The Anza™ Restriction Enzyme Cloning System employs a single buffer and protocol for all Anza restriction enzymes. The single Anza buffer allows multiple restriction enzymes to be used simultaneously in one reaction. This eliminates the need for sequential digests and the DNA clean-up necessary when changing buffers. Additionally, the Anza buffer and restriction enzymes have been designed for flexibility in digestion times. Digestions of up to 16 hours can be carried out without star activity.

Anza Red Buffer, which contains a density gradient and tracking dyes, is supplied for added convenience. Digestion reactions are as efficient in the Anza Red Buffer as in the Anza Buffer. Use of the Anza Red Buffer helps reduce pipetting steps when preparing multiple digestion reactions and a subsequent step includes agarose gel electrophoresis. For applications that require product analysis by fluorescence excitation, the colorless Anza Buffer without the tracking dyes is recommended.

Reaction Buffer with MgCl2 for DNase I (10X) Thermo Scientific™

Thermo Scientific 10X Reaction Buffer with MgCl2 is used with RNase-free DNase I, an endonuclease that digests single- and double-stranded DNA.

Fluorescein Di-β-D-Glucuronide (FDGlcU) Invitrogen™

Fluorescein di-β-D-glucuronide (FDGlcU) is colorless and nonfluorescent until it is hydrlozed to the monoglucuronide and then to the highly fluorescent fluorescein.

Ambion™ Recombinant RNase A Invitrogen™

Ambion® Recombinant RNase A for the production of plasmid DNA that is substantially free of host RNA is now available for research applications. For large batch sizes, the use of RNase is still the only effective technique applicable to remove host RNA. Other methods such as selective precipitation, gel filtration, chromatography, and endogenous E. coli RNase digestion have only been effective for small-scale or bench-scale purification. This new product was designed specifically to minimize the risk of introducing pathogens into bioprocessing. Recombinant RNase A is rigorously tested for contaminating nonspecific endonuclease, exonuclease, and protease activity.

Vivid™ BOMR Substrate Thermo Scientific™

Vivid® Substrates are blocked dyes that yield a minimal fluorescence signal until oxidation occurs. Cytochrome P450 metabolism at either of two potential oxidation sites releases the highly fluorescent product. They have superior fluorescence, solubility, and kinetic properties compared to conventional fluorogenic probes. This results in higher sensitivity, greater signal-to-noise ratio, and better assay reproducibility. When used in combination with CYP450 BACULOSOMES® Plus Reagents as part of the Vivid® CYP450 Screening Kits, the Vivid® Substrates enable rapid, robust detection of cytochrome P450 inhibitors. This particular substrate (Vivid ® BOMR Substrate) yields a product that emits a red fluorescence.

Thermo Scientific FastDigest MreI restriction enzyme recognizes CG^CCGGCG site and cuts best at 37°C in 5–15 minutes using universal FastDigest Buffer.

Thermo Scientific FastDigest MreI is one of an advanced line of fast restriction enzymes that are all 100% active in the universal FastDigest and FastDigest Green reaction buffers.

The universal buffer allows rapid single-, double-, or multiple DNA digestion within 5–15 minutes eliminating any need for buffer change or subsequent DNA clean-up steps. Ambion® RNase A is an endonuclease that specifically cleaves 3' of U and C residues. Affinity Purified RNase A is intended for critical applications when the absence of DNase and other nonspecific nuclease activities is essential. Supplied in one tube containing 1 mg (1 mg/mL). Note: This preparation contains RNase B, a carbohydrate isoform of RNase A. RNase A is rigorously tested for contaminating nonspecific endonuclease, exonuclease, and protease activity.
