The complete formulation is available.

Collagenase, Type I, powder Gibco™

Collagenase is a protease that cleaves the bond between a neutral amino acid (X) and glycine in the sequence Pro-X-Gly-Pro, which is found with high frequency in collagen. Collagenase is unique among proteases in its ability to degrade the triple-helical native collagen fibrils commonly found in connective tissues such as skin, tendon, blood vessels, and bone. Collagenase disaggregation is suitable for the culture of human tumors, mouse kidney, human adult and fetal brain, and many other tissues including epithelium. Collagenase is relatively gentle, dissociates well at physiological temperature and pH, and requires no mechanical agitation or special equipment.

Gibco® Collagenase Type I is isolated from Clostridium histolyticum and packaged as a lyophilized, non-sterile powder for research use in cell or tissue dissociation, and organ perfusions. Gibco® Collagenase Type I activity is guaranteed to be greater than 125 units/mg. Compared to other collagenase preparations, Gibco® Collagenase Type I has average levels of collagenase, caseinase, clostripain, and tryptic activities, and is well-suited for the digestion of fat, adrenal, and liver cells or tissues.

CD BHK-21 Production Medium Gibco™

Gibco® CD BHK-21 Production Medium is a chemically defined, protein-free cell culture medium. It is designed to support the growth of BHK-21 cells in serum-free suspension culture. This medium is suitable for large-scale manufacturing applications.

CD BHK-21 Production Medium is manufactured as follows:
WithWithout
• L-glutamine• Phenol Red
• Dry powder formatProtein hydrolysates

Gibco® CD BHK-21 Production Medium is the first cell culture medium supporting complete serum-free and protein-free processes for BHK-21 cells. EpiLife™ Medium, with 60 µM calcium Gibco™

EpiLife is a sterile, liquid medium prepared with 60 µM calcium chloride for the long-term, serum-free culture of human epidermal keratinocytes and human corneal epithelial cells. This basal medium requires the addition of an appropriate growth supplement prior to use. EpiLife Medium greatly extends the in vitro lifespan of human epidermal keratinocytes compared to other serum-free media systems. This basal media is animal origin-free and contains essential and non-essential amino acids, vitamins, other organic compounds, trace minerals, and inorganic salts. It does not contain antibiotics, antimycotics, hormones, growth factors, or proteins. EpiLife Medium is HEPES and bicarbonate buffered and designed for use in an incubator with an atmosphere of 5% CO2 and 95% air.

To support plating and long term proliferation of human keratinocytes, EpiLife Medium must be supplemented with selected hormones and growth factors. We offer a choice of growth supplements for culturing human keratinocytes. Please refer to the EpiLife Medium user guide to determine the best supplement choice for your purposes. The supplements contain all of the growth factors, hormones, and extracts necessary for growth of human keratinocytes in EpiLife Medium.

Gibco® FreeStyle™ F17 Medium supports 293-F cells at a peak density of 6–7 × 106 viable cells per mL, and supports CHO-S cells at a peak density of 8–9 × 106 viable cells per mL. Cells cultured in Gibco® FreeStyle™ F17 can be transfected using a wide variety of transfection reagents, including lipid-based transfection reagents, such as 293fectin™ and Freestyle™ MAX Reagent. Protein expression typically exceeds 75–150 µg/mL IgG in 293-F cells.

Animal origin–free, protein-free, and chemically defined formulation
Gibco® FreeStyle™ F17 Medium is animal origin–free, protein-free, and chemically defined, allowing for easier purification of your protein of interest. Gibco® chemically defined media contain no proteins, hydrolysates, or components of unknown composition.

Scalability in spinner flasks and bioreactors
Protein production using Gibco® FreeStyle™ F17 Medium can be scaled up in spinner flasks or bioreactors. The appropriate spinner or impeller speed and seeding density should be optimized for each system. Depending on the impeller design and speed, it may be necessary to supplement Gibco® FreeStyle™ F17 Medium with additional Pluronic® F-68 to avoid sheer stress in the culture.

cGMP manufacturing and quality system
Gibco® FreeStyle™ F17 Medium is manufactured at a cGMP-compliant facility located in Grand Island, New York. The facility is registered with the FDA as a medical device manufacturer and is certified to ISO 13485 standards.

CHO CD EfficientFeed™ A AGT™ Nutrient Supplement Gibco™

Gibco® CHO CD EfficientFeed™ A nutrient supplement is an animal origin-free, chemically defined complete feed formulation that provides up to 2-5 fold improvement in titers used in fed batch versus batch culture. It contains no proteins, hydrolysates, or components of incompletely defined composition. CHO CD EfficientFeed™ A AGT™ (Advanced Granulation Technology™) Kit Nutrient Supplement provides all the benefits of CHO CD EfficientFeed™ A liquid supplement in a simpler to scale up and reconstitute 2-part kit that is primarily AGT™ dry format material.

The AGT™ format is designed for large scale manufacturing and ease of use. This product is also available in liquid form for small scale use and experimentation.
• Expect up to 2- to 5-fold increases in productivity
• Avoid issues associated with the use of hydrolysates and other alternatives while gaining superior performance
• Simplify scale-up by leveraging the value of AGT™ simple reconstitution
• Achieve performance comparable to original liquid supplement
• Ready for larger scale use with convenient packaging options (10 L and 100 L)

Which EfficientFeed™ Nutrient Supplement to Choose?
Consider the CHO CD Efficient Feed™ Kit when:
• You are using a base medium with less than 6 g/L glucose and no hydrolysate at day 0

Consider CHO CD EfficientFeed™ A supplement when:
• You are currently using CD OptiCHO™ as your base cell growth medium

Consider CHO CD EfficientFeed™ B supplement when:
• You are currently using CD CHO as your base cell growth medium

Consider CD EfficientFeed™ C AGT™ supplement when:
• You want higher productivity in GS CHO and other selected cell lines
• You are currently using CD FortiCHO™ as your base cell growth medium
• You are using a base medium with greater than 6 g/L glucose or a hydrolysate at day 0

Increased Titers
CHO CD EfficientFeed™ supplements have been shown to increase titers 2-5 fold versus batch culture, and are an excellent starting point for process development (Figures 1a & 1b). Furthermore, they outperform competitive alternatives, including less consistent hydrolysates (Figures 2, 3a, & 3b).

Simple Scale-Up and Cost Savings
The ability to scale up in a dry format provides ease of handling and storage in manufacturing and other bioprocess settings. With AGT™ (Advanced Granulation Technology™) dry format, it is possible to store and use the product even more easily and safely than traditional dry format products. CHO CD EfficientFeed™ A AGT™ supplement is offered in 10 L and 100 L packaging that allows scale-up from the 1 L liquid supplement as a seamless process, as well as allow for cost savings when purchasing production level volumes.

Comparable Performance to Liquid Supplement
Equally important as the convenience and simplicity of AGT™ (Advanced Granulation Technology™) dry format is the comparable performance to the liquid CHO CD EfficientFeed™ A version (Figure 4). Both forms have performed similarly in our testing, and the AGT™ version has been used successfully as a prototype in many processes by multiple companies.

AGT™ Format Ease of Use
CHO CD EfficientFeed™ A AGT™ Kit Nutrient Supplement delivers dry format scale-up convenience as well as simple reconstitution with approximately 1% concentrated solution add back. For more details, consult the supporting documents provided below.
