McCoy's 5A (Modified) Medium Gibco™

McCoy's 5A (modified) Medium is a general purpose medium that supports the propagation of many types of primary cells, established cell lines, and explants from biopsy tissues. This medium will support the growth of primary mammalian cells derived from normal bone marrow, skin, spleen, kidney, lung, rat embryos, and other tissues.

Using McCoy's 5A (modified) Medium
Dr. Thomas McCoy originally formulated McCoy's 5A medium as a modification of Basal Medium 5A. Unlike other media, McCoy's 5A contains the reducing agent glutathione, bacto-peptone, and a high level of glucose. This product also includes Dr. Hsu's addition of Hanks' salts to enable use outside a CO2 incubator. McCoy's 5A (modified) Medium requires serum supplementation, commonly with 10% Fetal Bovine Serum (FBS). McCoy's 5A (modified) Medium uses a sodium bicarbonate buffer system (2.2 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

Advanced DMEM Gibco™

Advanced DMEM (Dulbecco's Modified Eagle Medium) is a widely used basal medium that allows the culture of mammalian cells with reduced Fetal Bovine Serum (FBS) supplementation. Compared to classic DMEM, serum supplementation can be reduced by 50–90% with no change in growth rate or morphology. Cells successfully cultured in Advanced DMEM, with no adaptation, include MDBK, HepG2, COS-7, A549, MDCK, WI-38, and Vero.

Using Advanced DMEM
Advanced DMEM is unique from other media due to the addition of the following ingredients to allow for serum reduction: ethanolamine, glutathione, ascorbic acid, insulin, transferrin, AlbuMAX® I lipid-rich bovine serum albumin for cell culture, and the trace elements sodium selenite, ammonium metavanadate, cupric sulfate, and manganous chloride. Advanced DMEM requires supplementation with 1–5% Fetal Bovine Serum and 4 mM L-glutamine or GlutaMAX™ supplement. Many cell lines do not require adaptation to this media. The FBS concentration must be optimized for each cell line to obtain maximum serum reduction. Advanced DMEM uses a sodium bicarbonate buffer system (3.7 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

RPMI 1640 Medium, low HEPES, low bicarbonate, no glutamine Gibco™

Roswell Park Memorial Institute (RPMI) 1640 medium was originally developed to culture human leukemic cells in suspension and as a monolayer. RPMI 1640 has since been found suitable for a variety of mammalian cells including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. Gibco® RPMI 1640 is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 contains biotin, vitamin B12, and PABA which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations.

Minimum Essential Medium (MEM) α is widely used for mammalian cell culture as well as selection for transfected DHFR negative cells. MEM α can be used with a variety of suspension and adherent mammalian cells, including keratinocytes, primary rat astrocytes, and human melanoma cells. Gibco® MEM α is a modification of Minimum Essential Medium (MEM) that contains non-essential amino acids, sodium pyruvate, lipoic acid, vitamin B12, biotin, and ascorbic acid. MEM α is available without nucleosides for use as a selection medium for DG44 and other DHFR negative cells. This product is made with Earle’s salts.

Ready-to-use format
Gibco® AmnioMAX™ - II Complete Medium is a frozen, 1X medium, ready to use upon thawing. No supplementation is required. Thawed medium can be stored at 2–8°C for up to ten days.

Quality and performance testing
Every lot of Gibco® AmnioMAX™ - II Complete Medium is performance tested by a certified US reference cytogenetics laboratory to ensure consistently superior performance. Pooled primary human amniotic fluid samples are cultured for six days in Gibco® AmnioMAX™ - II Complete Medium before measuring the total number of colonies and total number of mitotic colonies. In addition, each lot is tested for sterility, pH and osmolality.

Unique, optimized formulation
Gibco® AmnioMAX™ - II Complete Medium contains Fetal Bovine Serum (FBS), gentamicin, and L-glutamine to maximize cell attachment and growth. This optimized medium also has an enhanced buffering system that provides greater pH stability during culture manipulations.

Medium 199, HEPES Gibco™

Medium 199 was originally developed for nutritional studies of chick embryo fibroblasts. It has broad species applicability, particularly for cultivation of non-transformed cells. Medium 199 is widely used in virology, vaccine production, and in vitro cultivation of primary explants of mouse pancreatic epithelium, and rat lens tissues. Classic formulation optimized for better performance
For many years, researchers have used a common recipe for making freezing medium: High-glucose Dulbecco's Modified Eagle Medium (DMEM) supplemented with 10% serum and 10% DMSO. Recovery™ Cell Culture Freezing Medium is an optimized version of this classic formulation, using high-quality Gibco® Bovine Serum and Fetal Bovine Serum to provide improved cell recovery and viability after thawing.

Suitability for a wide variety of mammalian cells
Recovery™ Cell Culture Freezing Medium can be used to freeze most mammalian cells cultured in DMEM, DMEM/F-12, MEM, RPMI 1640, and Ham's F-12, including CHO-S, CHO-K1, HEK 293, Jurkat, and NIH 3T3 cells.

Opti-MEM® I medium is also recommended for use with cationic lipid transfection reagents, such as Lipofectamine™ reagent. These additional components allow for a reduction in serum supplementation by at least 50%. Opti-MEM® I medium uses a sodium bicarbonate buffer system (2.4 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.
