The 3.5K molecular-weight cut off (MWCO) cassettes use a cellulose membrane that is rated to completely retain proteins and other macromolecules that are larger than 3500 daltons while allowing removal of buffer salts and small contaminants. The cassette membrane is composed of low-binding regenerated cellulose. The membranes and cassettes are manufactured using clean-room conditions to ensure that are contaminant-free.

The Slide-A-Lyzer G2 Dialysis cassettes are available in five precise membrane MWCOs (2K, 3.5K, 7K, 10K and 20K) for dialyzing sample volumes up to 70 mL. The Slide-A-Lyzer G2 Dialysis Cassettes allow samples to be directly injected with a syringe or with a pipette through the unique sample port. The Slide-A-Lyzer Cassette design allows air to be removed from inside the cassette to maximize the surface area-to-volume ratio, providing better dialysis performance than dialysis tubing and tube-based systems. The sample is easily recovered through either the pipette or syringe sample ports.

These devices are easier to use than traditional dialysis tubing, providing both faster buffer exchange and more complete sample recovery. Slide-A-Lyzer Cassettes can be used for a wide range of applications including low molecular-weight contaminant removal, buffer exchange, desalting, equilibrium dialysis, and sample concentration. Liquid samples are easily added and removed by penetrating the self-sealing gasket with a hypodermic needle attached to a syringe. No knots, clips or caps are needed to seal the units and prevent leaking and sample-loss during the dialysis procedure.

Gamma-irradiated Dialysis Cassettes:
Gamma-irradiated Slide-A-Lyzer Dialysis Cassettes are ideal for researchers culturing cells and microorganisms; purifying viruses, DNA and RNA; or performing sample preparation for other applications requiring conditions that minimize the risk of sample contamination. The gamma-irradiated cassettes are treated with 20-36 kGy of gamma radiation, a dosage that has been determined to sterilize the units based on both trypticase soy broth medium for identifying total growth and fluid thioglycollate medium for identifying anaerobic growth. However, a confirmation of sterility is not provided for individual lots, as no further sterility testing is conducted post irradiation.

Slide-A-Lyzer Dialysis Cassettes are constructed from two sheets of low-binding, regenerated-cellulose dialysis membrane that are hermetically sealed on either side of a silicone-like gasket inside an inert plastic frame. The membrane and cassette materials are compatible with most common laboratory chemicals and buffers.

Pierce Strong Ion Exchange Spin Columns use membrane-adsorption as a chromatographic method to fractionate proteins based on their charge differences. The matrix has a highly porous structure with pores larger than 3000nm, providing proteins easy access to the membrane's charged ligands. Therefore, adsorptive membranes maintain high efficiencies at high-flow rates and when fractionating large biomolecules with small diffusivities.

Ion exchange chromatography separates molecules based on differences in their accessible surface charges. This technique is widely used in the pre-fractionation or purification of a target protein from crude biological samples. Novel membrane-based ion exchange chromatography is attracting attention because of its advantages over resin-based column chromatography. Membrane-based ion exchange chromatography has shorter diffusion times than resin-based chromatography. Interactions between molecules and active sites on the membrane occur in a convective manner through pores, which shortens the diffusion time compared with fluid inside the pores of a resin particle. Also, the relatively mild binding and eluting conditions of this separation method produce high protein recovery with intact biological activity.

SnakeSkin Dialysis Tubing, 3.5K MWCO, is composed of regenerated-cellulose dialysis tubing and supplied as an open, pleated (telescoped) tube. SnakeSkin Dialysis Tubing is supplied in eight inch (20 cm) sticks containing 35 feet of dialysis tubing having a 16, 22, or 35mm circular internal diameter (I.D.). With similar capacity to 10.5 meters of 23 to 50 diameter dry flat width tubing, the hydrated SnakeSkin Dialysis Tubing holds ~2 to 10 mL of sample per centimeter of length.

The easy-open, pleated format of SnakeSkin Dialysis Tubing was developed to simplify large volume sample dialysis. To use, simply pull out and cut off the required length of tubing, fold over one end of tubing and apply a dialysis clip to close it, add sample at the open end and use a second clip to close the remaining end. Clips are sold separately. In comparison, many traditional dialysis tubing membranes that are supplied as dry, flattened rolls that are difficult to open and often require extensive soaking before they can be used.

The SnakeSkin membrane-pleating process does not change the tubing's MWCO. Also, any low MW contaminants present are removed during the dialysis process. Because SnakeSkin Dialysis Tubing is made from the same type of regenerated cellulose as flat tubing, its dialysis performance matches that of conventional tubing.

The RED Device is an apparatus for performing equilibrium dialysis experiments in a high throughput, automation-compatible format, and consists of disposable inserts and a base plate formatted to a standard microplate footprint. It has been extensively validated for plasma-binding assays and produces results consistent with those reported in the literature. The RED Device also offers significant improvements in automation, time requirements, versatility and product reliability compared to other commercially available equilibrium dialysis systems.

Features of the RED Device Inserts:
Each single-use, disposable insert is made of two side-by-side chambers separated by a vertical cylinder of dialysis membrane (in 8K or 12K MWCO) validated for minimal nonspecific binding. This format requires no extensive assembly steps or specialized equipment, and each chamber or well is easily accessible from the top of the insert after insertion in the base plate. Additionally, the high surface-to-volume ratio of the membrane compartment allows rapid dialysis, where equilibrium can be reached in 4 hours with high levels of reproducibility and accuracy. In many cases, experiments can be completed in less than 100 minutes.

These 3.5K MWCO dialysis devices are available in three sizes (0.1, 0.5 and 2 mL capacities), and are designed to allow easy sample addition and removal using a standard laboratory pipette. The MINI Devices are effective and convenient for accomplishing typical sample desalting or buffer exchange in 4 to 8 hours. Unlike other small sample separation devices, there is no equipment to assemble, no need for syringe adapters and no laborious steps necessary to manipulate the small volume sample. The regenerated cellulose membrane is compatible with a number of common chemicals and buffers.

Slide-A-Lyzer MINI Dialysis Devices were developed as a convenient means for dialysing small-volume samples of nucleic acids and proteins without undue sample loss. Low molecular weight contaminant removal, buffer exchange, desalting, equilibrium dialysis, micro conjugation reactions, and concentration can be accomplished with these devices. Slide-A-Lyzer MINI Dialysis Devices are manufactured in a HEPA filter/clean environment and auto-bagged after assembly to ensure that they are free of contaminants and ready to use.

The 0.5 mL and 2 mL MINI Dialysis Devices fit in and are capped by inserting them into the included 15 mL and 50 mL conical tubes, respectively. The tubes can be securely placed in standard laboratory racks or shakers; this feature saves space, minimizes the risk of spills or contamination, and eliminates the need for floats or large beakers of buffer. Using these MINI Dialysis Devices, low molecular weight contaminant removal, buffer exchange and desalting, can be accomplished within 4 to 8 hours with efficiencies, rates, and recoveries very similar to conventional dialysis using a large dialysate volume (view data).

The 0.1 mL MINI Dialysis Devices are designed to be capped (caps included) and inserted into standard 1.5 mL micro centrifuge tubes. Alternatively, as many as 25 MINI Devices can be placed in a Slide-A-Lyzer MINI Dialysis Device Float for dialysis of multiple inside a large beaker. Using the 0.1 mL MINI Device, 100 µL of pH 2.8 glycine buffer can be equilibrated to 1 liter of pH 9.4 bicarbonate buffer in less than 10 minutes.

As with all dialysis applications, the observed rate of dialysis for each sample used with these devices depends on a variety of factors including membrane to sample volume, temperature and agitation rates; as well as the molecular weight, shape, degree of hydration, and charge of the molecules passing through the membranes and the pH, ionic strength, and solvent characteristic of the buffers being used.

The 3.5K molecular-weight cut off (MWCO) cassettes use a cellulose membrane that is rated to completely retain proteins and other macromolecules that are larger than 3500 daltons while allowing removal of buffer salts and small contaminants. The cassette membrane is composed of low-binding regenerated cellulose. The membranes and cassettes are manufactured using clean-room conditions to ensure that are contaminant-free.

The Slide-A-Lyzer G2 Dialysis cassettes are available in five precise membrane MWCOs (2K, 3.5K, 7K, 10K and 20K) for dialyzing sample volumes up to 70 mL. The Slide-A-Lyzer G2 Dialysis Cassettes allow samples to be directly injected with a syringe or with a pipette through the unique sample port. The Slide-A-Lyzer Cassette design allows air to be removed from inside the cassette to maximize the surface area-to-volume ratio, providing better dialysis performance than dialysis tubing and tube-based systems.
