Human Suspension Hepatocytes, Polymorphic Donors, (9-12 x 106 cells) Gibco™

Each lot of cryopreserved human hepatocytes is suitable for use in suspension metabolism applications and has been pharmacogenetically characterized as an homozygous null allele (e.g. CYP2D6*4⁄*4) via genotyping analysis (TaqMan®). The commonly tested Phase I (P450) and Phase II (UGT, SULT) drug metabolizing enzymes are analyzed for metabolic activity in each lot. Metabolite formation is measured by standard biochemical assays using LC-MS/MS or HPLC.

The PichiaPink™ Expression Strain Set contains the 4 Pichia pastoris strains for use with the PichiaPink™ Expression System Kits. Each strain comes as a frozen stock in YPD media with 15% glycerol. All PichiaPink™ strains have the ade2 deletion. 3 strains have deletions of two protease genes, PEP4 and PRB1. Select Transformants by Color
The PichiaPink™ Yeast Expression System makes it easy to select for transformed colonies by color. Untransformed PichiaPink™ strains form pink colonies. When grown on plates without adenine, transformed PichiaPink™ strains form white colonies (Fig 1).

Protease-deficient strains reduce the need for protease inhibitors when growing your cells and improve the yield of your protein. Mock (G) SLC Transporter Cells Gibco™

These cells are intended for use as a negative control in experiments utilizing TRANSiPORT solute carrier (SLC) transporter-overexpressing HEK293 cells. These mock cells do not overexpress the SLC transport protein and so can be used to determine levels of endogenous transport activity as well as non-specific binding of compounds being tested.

Mouse (ICR) Inactivated Embryonic Fibroblasts Gibco™

Mouse embryonic fibroblasts (MEFs) are used as a feeder layer when culturing mouse and human embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs). They are isolated from mouse (strain ICR) embryos. The cells are expanded and inactivated by γ irradiation and cryopreserved. Prior to cryopreservation, the cells are tested for viability (>80% post-thaw viability), verified for growth arrest by plating inactivated cells at low density and monitoring for foci of cell growth after 14 days in culture, and also tested for their ability to support growth of mouse embryonic stem cells (mESCs), human embryonic stem cells (hESCs), and marker expression (Oct 4).

Human MATE1 (G) SLC Transporter Cells Gibco™

TRANSiPORT Human MATE1 (G) SLC Transporter Cells are HEK293 cells that transiently overexpress the OATP2B1 solute carrier (SLC) transport protein. Solute carriers are a super-family of membrane transporters that can affect pharmacokinetics and drug exposure by governing the transport of solutes in and out of cells. TheMATE1 SLC transporter protein is found in hepatocytes.

Choice of measurement system
Cell-based assays can be performed using radioisotope-labeled compounds, fluorescence-labeled compounds, or non-labeled compounds. The amount of substrate transported into the cells can be measured directly using a liquid scintillation counter, fluorescence plate reader, or LC-MS/MS, thereby allowing direct evaluation of SLC transporter activity.

Assay reliability
Mock (G) SLC Transporter Cells (Cat. No. GM1101G) that do not overexpress the SLC transporter are available for use as a negative experimental control. Some compounds may demonstrate high background levels of transport due to the presence of endogenous transporters or non-specific binding.

Rapid results

GeneBLAzer™ RXRα-UAS-bla HEK 293T Cells Thermo Scientific™

RXR alpha-UAS-bla HEK 293T cells contain the ligand-binding domain (LBD) of the human retinoid X receptor-alpha (RXR alpha) fused to the DNA-binding domain of GAL4 stably integrated in the GeneBLAzer®UASbla HEK 293T cell line. GeneBLAzer®UAS-bla HEK 293T cells stably express a betalactamase reporter gene under the transcriptional control of an upstream activator sequence (UAS). When an agonist binds to the LBD of the GAL4 (DBD)-RXR alpha (LBD) fusion protein, the protein binds to the UAS, resulting in expression of alpha-lactamase. Division Arrested (DA) cells are available in two configurations- an Assay Kit (which includes cells and sufficient substrate to analyze 1 x 384-well plate), and a tube of cells sufficient to analyze 10 x 384-well plates. RXR alpha-UAS-bla HEK 293T cells are functionally validated for Z' and EC50 concentrations of 9-cis-retinoic acid. In addition, RXR alpha-UAS-bla HEK 293T cells have been tested for assay performance under variable conditions, including DMSO concentration, cell number, stimulation time, and substrate loading time (data available upon request). Additional testing data using alternate stimuli are also available.

Expi293F™ Cells Gibco™

Expi293F human cells are derived from the 293 cell line, and are a core component of the Expi293 Expression System. They are maintained in suspension culture and will grow to high density in Expi293 Expression Medium. Expi293F cells are highly transfectable and generate superior protein yields compared to standard 293 cell lines in transient protein expression.

Expi293F Cells attributes include:

Demonstrated higher protein expression levels than other 293 cells adapted to Expi293 Expression Medium

Frozen cells are supplied in a vial containing 1 ml of cells at 1 x 107 viable cells/ml in Expi293 Expression Medium and 10% DMSO. The cells should be thawed directly into Expi293 Expression Medium. The six-vial multipack eliminates the need to expand and rebank the cells in the lab.

Cells are typically grown in flasks on a shaker platform in a humidified, CO2 cell culture incubator. However, cultures are scalable from volumes of <1 mL in multiwell plates to >10 L in bioreactors. Protein harvest can be performed at 2 to 7 days post-transfection, depending on the protein expressed. For expression of recombinant antibodies, harvest is typically at 5 to 7 days post-transfection. There is no need for culture medium supplementation or batch feeding during expression.
