Kupffer Cells Provide for Improved Physiological Modeling
Growing evidence shows that under both normal and pathological conditions, many hepatocyte functions are regulated by substances released from neighboring non-parenchymal cells (NPC). These cells, particularly Kupffer cells, play an important role in the modulation of xenobiotic metabolism in the liver. Kupffer cells secrete potent mediators of the inflammatory response that controls liver inflammation. These cytokine mediators control hepatocyte metabolic rates through direct interactions with phase I and phase II enzymes. Kupffer cell and hepatocyte co-cultures can self-assemble within 72 hours of treatment with pro-inflammatory cytokines or LPS, and can function by effectively modulating P450 expression, thus giving a more physiologically relevant result. Studies indicate that co-culture of hepatocytes with NPCs better represent both normal liver physiology as well as disease states.

Mock (G) SLC Transporter Cells Gibco™

These cells are intended for use as a negative control in experiments utilizing TRANSiPORT solute carrier (SLC) transporter-overexpressing HEK293 cells. These mock cells do not overexpress the SLC transport protein and so can be used to determine levels of endogenous transport activity as well as non-specific binding of compounds being tested.

GeneBLAzer™ ERRα-UAS-bla HEK 293T Cells Thermo Scientific™

ERR alpha-UAS-bla HEK 293T cells contain the ligand-binding domain (LBD) of the human Estrogen related receptor alpha (ERR alpha) fused to the DNA-binding domain of GAL4 stably integrated in the GeneBLAzer®UAS-bla HEK 293T cell line. GeneBLAzer®UAS-bla HEK 293T cells stably express a beta-lactamase reporter gene under the transcriptional control of an upstream activator sequence (UAS). When an agonist binds to the LBD of the GAL4 (DBD)-ERR alpha (LBD) fusion protein, the protein binds to the UAS, resulting in expression of beta-lactamase. Division Arrested (DA) cells are available in two configurations- an Assay Kit (which includes cells and sufficient substrate to analyze 1 x 384-well plate), and a tube of cells sufficient to analyze 10 x 384-well plates. ERR alpha-UAS-bla HEK 293T cells are functionally validated for Z' and EC50 concentrations of XCT790, an inverse agonist. In addition, ERR alpha-UAS-bla HEK 293T cells have been tested for assay performance under variable conditions, including DMSO concentration, cell number, stimulation time, and substrate loading time (data available upon request). Additional testing data using alternate stimuli are also available.

Human OATP1B1 (G) SLC Transporter Cells Gibco™

TRANSiPORT Human OATP1B1 (G) SLC Transporter Cells are HEK293 cells that transiently overexpress the OATP1B1 solute carrier (SLC) transport protein. Solute carriers are a super-family of membrane transporters that can affect pharmacokinetics and drug exposure by governing the transport of solutes in and out of cells. The OATP1B1 SLC transporter protein is found in hepatocytes.

Choice of measurement system
Cell-based assays can be performed using radioisotope-labeled compounds, fluorescence-labeled compounds, or non-labeled compounds. Assay reliability
Mock HEK293 cells (Cat. No. GM1101G) that do not overexpress the SLC transporter are available for use as a negative experimental control. Some compounds may demonstrate high background levels of transport due to the presence of endogenous transporters or non-specific binding.
