cell health at harvest is a priority for downstream processing

Consider CD CHO medium when:
• Your GS CHO cells are recommended to be grown in CD CHO medium
• Your cells have been successfully cultured in CD CHO medium in the past and your process is nearing a regulatory filing (DMF available)

Consider CD DG44 medium when:
• You use parental DG44 or DXB11 CHO cells (dhfr-)

Dry Media Formulations
Gibco® liquid media are available in an AGT™ (Advanced Granulation Technology) dry media format that is designed to provide increased consistency and productivity across all stages of production from development to commercial manufacturing. AGT™ media are manufactured animal origin-free with the same complete, serum-free, protein-free, chemically-defined formulation and performance as their liquid media counterparts.

293 SFM II Gibco™

293 SFM II is a serum-free, low-protein (<10 µg/ml) formulation optimized for high-density suspension culture of 293 cells (1). (It is not suitable for adherent 293 cell cultures.) This product replaces 293 SFM (Cat. No. 10970-010). 293 SFM II is animal-origin free. Formulated without L-glutamine for increased stability; add 20 ml of 200 mM L-glutamine or 20 ml of GlutaMAX™-I Supplement per 1,000 ml of medium prior to use.

RPMI 1640 Medium (Dutch modification) Gibco™

Roswell Park Memorial Institute (RPMI) 1640 medium was originally developed to culture human leukemic cells in suspension and as a monolayer. RPMI 1640 has since been found suitable for a variety of mammalian cells including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. We offer a variety of Gibco® RPMI 1640 modifications for a range of cell culture applications. Find the right formulation using the media selector tool.

Gibco® RPMI 1640 is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 contains biotin, vitamin B12 and PABA which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations. This RPMI formulation has the Dutch modification (addition of HEPES, with sodium bicarbonate lowered to 1 g/L).

PSC Cryopreservation Kit Gibco™

The PSC Cryopreservation Kit contains xeno-free PSC Cryopreservation Medium, which is a ready-to-use solution for the cryopreservation of early passage pluripotent stem cells (PSCs), and RevitaCell™ Supplement (100X), a chemically defined recovery supplement for use in the post-thaw culture media. When used in combination, these reagents help minimize loss of cell viability, maximize post-thaw recovery, and minimize unwanted differentiation of pluripotent stem cells. This kit can also be used to cryopreserve and recover peripheral blood mononuclear cells (PBMCs) to improve post-thaw cell viability and recovery.

• Efficient recovery—routinely achieves 50% more viable cells than traditional methods of PSC cryopreservation
• Consistent—chemically defined media maintains normal morphology, pluripotency, and karyotype of pluripotent stem cells over 10 passages
