DNAzol® Reagent is a complete and ready-to-use organic reagent for the isolation of genomic DNA from solid and liquid samples of animal, plant, yeast, and bacterial origin. The DNAzol® Reagent procedure can be completed in typically 10–30 minutes with DNA recoveries of 70–100%. DNAzol® Reagent is:

Efficient isolation of genomic DNA from a variety of sample types
The DNAzol® Reagent procedure uses a novel guanidine-detergent lysing solution that permits selective precipitation of DNA from cell lysate. 1 mL of DNAzol® Reagent can be used to isolate genomic DNA from 1–3 x 107 cells, from 0.1 mL of whole blood, or from 25–50 mg tissue.

Rapid isolation and high recovery of genomic DNA
The DNAzol® Reagent combines reliability, efficiency, and simplicity in its DNA isolation protocol. During isolation, a biological sample is lysed (or homogenized) in DNAzol® Reagent, and the genomic DNA is then precipitated from the lysate with ethanol. Following an ethanol wash, DNA may be solubilized in either water or 8 mM NaOH. The entire procedure can be completed in 10–30 min with DNA recovery of 70–100%.

Isolated DNA can be used for a number of downstream applications
Genomic DNA extracted using the DNAzol® procedure is suitable for a variety of applications, including Southern blotting, cloning, PCR, restriction endonuclease digestion, and dot blot hybridization.

MPC™-6 (Magnetic Particle Concentrator) Thermo Scientific™

The MPC™-6 (Magnetic Particle Concentrator) holds six tubes with variable diameters (10–30 mm). Optimal working volume: 5–15 mL. Magnetic particle concentrators are designed to obtain optimal binding of Dynabeads™ with targets (e.g., cells, proteins, and nucleic acids).

The ChargeSwitch® Plasmid Yeast Mini Kit provides an efficient method for purifying high-quality plasmid DNA from 1 ml of Saccharomyces cerevisiae grown in selective minimal media. This kit typically yields up to 25 ng of plasmid DNA. Data generated from transformation and PCR experiments using ChargeSwitch® purified DNA shows improved performance versus DNA purified by other methods (Figure 1). This kit includes all the necessary reagents for the number of preps listed and requires the use of a magnetic accessory. GeneJET Plasmid Maxiprep Kit Thermo Scientific™

Thermo Scientific GeneJET Plasmid Maxiprep Kit, sufficient for 25 preps, utilizes silica-based membrane technology in the form of convenient spin column. Each prep recovers up to 750 µg of high copy plasmid DNA that can be used in a wide variety of molecular biology procedures, such as restriction endonuclease digestion, PCR, cloning, transformation, automated sequencing, in vitro transcription, and transfection of robust cell lines.

UltraPure™ Salmon Sperm DNA Solution Invitrogen™

UltraPure™ Salmon Sperm DNA Solution is a ready-to-use, sheared DNA solution that is used directly in the preparation of prehybridization and hybridization solutions. This DNA solution is prepared from highly pure, phenol/chloroform-extracted DNA and DNase-free, RNase-free (DEPC-treated), distilled, deionized water. The DNA is sheared to an average size of ≤2,000 bp. The concentration is adjusted to 10 mg/mL.

Applications
Such solutions are used to block the nonspecific attachment of probe to the surface of a membrane.

Performance and quality testing
Concentration, size verification, viscosity, and ultraviolet absorbance (A260/A280) are analytically tested.

Designed for performance
The column design is based on the description of gel filtration for the purification of DNA from nick translation reactions by Sambrook, et.al. Each Centri-Sep™ unit consists of a specially fritted microfuge tube, dry gel, a wash tube, and a sample collection tube, all specifically designed for this purpose.

Excellent recovery
The Centri-Sep™ gel provides excellent recovery of DNA fragments with sizes greater than 16 base pairs or a 25-mer, while removing >98% of salts, NTPs, and other unwanted low molecular weight impurities.

Versatile
The column gel is hydrated with either reagent-grade water or a buffer of your choice, and spun in a microcentrifuge or swinging-bucket centrifuge to remove the intersititial fluid. Improved Packaging and Configuration

Easy-DNA™ gDNA Purification Kit Invitrogen™

The Easy-DNA™ Kit is a fast and easy way to isolate high-quality, high-molecular weight genomic DNA (gDNA) from a wide range of cells and tissue types. Sample sizes can range from a single hair follicle up to 1 gram of mammalian tissue. Other samples tested include mouse tails, plant leaves, yeast, and E. coli cells. These samples yield high-quality DNA with an average size between 100 kb and 200 kb, which is suitable for PCR, DNA hybridization, genomic DNA library construction, and other applications. Advantages of the Easy-DNA™ Kit include:

Reliability—high-quality genomic DNA from large or small samples of cells, tissue, plant, yeast, E. coli, blood, and even hair follicles
Simplicity—for most sample types, only 4 steps, typically requiring less than 90 minutes are needed to isolate high-quality DNA
Versatility—one kit is supported by optimized protocols for a variety of cell and tissue types

Simple workflow, quality results
Optimized protocols for a variety of cell and tissue types are included in the instruction manual supplied with the kit. Generally, small-scale gDNA preparations are performed in microfuge tubes, in which samples are mixed with Lysis Solution and incubated at 65°C for 10 minutes. After extraction with chloroform (not included in kit), precipitation, and reconstitution of the DNA pellet, the preparation is complete. The kit contains a protein degradation reagent (for use with high-protein–containing samples such as connective tissue), mussel glycogen for use as a co-precipitant for dilute DNA samples, and an RNase solution.

ChargeSwitch™ gDNA Normalized Buccal Cell Kit Invitrogen™

The ChargeSwitch® gDNA Normalized Buccal Cell Kit uses magnetic bead-based technology to allow rapid and efficient isolation of genomic DNA from human buccal swabs in less than 15 minutes after lysate preparation.

Rapid Purification Protocol
The ChargeSwitch® gDNA Normalized Buccal Cell Kit allows purification of genomic DNA from human buccal cell swabs or pelleted cells from a mouthwash in less than 15 minutes after lysates preparation. ChargeSwitch® gDNA Normalized Buccal Cell Kit uses magnetic beads with a unique, ionizable functional group whose affinity for nucleic acids is pH-dependent to facilitate nucleic acid purification. High-quality genomic DNA is purified at a normalized concentration of 1-3 ng/ µl in a total volume of 150 µl (Figure 1) by eliminating the need for lengthy quantitation and dilution steps. Samples can be stored for up to 2 weeks at 4°C before processing without a noticeable loss in DNA yield or quality.

Note: DNA yield varies and is dependent on several factors including the technique of the person taking the swab, whether the donor is a high or low shedder, and the type of swab used.

Validated for Single Tandem Repeat (STR) Profiling
After employing the ChargeSwitch® gDNA Normalized Buccal Cell Kit, purified gDNA is suitable for use in STR analysis of poor or environmentally degraded samples, and for downstream applications such as PCR.

Compatible with Single Tube and High-Throughput Protocols
The ChargeSwitch® gDNA Normalized Buccal Cell Kit is designed for automated processing of both small numbers of samples individually, as well as large numbers of samples using a liquid handling robot or a multi-channel pipette.

For processing up to 24 individual samples at a time the MagnaRack™ two-piece magnetic separation tube rack is required. This rack consists of a magnetic base station and a removable tube rack, which can hold up to 24 microcentrifuge tubes and can be used with most ChargeSwitch® products.

For high-throughput applications utilizing 96 well blocks, the 96-Well Magnetic Separator is required. This rack can hold and separate up to 96 magnetic bead samples in a deep well plate when placed on top of the magnetic base station.
