The ExpiFectamine™ 293 Transfection Kit is specifically designed to work with Expi293F™ cells in the Expi293™ Expression System. The transfection kit consists of the cationic lipid-based ExpiFectamine™ 293 Reagent and ExpiFectamine™ 293 Transfection Enhancers 1 and 2. The enhancers are added 16 to 18 hours post-transfection to boost transfection, cell viability, and protein expression. The protocol for use of ExpiFectamine™ 293 Transfection components is very similar to transfection protocols for other transient expression reagents, such as 293fectin™ and FreeStyle™ MAX.

siPORT™ NeoFX™ Transfection Agent with manual Invitrogen™

This Ambion® lipid-based formulation can be used to efficiently transfect adherent cells with siRNA as they are subcultured—without increased cytotoxicity. It is provided in one tube containing 1 mL.

Reverse-Transfect Cells as They are Plated, Saving a Day
Just add siPORT™ NeoFX to your diluted siRNA, incubate to form transfection complexes, add the complexes to the culture wells, and overlay with cells. This new streamlined protocol, named "neofection", will save researchers a full day of valuable time when compared to traditional plated transfection procedures. The transfection complexes are active and stable even in the presence of serum, so there is no need to remove or replace media following transfection.

Optimized for Lower siRNA Concentrations and High Reproducibility
Recent reports indicate that siRNA concentrations of 100 nM or higher can lead to nonspecific changes in gene expression (off-target effects) in mammalian cultured cells. Reducing the amount of siRNA used for transfections to 1–20 nM minimizes these nonspecific effects, while still providing extensive silencing of the target gene. siPORT™ NeoFX Transfection Agent efficiently transfects low concentrations of siRNA. Furthermore, this agent delivers high performance among multiple wells for replicate samples and between different lots of the transfection agent.

Compatible with a Broad Range of Cell Lines

TurboFect in vivo Transfection Reagent Thermo Scientific™

Thermo Scientific TurboFect in vivo Transfection Reagent is highly effective for DNA plasmid delivery in vivo, while not inducing an inflammatory response. TurboFect in vivo has been assessed for plasmid DNA delivery in both mice and rats and is suitable for DNA administration via various routes, including intravenous, intraperitoneal, and intratumoral injection. The amount of DNA and suggested injection volume will depend on the experimental animal and the route of administration as well as the targeted tissue or organ and the expression vector.

TurboFect in vivo is a sterile solution of a cationic polymer in water. The polymer forms compact, stable, positively charged complexes with DNA. These complexes protect DNA from degradation and facilitate gene delivery in vivo.

Lipofectamine™ Stem Transfection Reagent Invitrogen™

Lipofectamine Stem Transfection Reagent is optimized to achieve maximum efficiency with minimal early differentiation in a wide range of stem cells. It can co-deliver DNA, RNA, and Cas9 ribonucleoprotein (RNP) complexes. Lipofectamine Stem reagent is compatible with a variety of media systems, including feeder-free, helping support and simplify your stem cell culture workflow. Researchers can achieve up to 80% or better transfection efficiency in pluripotent stem cells (PSCs) and neural stem cells (NSCs), and up to 45% in mesenchymal stem cells (MSCs).

Transfecting stem cells without inhibiting cell viability and cell growth can be challenging due to the sensitivity of these cells. Lipofectamine Stem is a low toxicity reagent, requiring low amounts of nucleic acid, allowing your stem cells to stay healthy and continue proliferating without inducing differentiation. Achieve high efficiency gene editing in stem cells
Gene editing applications typically require the ability to co-deliver DNA, RNA, and/or protein into cells. Lipofectamine Stem Transfection Reagent is designed to co-deliver CRISPR/Cas9 complexes into human stem cells for high efficiency gene editing. This one reagent can be used to transfect large DNA plasmids such as all-in-one CRISPR/Cas9 vectors and co-deliver a range of RNAs, such as large Cas9 mRNA with tracr/crRNA complexes or single guide (sg) RNA, and protein such as Cas9 protein combined with tracr/crRNA or sgRNA.

A low-toxicity alternative to electroporation
Transfecting stem cells via electroporation methods can be challenging and cumbersome. Electroporation protocols recommend single cell suspension culture to ensure maximum efficiency and minimal toxicity. Using the reverse transfection protocol, the Lipofectamine Stem Transfection Reagent proves to be an effective and less toxic alternative to electroporation for stem cells. It requires low amounts of nucleic acid to achieve maximum transfection efficiency, on par with electroporation, while supporting continued proliferation without inducing differentiation.

DMRIE-C Transfection Reagent Invitrogen™

DMRIE-C Transfection Reagent is suitable for transfecting DNA and RNA into eukaryotic cells, and is particularly effective for transfecting suspension cells (e.g., Jurkat) and other lymphoid-derived cell lines. Refer to the Cell Lines database at for a list of cell types successfully transfected. DMRIE-C can also be used for in vivo delivery of DNA. DMRIE-C is a 1:1 (M/M) liposome formulation of the cationic lipid DMRIE (1,2-dimyristyloxy-propyl-3-dimethyl-hydroxy ethyl ammonium bromide) and cholesterol in membrane-filtered water.

FreeStyle™ 293-F Cells Gibco™

The FreeStyle™ 293-F cell line is derived from the 293 cell line and is intended for use with the FreeStyle™ MAX 293 Expression System or FreeStyle™ 293 Expression System. FreeStyle™ 293-F cells are adapted to suspension culture in FreeStyle™ 293 Expression Medium. Frozen cells are supplied in and may be thawed directly into FreeStyle™ 293 Expression Medium. The FreeStyle™ 293-F cell line is supplied in a vial containing 1 mL of cells at 1 x 107 viable cells/mL in 90% FreeStyle™ 293 Expression Medium and 10% DMSO. Characteristics of FreeStyle™ 293-F cells:

• Prepared from low-passage Master Cell Bank cultures derived from parental 293-F cells that were re-cloned by limiting dilution. The 293 clonal derived cultures are maintained in serum-free conditions for only 30 to 35 total passages.
• Adapted to high-density, serum-free, suspension growth and may be maintained in FreeStyle™ 293 Expression Medium.
• Demonstrate high transfection efficiencies with FreeStyle™ MAX reagent.
• Suspension cultures may be transfected in FreeStyle™ 293 Expression Medium without the need to change media.
• Permits transfection of cells at large volumes.

Competent Cell Sampler Invitrogen™

The Competent Cell Sampler kit contains a pair of four different strains of chemically competent E. coli cells in convenient One Shot formats. Each single-use vial of cells is ready to use, with no extra pipetting steps or freeze-thaw cycles which reduce transformation efficiency. Use this kit to evaluate which strains best suit the different types of DNA used in your lab.

One Shot Mach1 T1-phage resistant chemically competent E. coli cells have a faster doubling time than typical E. coli cells. As a result, Mach1 colonies are clearly visible within eight hours of plating your transformation on ampicillin selection plates. From an overnight colony, minipreps can be performed after four hours of growth.

Mach1 competent E. coli cells come in One Shot format (Cat. No. C862003) and our MultiShot StripWell and FlexPlate formats for medium and high-throughput applications (C869601 and C8681201).

One Shot OmniMAX2 T1-phage-resistant chemically competent E. coli cells are perfect for use in all cloning applications, including Gateway technology. They offer the highest transformation efficiency in a chemically competent One Shot format with 5 x 109 transformants with 1 µg of control DNA. Coupling the high transformation efficiency with the elimination of the mcrA, mrr, mcrBC, and hsdRMS restriction systems allows for the construction of more representative genomic libraries in a convenient chemically competent format.

OmniMAX 2 cells are offered in our One Shot format (C854003) and in our MultiShot FlexPlate format for medium to high throughput applications (C8581201).

OneShot Stbl3 chemically competent cells reduce the frequency of unwanted homologous recombination and are perfect for cloning unstable DNA such as lentiviral DNA containing long terminal repeats (LTRs). The DNA yield from Stbl3 cells is often >10-fold higher than standard cloning strains.

Stbl3 chemically competent cells are available in the One Shot format (C737303) and in the MultiShot StripWell and FlexPlate formats (C739601 and C7381201) for medium to high throughput applications.

One Shot TOP10 cells are perfect for a broad range of cloning techniques. They are often included with many of our cloning and expression kits like TOPO and Gateway cloning kits and complement our GeneArt High-Q Strings and Strings DNA fragments and libraries. One Shot TOP10 competent E. coli come with a transformation efficiency of 1 x 109 cfu/µg supercoiled DNA and are ideal for high-efficiency cloning and plasmid propagation. They allow stable replication of high-copy number plasmids.

TOP10 chemically competent cells are available in the One Shot format (C404003), in the MultiShot StripWell and FlexPlate formats (C409601 and C4081201) for medium to high throughput applications.

The Silencer® siRNA Starter Kit includes a high-quality, lipid-based transfection agent, siPORT™ NeoFX™, that effectively delivers siRNAs to a broad range of cell lines. It can be used for reverse transfection, a streamlined protocol that eliminates an entire day from the RNAi experimental procedure; it can also be used with standard transfection protocols. This reagent is not sensitive to serum and efficiently transfects low concentrations of siRNA. The kit also includes a well-characterized GAPDH siRNA, which has been shown to strongly down-regulate GADPH in human, mouse, and rat cells, as well as a nontargeting negative control siRNA, Silencer® Negative Control #1 siRNA. Because GAPDH is ubiquitously expressed and is easy to detect at both the mRNA and protein level, it is an ideal target for a positive control siRNA.

Three Detection Systems
The kit includes three different systems for monitoring GAPDH siRNA-induced knockdown. An anti-GAPDH antibody is provided to assess GAPDH protein levels by western blot or immunofluorescence. The KDalert™ GAPDH Assay is also provided to quickly measure GAPDH enzymatic activity. This assay can also be used to assess cell viability. Finally, RT-PCR primers are included, for real-time quantitative RT-PCR detection of GAPDH mRNA using SYBR® Green. The ViraPower™ HiPerform™ T-REx™ Gateway® Expression System includes all the components needed to generate lentivirus, including vector kit, 293FT cell line, and the support kit. Transfect primary cells efficiently
Primary cells are often difficult to transfect. TurboFect Transfection Reagent is a sterile solution of a proprietary cationic polymer.
