Cysteinyl leukotrienes play a role in the patho-physiology of numerous inflammatory disorders, including chronic asthmatic inflammation. The cysteine leukotriene receptors, CysLT1 and CysLT2, are thus primary targets for anti-asthma drugs. A number of cysteinyl leukotrienes (including LTC4, LTD4, and LTE4) have been identified, and are known to function as inflammatory mediators as well as being potent smooth muscle contractile agents.

Leukotriene receptor antagonists (LTRAs) reduce the effect of allergic inflammation and athsma by blocking cysteine leukotriene activity. Several CysLT antagonists, such as montelukast, zafirlucast, and pranlukast, are currently used in the treatment of asthma.

GeneBLAzer™ TACR2-NFAT-bla CHO-K1 Cells Thermo Scientific™

GeneBLAzer® TACR2-NFAT-bla CHO-K1 cells contain the human Tachykinin receptor 2 (TACR2), (Accession # NM_001057) stably integrated into the CellSensor® NFAT-bla CHO-K1 cell line. CellSensor® NFAT-bla CHO-K1 cells (Cat. no.K1078) contain a beta-lactamase reporter gene under control of the NFAT Response Element. TACR2-NFAT-bla CHO-K1 cells are functionally validated for Z’-factor and EC50 concentrations of (Nle10)-Neurokinin A (4-10) (Figure 1). In addition, TACR2-NFAT-bla CHO-K1 cells have been tested for assay performance under variable conditions.

CellSensor™ NFAT HEK 293T DA Assay Kit Thermo Scientific™

The CellSensor® NFAT HEK 293T DA (Division Arrested) cells contain a beta-lactamase reporter gene under the control of the Nuclear Factor of Activated T Cells (NFAT) response element stably integrated into HEK 293T cells. Fluorescence activated cell sorting (FACS) was used to isolate clones responsive to stimulation of the NFAT pathway by PMA (Phorbol 12-Myristate 13-Acetate) and Thapsagargin (Thaps). The cell line was validated for DMSO tolerance, cell number, stimulation time, substrate loading conditions, and EC50 concentration of Thaps at a fixed concentration of PMA. DA cells are irreversibly division arrested using a low-dose treatment of Mitomycin C, and have no apparent toxicity or change in cellular signal transduction.

This product can serve as a negative control in screening assays performed with specific G-protein coupled receptor (GPCR) DA cells that were made from this same background or these cells can be used in other assays where a sensitive readout to intracellular changes in calcium are needed.

GeneBLAzer™ P2RY2-NFAT-bla CHO-K1 Cells Thermo Scientific™

GeneBLAzer® P2RY2-NFAT-bla CHO-K1 cells contain the human Purinergic receptor P2, G protein-coupled, 2 (P2RY2) stably integrated into the CellSensor® NFAT-bla CHO-K1 cell line. CellSensor® NFAT-bla CHO-K1 cells (Cat. no. K1534) contain a beta-lactamase reporter gene under control of the Nuclear Factor of Activated T-cells (NFAT) response element.

P2RY2 expression has been identified in smooth muscle, skeletal muscle, heart, spleen, lymphocytes, macrophages, bone marrow, lung, intestine, placenta, brain, kidney, and liver. P2RY2 has been implicated in the induction of chloride secretion in airway epithelial tissue, increases in coronary blood flow by vasodilation, and epidermal homeostasis.

Tango™ GPR120S-bla U2OS Cells Thermo Scientific™

Tango™ GPR120 (short)-bla U2OS cells contain the human GPR120 (short) linked to a TEV protease site and a Gal4-VP16 transcription factor stably integrated into the Tango™ GPCR-bla U2OS parental cell line. The Quantifiler Trio DNA Quantification Kit is a robust kit that enables forensic laboratories to simultaneously obtain a quantitative and qualitative assessment of total human and human male DNA in a single, highly sensitive real-time PCR reaction. A Highly Informative Workflow Tool

Comparison to Quantifiler Duo Kits
There are several specific differences between the Quantifiler Trio DNA Quantification Kit and the Quantifiler Duo DNA Quantification Kit, including:
Target Loci: The Quantifiler Trio kit uses multiple-copy target loci for improved detection sensitivity. There are three human-specific target loci: Small Autosomal, Large Autosomal, and a Y-chromosome target. Each consists of multiple copies dispersed on various autosomal chromosomes (Small Autosomal and Large Autosomal) or multiple copies on the Y-chromosome.
Optimized Chemistry: The assay chemistry has been optimized for more efficient multiplexing, faster PCR cycle times (~1 hour), and better inhibitor tolerance. The overall performance improvements allow the Quantifiler Trio kit to better match the enhanced performance of newer STR kits that are designed to accommodate more challenging samples, like the Identifiler Plus, NGM SElect, or GlobalFiler PCR Amplification kits.
DNA Standard: The Quantifiler Trio DNA Standard is a pooled, human genomic, smaller molecular weight DNA for improved stability, homogeneity, and correlation with forensic samples. There is a five point standard curve protocol that helps improve efficiency with up to 6% more samples on a plate.

LanthaScreen™ STAT5 TF-1 Cell Line Thermo Scientific™

LanthaScreen® GFP Cellular Assays allow for the analysis of post-translational modifications for a number of target proteins in an live-cell context. In hematopoeitic cells, the JAK2⁄STAT5 signaling pathway plays an essential role in blood cell formation in response to cytokines such as GM-CSF, IL-3, and EPO. In this pathway, binding of these cytokines to their respective cell surface receptors results in the activation of JAK2, which in turn phospho-activates STAT5 proteins at specific tyrosine residues (Tyr-694⁄699). LanthaScreenTM-STAT5 TF-1 is a human hematopoeitic cell line which constitutively expresses GFP-STAT5 fusion proteins. The JAK2⁄STAT5 signaling pathway is known to be functionally intact in this cell line, therefore the GFP-STAT5 fusion protein serves as a substrate for the inducible phosphorylation by JAK2. Using this cell line, a homogenous immuno-assay has been developed in which the phosphorylation state of GFP-STAT5 is detected in cell lysates using a LanthaScreenTM Terbium-anti-mouse and anti-phospho STAT5 [pTyr694⁄699] antibody pair, in a time-resolved FRET (TR-FRET) readout. GFP-STAT5 Lentivirus was transduced into TF-1 cells followed by selection with Blasticidine. This cell line is a clonal population isolated by flow cytometry using GFP fluorescence as sorting marker, and has been screened for the constitutive expression of GFP-STAT5 fusion protein. Using a lytic TR-FRET immuno-assay, this cell line is validated for EC50 and Z' under optimized conditions using GM-CSF as an agonist for JAK2 -mediated GFP-STAT5 phosphorylation. This assay has also been tested for assay performance under variable experimental conditions, including cell plating density, stimulation time, DMSO tolerance and lysis⁄equilibration time.

In the LanthaScreen® Eu Kinase Binding Assay, binding of an Alexa Fluor® conjugate or 'tracer' such as Kinase Tracer 199 to a kinase is detected by addition of a Eu-labeled anti-tag antibody. Binding of the tracer & antibody to a kinase results in a high degree of FRET, whereas displacement of the tracer with a kinase inhibitor results in a loss of FRET.
