mirVana™ miRNA Inhibitor let-7c Positive Control
The mirVana™ miRNA Inhibitor let-7c Positive Control is intended for use as a positive control for mammalian cell culture experiments using mirVana™ miRNA Inhibitors. Endogenous let-7c miRNA has been shown to negatively regulate HMGA2 mRNA in cultured cells. HMGA2 is a ubiquitously expressed, nonhistone, chromatin protein that can modulate gene expression through changes in chromatin architecture. let-7c miRNA down-regulates levels of HMGA2 mRNA. When transfected into human and mouse cell lines, let-7c miRNA inhibitor blocks endogenous let-7c miRNA, resulting in increased levels of HMGA2 mRNA. Thus, let-7c miRNA inhibitor activity can be monitored in human or mouse cells using real-time reverse transcription PCR (RT-PCR) to detect HMGA2 mRNA.

First Generation Inhibitor Controls
Our first-generation miRNA inhibitors (and controls), Ambion® Anti-miR™ miRNA Inhibitors, have lower specificity and potency, plus a lower strand specificity compared to the new mirVana™ miRNA Inhibitors. These first-generation inhibitors are recommended for completing ongoing in vitro miRNA studies. What are Silencer® Select siRNAs?

The Pre-miR™ hsa-miR-1 miRNA Precursor is intended for use as a positive control in experiments utilizing Pre-miR miRNA Precursors. This precursor was chosen as a positive control because, upon delivery into cells, it effectively down regulates the expression of twinfilin-1, also known as PTK9, at the mRNA level (60-95% reduction. About Pre-miR miRNA Precursors

Pre-miR™ miRNA Precursor Molecules are small, chemically modified double-stranded RNA molecules designed to mimic endogenous mature miRNAs. These ready-to-use miRNA mimics, which are similar to, but not identical to siRNAs, can be introduced into cells using transfection or electroporation parameters similar to those used for siRNAs. They enable detailed study of miRNA biological effects via gain-of-function experiments (Figure 1). Positive and negative control Pre-miR miRNA Precursors (listed above) or the Pre-miR™ Precursor Starter Kit can be used to optimize transfection of Pre-miR miRNA Precursors.

Pre-miR miRNA Precursor Molecules have been carefully designed and modified to ensure that the correct strand, representing the desired mature miRNA, is taken up into the RNA-induced silencing-like complex responsible for miRNA activity (Figure 2). In contrast to miRNA expression vectors, these synthetic molecules can be used in dose response studies due to their introduction directly into the cell by transfection or electroporation. Thanks to their small size, they are easier to transfect than vectors, and can be delivered using conditions identical to those used for siRNAs.

What are Ambion® In Vivo siRNAs?
Ambion® In Vivo siRNAs are designed using the proven Silencer® Select algorithm and incorporate chemical modifications for superior serum stability with in vivo delivery. The added serum stability does not compromise the performance of the siRNAs. Ambion® In Vivo siRNAs have been shown to be non-toxic in vivo (mouse) and non-immunogenic (cell-based assays). Factor VII (FVII) Positive Control siRNA
Ambion® In Vivo Factor VII siRNA, In Vivo Ready, provides a positive control for experiments involving Ambion® In Vivo siRNA delivery to mice. Factor VII (FVII⁄F7; also known as proconvertin) is a vitamin K-dependent serine protease that functions as a central protein in the coagulation cascade. FVII is synthesized in the liver and secreted to the plasma where it circulates in an inactive form. After delivery of FVII-targeting siRNA to the liver, knockdown can be measured by quantifying FVII protein levels in blood by robust, commercially available chromogenic assays. Moreover, blood samples can be collected from the same animal at multiple time points post-siRNA injection. These features enable much more rapid and efficient knockdown measurements for FVII than for gene targets that require quantitation of mRNA levels in liver tissue, making FVII an ideal control for in vivo siRNA delivery.

In Vivo Ready Controls
Our "In Vivo Ready" Control siRNAs are subjected to that extra level of purification and testing required for the introduction of siRNAs into animals. After HPLC purification and annealing, each siRNA is further purified utilizing a process that removes excess salt via a semi-permeable membrane. The result is highly pure siRNA with minimal salt content, suitable for in vivo applications. In vivo siRNAs are then sterile filtered, and tested for the presence of endotoxin.
