StemPro™ BM Mesenchymal Stem Cells Gibco™

StemPro® BM Mesenchymal Stem Cells are cryopreserved human bone marrow mesenchymal stem cells (BM MSCs). These Off-the-Shelf cells are manufactured to meet Good Manufacturing Practice (GMP) manufacturing standards in a California-licensed facility and available for Research Use Only. They are isolated and expanded in culture to passage 4 under a low oxygen (ischemia tolerant (it)) proprietary clinical-scale manufacturing process using Bovine Growth Serum (BGS)-supplemented media and cryopreserved in an animal origin free cryopreservation medium. The cells are supplied at 1x106 and 5x106 viable cells/vial. A Certificate of Analysis is available for each lot which documents the quality control specifications, test results and donor information.

Immature, High Potency, Low Oxygen-Cultured MSCs
StemPro® BM Mesenchymal Stem Cells are derived from human bone marrow aspirates from qualified, traceable donors. The cells are isolated, cultured and expanded in compliance with GMP manufacturing standards in a California-licensed facility using a proprietary clinical-scale Reduced Oxygen Tension manufacturing process. Cells manufactured in reduced oxygen tension environments result in higher cell yields of highly potent immature stem cells compared to cells expanded in normal oxygen culture conditions.

GeneBLAzer™ MR-UAS-bla HEK 293T Cells Thermo Scientific™

MR-UAS-bla HEK 293T cells contain the ligand-binding domain (LBD) of the human Mineralcorticoid Receptor (MR) fused to the NA-binding domain of GAL4 stably integrated in the GeneBLAzer®UAS-bla HEK 293T cell line. GeneBLAzer®UAS-bla HEK 293T cells stably express a beta-lactamase reporter gene under the transcriptional control of an upstream activator sequence (UAS). When an agonist binds to the LBD of the GAL4 (DBD)-MR (LBD) fusion protein, the protein binds to the UAS, resulting in expression of beta-lactamase. Division Arrested (DA) cells are available in two configurations- an Assay Kit (which includes cells and sufficient substrate to analyze 1 x 384- well plate), and a tube of cells sufficient to analyze 10 x 384-well plates. MR-UAS-bla HEK 293T cells are functionally validated for Z' and EC50 concentrations of aldosterone. In addition, MR-UAS-bla HEK 293T cells have been tested for assay performance under variable conditions, including DMSO concentration, cell number, and substrate loading time. Additional testing data using alternate stimuli are also available.

Human MATE1 (G) SLC Transporter Cells Gibco™

TRANSiPORT Human MATE1 (G) SLC Transporter Cells are HEK293 cells that transiently overexpress the OATP2B1 solute carrier (SLC) transport protein. Solute carriers are a super-family of membrane transporters that can affect pharmacokinetics and drug exposure by governing the transport of solutes in and out of cells. TheMATE1 SLC transporter protein is found in hepatocytes.

Choice of measurement system
Cell-based assays can be performed using radioisotope-labeled compounds, fluorescence-labeled compounds, or non-labeled compounds. The amount of substrate transported into the cells can be measured directly using a liquid scintillation counter, fluorescence plate reader, or LC-MS/MS, thereby allowing direct evaluation of SLC transporter activity.

Assay reliability
Mock (G) SLC Transporter Cells (Cat. No. GM1101G) that do not overexpress the SLC transporter are available for use as a negative experimental control. Some compounds may demonstrate high background levels of transport due to the presence of endogenous transporters or non-specific binding.

StemPro™ Human Adipose-Derived Stem Cells Gibco™

Human Adipose Derived Stem Cells (ADSCs) are isolated from human lipoaspirate tissue and cryopreserved from primary cultures. Before being cryopreserved the ADSCs are expanded for one passage in MesenPRO RS™ Medium (Cat. no. 12746-012) - a revolutionary new reduced serum (2%) medium that results in reduced ADSC doubling times. Each lot of ADSCs originates from a single donor of human lipoaspirate tissue. Each vial of ADSCs contains cells that can differentiate into multiple mature cell phenotypes in vitro including adipocytes, osteoblasts and chondrocytes. ADSCs have demonstrated very similar phenotypic and functional characteristics to that of bone marrow-derived mesenchymal stem cells. The ADSCs express a flow cytometry cell surface protein profile as follows: positive CD29, CD44, CD73, CD90, CD105, CD166; negative CD14, CD31, CD45, Lin1. For optimal ADSC growth and expansion, MesenPRO RS™ Medium is recommended.

RXR alpha-UAS-bla HEK 293T cells contain the ligand-binding domain (LBD) of the human retinoid X receptor-alpha (RXR alpha) fused to the DNA-binding domain of GAL4 stably integrated in the GeneBLAzer®UASbla HEK 293T cell line. GeneBLAzer®UAS-bla HEK 293T cells stably express a betalactamase reporter gene under the transcriptional control of an upstream activator sequence (UAS). Gibco Rat Fetal Neural Stem Cells are isolated and cryopreserved from the cortex of Sprague Dawley Rats at day E14 of gestation. A flexible, ready-to-use, and quality alternative to fresh cells, each vial contains 2 million cells per mL. Upon proliferation in StemPro NSC SFM, >75% of the cells retain their undifferentiated phenotype with the capability to differentiate into neurons, astrocytes, and oligodendrocytes.
