UltraPure™ SDS Solution, 10% Invitrogen™

UltraPure™ 10% SDS Solution is a premixed aqueous solution containing 10% sodium dodecyl sulfate (SDS). SDS is a protein denaturant used in polyacrylamide gel electrophoresis for molecular weight determinations. It also is used to dissociate nucleic acid-protein complexes in DNA extraction protocols, to disrupt cell membranes, and to prepare prehybridization and hybridization solutions. UltraPure™ 10% SDS Solution is filtered and ready to use.

UltraPure™ TEMED Invitrogen™

UltraPure™ TEMED (N,N,N´,N´-tetramethylethylenediamine) is the most commonly used accelerator for the polymerization of acrylamide gels.

UltraPure™ Sodium Dodecyl Sulfate (SDS) Invitrogen™

UltraPure™ Sodium Dodecyl Sulfate (SDS) is a detergent that functions as a protein denaturant. It is used in the determination of protein molecular weight by polyacrylamide gel electrophoresis. It is also widely used to disrupt cell walls and dissociate nucleic acid-protein complexes in DNA and RNA extraction protocols.

UltraPure™ Agarose-1000 Invitrogen™

UltraPure™ Agarose-1000 is a polysaccharide (see structure) used for size-based separation of nucleic acids in agarose gel electrophoresis. UltraPure™ Agarose 1000 is specifically formulated for the high- resolution separation of small (<1,000 bp) DNA, RNA, and PCR fragments. This standard melting temperature agarose can resolve DNA fragments differing in as little as 10 bp in size. Benefits of using UltraPure™ Agarose 1000:

• Ideal for analysis and recovery of small DNA and RNA fragments
• Strong gel structure allows for better handling
• Improved clarity allows for enhanced visibility of fragments

Improved packaging
The new environmentally friendly packaging uses 75% less plastic than the original bottles (see figure). This means less energy and raw material used in manufacture, and less waste in landfills. Additionally, the easy-pour spout reduces the likelihood of spills and contamination.

Performance and quality testing
The performance of UltraPure™ Agarose 1000 is evaluated to satisfy specifications in appearance, moisture, gel strength, gelling temperature, melting temperature, sulfate content, electroendosmosis, and DNase/RNase assay.

Note: UltraPure™ Agarose 10000 (Cat. No. 16550-100) is a replacement for the item previously sold under Cat. No. 10975-035.

MagMAX™ Cell-Free Total Nucleic Acid Isolation Kit Applied Biosystems™

The MagMAX Cell-Free Total Nucleic Acid Isolation Kit is designed for enrichment of all nucleic acid species present in cell-free biological samples such as plasma. The kit utilizes MagMAX magnetic-bead technology, ensuring reproducible recovery of high-quality nucleic acid that is suitable for a broad range of applications, including real-time PCR, digital PCR, and next-generation sequencing.

The magnetic bead–based purification format allows you to easily process a wide range of sample input volumes (1 mL to 6 mL). When used with the KingFisher Duo Prime or KingFisher Flex Magnetic Particle Processor, 6 to 24 plasma samples of 2-mL input volume can be isolated at once. Alternatively, samples can be processed manually with the use of a magnetic stand.

Features of the MagMAX Cell-Free Total Nucleic Acid Isolation Kit include:
• Automation-ready, phenol-free extraction
• Fast procedure allows 6 to 24 samples to be processed in 90 minutes or less when used with the KingFisher Duo Prime or KingFisher Flex Magnetic Particle Processor with a 24 Deep Well Head
• Flexible protocol accommodates sample volume inputs ranging from 1 mL to 6 mL of plasma
• No need for carrier RNA
• Elution volumes ranging from 15 µL to 60 µL
• Compatible with the Oncomine Lung cfTNA Research Assay

Go beyond cell-free DNA and get the total picture
Isolating and analyzing circulating cell-free total nucleic acid (cfTNA) from plasma and other cell-free samples serves as a non-invasive method to detect and monitor targets associated with certain disease states such as cancer.

The first step in attaining this valuable information is efficient nucleic acid isolation that specifically recovers the circulating nucleic acid species that contains information related to the disease state. The importance of cell-free DNA (cfDNA) is becoming established, but it may not tell the whole story. Cell-free RNA (cfRNA) sequences serve as critical biomarkers as well in the form of microRNAs and gene fusions. This inclusion of the cfRNA with the cfDNA provides an opportunity for researchers to expand their studies and advance human health.

The MagMAX Cell-Free Total Nucleic Acid kit is developed by the same Thermo Fisher Scientific scientists that brought you the MagMAX Cell-Free DNA Isolation Kit. The difference is the inclusion of all cfRNA species in the eluate, allowing you to increase the amount of information available from a single cell-free sample. Just like the cfDNA Isolation Kit, the cfTNA kit is designed for both manual and automated isolation of nucleic acid from a wide sample volume input range of 1 to 6 mL. The phenol-free, magnetic-bead format makes the kit fully amenable to automated isolation.

Thanks to the scalable format that allows for various sample inputs, it is possible to vary the number of samples processed with a single kit. For example, enough reagents are provided to process 50 samples of 2 mL plasma each. Alternatively, with the same kit it is possible to process 25 samples of 4 mL plasma each. Higher and lower input volumes are possible and the number of samples that can be processed will vary accordingly. This unique product attribute allows you to tailor the product to your specific needs.

Benefits of MagMAX magnetic bead–based purification
Magnetic beads offer many benefits compared to other technologies for isolating nucleic acid. Beads bind the DNA more efficiently than glass-fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of filter clogging due to cellular particulates in samples. This clogging issue is of particular concern with protein-rich, large-volume samples such as plasma that are commonly used in circulating cell-free nucleic acid studies.

MagMAX™ Cell-Free DNA Elution Solution Applied Biosystems™

This solution is included in the MagMAX Cell-Free DNA Isolation Kit (Cat. No. A29319). It is made available separately for applications that require more Elution Solution than is provided in the kit.

Benefits of MagMAX magnetic bead–based purification
The magnetic bead–based purification format of the kit allows for easy scaling of the number of samples processed either manually with a magnetic stand or with a magnetic particle processor such as the KingFisher™ Duo Prime or KingFisher™ Flex processors.

Magnetic beads offer many benefits compared to other technologies for isolating nucleic acids. Beads bind the RNA and DNA more efficiently than glass fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of undigested tissue particulates clogging these items during the extraction process. This clogging issue is of particular concern with fibrous plant tissue samples that that are not fully lysed during the disruption and homogenization steps.

Optimized bead surface chemistry
The Dynabeads™ MyOne™ Carboxylic Acid utilized in the MagMAX Plant DNA Kit has an optimized surface chemistry with high specific surface area that allows efficient kinetics and high sensitivity in nucleic acid capture. These aspects make Dynabeads magnetic technology ideal for the recovery of the DNA targets found in a variety of plant samples. Once captured, the DNA is then eluted in 150 µL and ready for downstream applications.
