Pepsin: Cleaves proteins preferentially at carboxylic groups of aromatic amino acids such as phenylalanine, tryptophan and tyrosine. The optimum pH range for pepsin activity is 1.0–3.0.
Protease XIII: Cleaves proteins preferentially at carboxylic groups of basic amino acids (arginine, lysine, histidine). Like pepsin, Proteinase XIII is active at low pH. (1)

Thermo Scientific SMART Digest Immobilized Ribonucleases (Rnases) Columns are designed using the same proven technology as the Thermo Scientific SMART Digest Rnase kits to provide simple, fast and highly reproducible digestion of ribonucleic acid (RNA) in a flow through format.
Thermo Scientific SMART Digest Dual Enzyme columns containing pepsin and protease XIII provide fast, reliable digestion of proteins for for HDX-MS analysis helping to provide the sequence characterization of biotherapeutics.
Sample preparation is a key step in analytical workflow. Our solutions include a multitude of consumables and automated platforms to enable fast, reproducible and easy sample preparation whatever your workflow requirements may be.
Liquid chromatography-mass spectrometry (LC-MS) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) are key techniques in proteomic research studies. High-quality preparation of protein to peptide samples for LC-MS and LC-MS/MS is essential for high-quality results.
Can I use the AccelerOme TMT11plex MS Sample Preparation Kits and the AccelerOme TMTpro 16plex MS Sample Preparation Kits for samples with protein amounts lower than 10 µg?
When using the AccelerOme TMT11plex MS Sample Preparation Kits and the AccelerOme TMTpro 16plex MS Sample Preparation Kits, should I add a protease inhibitor cocktail to the lysis buffer? How about phosphatase inhibitors?
