Dynabeads™ Pan Mouse IgG (Invitrogen™)

Dynabeads® Pan Mouse IgG binds monoclonal mouse IgGs of your choice for specific cell isolation or depletion. It will bind all subclasses of mouse IgG and is Fc-specific. Low amount of IgG is required for binding. All cells from all species except from mouse can be isolated with this product, depending only on the specific antibody of choice.

• Depletion: cells can be depleted from a sample
• Positive isolation: cells can be positively isolated from a sample for molecular applications
• Negative isolation: cells can be negatively isolated from a PBMC sample by adding an antibody cocktail of mouse IgGs towards the unwanted cells. Compatible with flow cytometry.

Note: For cellular assays we recommend positive isolation followed by release of beads from the cells. Recommended product for bead-free cells: Dynal® CELLection™ Pan Mouse IgG (Cat. no 115-31D) in combination with your mouse IgG.

Excellent Yield, Purity, and Viability of Recovered Cells
Dynabeads® Mouse CD4 are uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of mouse CD4+ cells from spleen or lymph nodes, although any single-cell suspension may be used. Dynabeads® Mouse CD4 beads are coated with a primary monoclonal antibody specific for the CD4 membrane antigen mainly expressed on mouse T helper cells. Dynabeads® Mouse CD4 binds to CD4+ cells during a short incubation period in a tube, after which the bead-bound cells are separated from unbound cells using a magnet. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the isolated CD4+ cells.

Ideal for Both Depletion and Positive Isolation for Downstream Molecular Assays
Because of the powerful magnets used for separation, Dynabeads® Mouse CD4 can be used in viscous samples such as single cell suspensions from tissue, whole blood and bone marrow, and enable efficient depletion of mouse CD4+ cells in about 30 minutes. Dynabeads® Mouse CD4 will also afford excellent recovery of high-purity, viable cells when used for positive isolation for downstream studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. About Dynabeads® CD4

Magnetic bead–based separation offers easy handling
Dynabeads® CD4 is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet, it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. Starting samples can be whole blood, buffy coat, bone marrow, PBMC, or tissue digests.

Dynabeads™ Untouched™ Mouse CD8 Cells Kit (Invitrogen™)

Dynabeads® Untouched™ Mouse CD8 Cells supplies both 4.5 µm superparamagnetic Dynabeads® (with a polyclonal sheep anti-rat IgG antibody covalently bound to the bead surface) and a monoclonal Antibody Mix, together designed for the depletion of CD8-negative leucocytes from mouse spleen or lymph node single-cell suspensions. Following the depletion procedure, the negatively isolated, untouched target mouse CD8+ cells are pure, viable, and ready to be used in any downstream application.

Excellent yield, purity, and viability of recovered cells
The Antibody Mix contains a cocktail of rat IgGs that binds mouse CD4+ T cells, B cells, NK cells, monocytes/macrophages, dendritic cells, erythrocytes, and granulocytes. The Antibody Mix is first added to the sample in a tube to bind the unwanted CD8-negative cells. The cells are then washed to remove excess antibodies. Mouse Depletion Dynabeads® (coated with a rat IgG antibody) are added to bind the antibody-labeled, CD8-negative cells. After a short incubation, the bead-bound cells are then separated from unbound cells in 1–2 minutes using a magnet. The purified target mouse CD8+ cells in the supernatant can be transferred to a new tube for use in any downstream application.

This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the untouched CD8+ cells. The protocol can be scaled up according to your sample size.

The Dynabeads® Untouched™ Human CD8 T Cells Kit is used to isolate pure and viable untouched CD8+ T cells from PBMC by negative isolation. The kit depletes CD4+ T cells, B cells, NK cells, monocytes, platelets, dendritic cells, granulocytes, and erythrocytes. The negatively isolated human CD8+ T-cells are left in the sample and have not been in contact with the Dynabeads®. Use this kit to isolate pure and viable untouched B cells from mouse lymphoid organs by negative isolation. The kit depletes T cells, monocytes/macrophages, NK cells, plasma cells, CD5+ B1 cells, platelets, dendritic cells, granulocytes and erythrocytes. The negatively isolated mouse B cells are left in the sample and have not been in contact with the Dynabeads. Methodology:
