Thermo Scientific HisPur Ni-NTA Resin is a high-capacity, high-performance nickel-IMAC resin for routine affinity purification of His-tagged fusion proteins.

The expression and purification of recombinant proteins is central to protein regulation, structure and function studies. The majority of recombinant proteins are expressed as fusions with short affinity tags, the most popular being the polyhistidine (6xHis) tag. The method used to purify recombinant His-tagged proteins is immobilized metal affinity chromatography (IMAC), consisting of chelating resins charged with either nickel or cobalt ions that coordinate with the histidine side chains.

HisPur Ni-NTA Resin effectively purifies high levels of overexpressed His-tagged fusion proteins from bacterial lysates, such as those that are attained with Thermo Scientific B-PER Bacterial Protein Extraction Reagents. The resin performs well in batch-binding and spin-column procedures at a variety of scales. Performance equals or exceeds popular Ni-NTA resins from other suppliers. HisPur Ni-NTA Resin is a high-quality, stable and resilient affinity support. Tests confirm that no decrease in performance occurs after at least five repeated uses. These data indicated that the resin is highly resistant to structural degradation or nickel ion leaching during normal use.

HisPur Ni-NTA Resin and HisPur Cobalt Resin are alternative forms of IMAC, the former using nickel and the latter using cobalt as the chelated metal ion responsible for His-tag binding. Ni-NTA resins have been the most common IMAC resin choice for 6xHis-tag protein purifications because of the four metal-binding sites on the chelate, which enables high-protein binding and low-metal ion leaching. Cobalt binds less strongly and therefore more discriminantly, enabling greater purity but usually less yield in His-tagged protein purification.

Pierce™ 3x DYKDDDDK Peptide Thermo Scientific™

Pierce 3x DYKDDDDK Peptide is a lyophilized peptide with the sequence Met-Asp-Tyr-Lys-Asp-His-Asp-Gly-Asp-Tyr-Lys-Asp-His-Asp-Ile-Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys (MDYKDHDGDYKDHDIDYKDDDDK). It has a molecular weight of 2861.9 g/mol. For use, reconstitute to 1.5 mg/mL in PBS.

This tricosa-peptide can be used to competitively elute DYKDDDDK-tagged fusion proteins from immobilized anti-DYKDDDDK affinity supports, especially when a DYKDDDDK-tagged protein is sensitive to low pH. Note that Pierce 3x DYKDDDDK Peptide may interfere with protein determination assays and absorbance at 280 nm. However, the peptide can be removed from the eluate by desalting (e.g., with Zeba Spin Desalting Columns, Cat. No. 89891).

NHS-activated agarose is crosslinked, 6% beaded agarose resin that contains N-hydroxysuccinimide (NHS) functional groups. The activated resin reacts with primary amines to form stable amide linkages that covalently immobilize antibodies or other proteins for use in affinity purification procedures. Pierce NHS-Activated Agarose has a coupling capacity greater than 30 mg/mL for the slurry.

Applications:
• Rapid Immobilization of goat anti-mouse and anti-rabbit antibodies in order to purify IgG produced in animals or hybridomas.
• Bulk immobilization of Protein A for purification of monoclonal antibodies
• Immobilization of ligands for purification of recombinant proteins

Pierce NHS-Activated Agarose resin uses reliable NHS-ester Chemistry and does not require hazardous chemicals for immobilization. Other amine-reactive supports, such as periodate-oxidized resins, use toxic sodium cyanoborohydride to stabilize the reaction linkage to primary amines and take 4 to 6 hours to complete. Traditional methods such as cyanogen bromide-activated supports also couple amines; however, this chemistry results in nonspecific binding and constant slow leakage of the coupled ligand. Reactions with Pierce NHS-Activated Agarose are complete in less than one hour and yield much more stable linkages.

The NHS-Activated Agarose coupling reaction is performed in an amine-free buffer at pH 7-9. Protein coupling efficiency is typically greater than 80%, regardless of the ligand's molecular weight or pI. Once a ligand is immobilized, the prepared resin can be used for multiple affinity purification procedures. The crosslinked beaded agarose has fast linear flow potential, making it useful for gravity-flow and low- to medium-pressure applications.

The AKT signaling pathway serves as a master regulatory pathway for cell growth and proliferation. Aberrant activation of this pathway promotes survival of tumor cells in many types of cancer. This module is ideal for the multiplexed immuno-enrichment of key proteins in the AKT signaling pathway and has been optimized for use with the SureQuant quantitation modules.

The SureQuant AKT Pathway IP and MS Sample Preparation Module enables highly effective antigen immunoprecipitation (IP) and co-immunoprecipitation (co-IP) for MS analysis. Cells are first lysed and incubated with an IP-MS-verified biotinylated antibody mixture. The antibody-antigen complex is then captured on MS-compatible streptavidin magnetic beads, with non-specific proteins removed through washing with MS-compatible buffers. The target proteins are eluted using trypsin and are directly compatible with MS sample preparation. The MS sample preparation has been streamlined and can be completed in ~4 hours for same-day MS analysis.

This kit can be used with the either the SureQuant AKT Pathway Absolute Quantitation Module or the SureQuant AKT Pathway Relative Quantitation Module for protein targeted quantitation. When used together, these modules provide a complete solution for sample preparation and analysis of the target proteins in the pathway.

The glutathione is immobilized through its central sulfhydryl onto 6% crosslinked agarose resin. The expression and purification of recombinant proteins is central to protein regulation, structure and function studies. The majority of recombinant proteins are expressed as fusions with short affinity tags or small proteins, such as glutathione S-transferase (GST). This protein binds specifically to reduced glutathione (GSH) in near-neutral, nondenaturing conditions (e.g., Tris buffer). Bound protein is easily dissociated (eluted) by competitive displacement with buffer containing free, reduced GSH (oxidized glutathione, GSSH is not effective for this purpose). When proteins of interest are expressed as fusions with GST and glutathione is immobilized to an solid support, this protein-substrate system enables affinity purification of recombinant proteins, as well as various other experiments with those proteins.
