Ham's F-12 Nutrient Mix Gibco™

Ham's F-12 Nutrient Mixture (F-12) was designed for serum-free, single-cell plating of Chinese Hamster Ovary (CHO) cells. F-12 has since been used for serum-free growth of CHO cultures as well as serum-supplemented growth of other mammalian cells, including chondrocytes and rat prostate epithelial cells. Using F-12
Compared to other basal media, F-12 contains a wider variety of components, including zinc, putrescine, hypoxanthine, and thymidine. F-12 contains no proteins or growth factors. Therefore, F-12 requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). F-12 uses a sodium bicarbonate buffer system (1.176 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

cGMP manufacturing and quality system
For supply chain continuity, we manufacture F-12 at two separate facilities; one located in Grand Island, NY and the other in Scotland, UK. Both sites are compliant with cGMP-manufacturing requirements, are certified to ISO 13485, and are registered with the FDA as medical device manufacturers.

CHO-S Cells (cGMP banked) and Media Kit Gibco™

GIBCO® CHO_S Cells (cGMP Banked) and Media Kit have been developed for the growth of Chinese Hamster Ovary (CHO) cells and expression of recombinant proteins in suspension culture. Parental CHO-S cells have been produced, banked and tested to meet cGMP quality standards. The cells have been adapted to CD CHO Medium for serum free suspension growth. The CD CHO Medium is made from animal origin free and chemically defined components. It contains no proteins, hydrolysates, or components of unknown composition. The medium is formulated without L-glutamine for greater stability, and without phenol red to minimize potential for estrogen-like effects.

ExpiCHO™ Stable Production AGT Medium Gibco™

Gibco ExpiCHO Stable Production Medium (SPM) is specifically designed to simplify your cell line development process into production with minimal effort and a high degree of confidence. This chemically defined, protein-free, animal origin component-free formulation is fully optimized for large-scale fed-batch culture with ExpiCHO-S cells. ExpiCHO SPM supports an easy transition from transient to stable production, provides high titers without the need for additional medium optimization (see figures below), and works well with existing EfficientFeed C+ supplement.

ExpiCHO SPM is part of the ExpiCHO family, an end-to-end solution that empowers you to carry out your entire workflow from transient to stable expression without having to screen or optimize media and feeds.

Hassle-free scale-up
Simplify your process as you transition into stable clones with ExpiCHO SPM—no cell adaptation, no media optimization, no changes in cell doubling time and peak density. ExpiCHO SPM eases the transition using ExpiCHO-S cells and provides optimal performance with less need for fine-tuning from development scientists to get you to the next step faster. This medium supports high titers under standard fed-batch process conditions in conjunction with EfficientFeed C+ supplement. The power to streamline transfer to manufacturing
ExpiCHO SPM liquid format and AGT dry format demonstrate comparable cell growth and titer performance (see figure below). Therefore, scale-up and manufacturing using AGT dry format is simplified by knowing that the reconstituted AGT dry format will behave similarly to the liquid format used prior to scale-up. The AGT dry format is easy to reconstitute; just add water and it will automatically adjust pH and osmolality. This can help provide efficiencies in supply chain complexity, reconstitution complexity, and, therefore, overall expense.

When to use ExpiCHO Stable Production Medium:
• When you are using the ExpiCHO-S cell line for transient expression and want to scale up
• After you have completed the stable transfection of ExpiCHO-S cells in ExpiCHO Expression Medium. (Follow our recommended protocol.)
• When you do not have the time to perform medium adaptation or optimization

When NOT to use ExpiCHO Stable Production Medium:
• When you have an existing platform cell-line or feeds and are looking for a broad medium to support a wide variety of cell-lines
• When you need a medium for transfection. For transient and stable transfection, ExpiCHO Expression Medium is recommended.

• A stable cloning protocol designed specifically for the ExpiCHO family of products (must be used for optimal performance)

PFHM-II Protein-Free Hybridoma Medium (powder) Gibco™

Gibco® Protein Free Hybridoma Medium II (PFHM II) is a serum-free, protein-free medium specifically developed for the ability to support the growth of hybridomas for antibody production. Gibco® PFHM II features:

RPMI 1640 Medium, HEPES Gibco™

RPMI 1640 Medium was originally developed to culture human leukemic cells in suspension and as a monolayer. Roswell Park Memorial Institute (RPMI) 1640 Medium has since been found suitable for a variety of mammalian cells, including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. This RPMI is modified as follows:

Using RPMI
RPMI 1640 Medium is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 Medium contains biotin, vitamin B12, and PABA, which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations. RPMI 1640 Medium contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 Medium requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 Medium uses a sodium bicarbonate buffer system (2.0 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

Medium 254CF, Kit Gibco™

A sterile, liquid medium for the culture of human epidermal melanocytes. Medium 254CF is Medium 254 prepared without calcium chloride. Calcium chloride is provided as a separate component with each bottle of medium. This basal medium requires the addition of HMGS (S-002-5) or HMGS-2 (S-016-5) prior to use.

Expi293™ Expression Medium Gibco™

Gibco Expi293 Expression Medium is a chemically defined, serum-free, protein-free medium for growth and transfection of suspension-adapted human embryonic kidney (HEK) 293 cells. It is specifically designed as a core component to support high density culture of Expi293F cells in the Expi293 Expression System for scalable transient protein expression.

• Supports growth of suspension 293 cultures to densities of over 1.5 x 10^7 cells/mL
• Transfection compatible serum-free suspension culture medium enables transfection efficiencies of greater than 70% using ExpiFectamine transfection reagent
• Enables sustained, high level expression of high density transiently transfected cultures, achieving yields of up to 1 gram per liter of recombinant protein
• Supports growth and transfection of 293 cells in culture formats of less than 1 mL in multiwell plates to greater than 10L in disposable bioreactors

Expi293 Expression Medium is formulated with GlutaMAX-I reagent. It is ready to use, with no supplementation required. The chemically defined formulation results in high reproducibility and lot-to-lot reliability. Expi293 Expression Medium contains no human or animal-origin components. The medium is not recommended for adherent cell cultures.

LHC-8 Medium (1X), without gentamicin Gibco™

LHC serum-free media are the most widely used products for culture of bronchial epithelial cells. This is a supplemented medium. For maximum flexibility and convenience, we also offer LHC basal medium.

Applications:
Engineered specifically for the growth of bronchial epithelial cells such as IB3-1, S9, and C38, BEAS-2B, BZR, BBM, Het-1A, NHBE (normal human bronchial epithelial cells), and BBE (bovine bronchial epithelial cells) for asthma, allergy, lung cancer, cystic fibrosis, pulmonary, and esophageal cancer research without further supplements.

Performance and quality testing:
Performance tested in a growth assay using BEAS-2B cells.

DMEM, high glucose, no glutamine, no methionine, no cystine Gibco™

Dulbecco's Modified Eagle Medium (DMEM) is a widely used basal medium for supporting the growth of many different mammalian cells. Cells successfully cultured in DMEM include primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells, as well as cell lines such as HeLa, 293, Cos-7, and PC-12. DMEM is unique from other media as it contains 4 times the concentration of amino acids and vitamins than the original Eagle's Minimal Essential Medium. DMEM was originally formulated with low glucose (1 g/L) and sodium pyruvate, but is often used with higher glucose levels, with or without sodium pyruvate.

CD CHO AGT™ (Advanced Granulation Technology) medium is a protein-free, chemically-defined medium optimized for the growth of Chinese hamster ovary (CHO) cells and expression of recombinant proteins in suspension culture. The AGT™ dry media format is designed to provide increased consistency and productivity across all stages of production from development to commercial manufacturing. CD CHO AGT™ medium contains no proteins or peptide components of animal, plant, or synthetic origin, as well as no undefined lysates or hydrolysates.

When to Use CD CHO Medium
• Your GS CHO cells are recommended to be grown in CD CHO medium
• Your cells have been successfully cultured in CD CHO medium in the past and your process is nearing a regulatory filing (DMF available)

When to Consider Another Gibco® Medium

Consider CD FortiCHO™ medium when:
• The cell line you are using is a transfected CHO K1, GS CHO or CHO-S™ cell line
• Maximum batch culture cell densities and protein titers are needed
• You have time to adapt cells into a new medium in an effort to maximize titers
• Cell health at harvest is a priority for downstream processing

Consider CD OptiCHO™ medium when:
• You are using a transfected CHO cell line other than CHO K1, GS CHO or CHO-S™ cell lines
• Optimization of feeding strategies will be part of your base medium selection testing in an effort to maximize productivity
• Your CHO cell line is “finicky," i.e. hard to grow or adapt

Consider CD DG44 medium when:
• You use parental DG44 or DXB11 CHO cells (dhfr-)

Liquid Media Formulations

MEM α (Minimum Essential Medium α) is widely used for mammalian cell culture as well as selection for transfected DHFR-negative cells. Using MEM α
MEM α is a modification of Minimum Essential Medium (MEM) that contains non-essential amino acids, sodium pyruvate, lipoic acid, vitamin B12, biotin, and ascorbic acid. MEM α is available without nucleosides for use as a selection medium for DG44 and other DHFR-negative cells. This product is made with Earle’s salts. MEM α contains no proteins, lipids, or growth factors. Therefore, MEM α requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). MEM α uses a sodium bicarbonate buffer system (2.2 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

Minimum Essential Medium (MEM) is one of the most commonly used of all cell culture media. MEM can be used with a variety of suspension and adherent mammalian cells, including HeLa, BHK-21, 293, HEP-2, HT-1080, MCF-7, fibroblasts, and primary rat astrocytes. Gibco® MEM, developed by Harry Eagle, was based on his earlier formulation of Basal Medium Eagle (BME). Many other modifications of MEM followed, including Glasgow’s MEM, MEM α, DMEM, and Temin’s Modification. MEM is available with Earle’s salts for use in a CO2 incubator, or with Hanks' salts for use without CO2. This product is made with Earle’s salts.
