Dynabeads™ Regulatory CD4+/CD25+ T Cell Kit (Invitrogen™)

Use this kit to isolate highly pure CD4+ CD25+ regulatory T-cells (Treg) that express the intracellular transcription factor Foxp3. The CD4+ CD25- cell fraction can be used as effector cells in downstream inhibitory assays. You can expect >95% purity (CD4+ CD25+ expression) and >80% Foxp3 expression.

Methodology

The isolation procedure includes the following steps:

• Negative isolation of untouched human CD4+ T-cells from PBMC.
• Positive isolation of CD25+ T-cells from the untouched CD4+ T-cell subset.
• Removal of beads from the CD4+CD25+ Treg cells with DETACHaBEAD®.

Dynabeads™ FlowComp™ Mouse CD4+CD25+ Treg Cells Kit (Invitrogen™)

This gentle and easy-to-use kit allows you to isolate highly pure and functional mouse Treg cells from mouse lymph nodes or spleen samples. Your cells are not exposed to the stress of being passed through a column. The isolated CD4+CD25+Foxp3+ Treg cells are bead-free and flow-compatible. The cells can be readily expanded and retain their functional suppressive capacity. You can expect a purity level of >90% Treg cells. Over 88% of the isolated CD25+ cells express Foxp3. You will save time and money, and get increased confidence in your results.

Methodology:
Negative isolation of CD4+ T cells, followed by positive isolation of CD25+ regulatory T cells. First, an antibody mix is added to the sample and the Ab will bind the non-CD4+ T cells. Mouse Depletion Dynabeads® are then added and will bind to the Ab-cell complex during a short incubation. Bead-bound cells are separated on a magnet and discarded. The remaining untouched mouse CD4+ T-cells are then labeled with FlowComp™ CD25 Antibody and positively isolated using FlowComp™ Dynabeads®. A short incubation with FlowComp™ Release Buffer then releases the isolated mouse regulatory T-cells from the Dynabeads®. The isolation method is easy to use and to scale up.

Dynabeads™ Human DC Enrichment Kit (Invitrogen™)

Use this kit to enrich viable human dendritic cells (DC's) from PBMC by negative isolation. The kit depletes T cells, B cells, monocytes/ macrophages, granulocytes, NK cells and erythrocytes. Highly enriched DC's are bead and antibody-free, and can be used in any functional assay. The cells should be further sorted to obtain high purity of the specific DC sub-populations of choice.

• Obtain high recovery (80-100%) of viable DC's of all sup-populations.
• Substantial reduction in sorting time and volume compared to sorting only, thus saving money as well.

Methodology:
An antibody mix towards the non- dendritic cells is added to the sample and allowed to bind to the cells. Dynabeads® are added and will bind to the antibody-labeled cells during a short incubation. Dynabeads® CD15 are superparamagnetic beads covalently coupled with an anti-human CD15 antibody that efficiently depletes human CD15+ myeloid cells (predominantly neutrophil and eosinophil granulocytes) directly from whole blood, buffy coat, or PBMC samples. About Dynabeads® CD15
Dynabeads® CD15 are uniform, superparamagnetic polystyrene beads (4.5 µm diameter) coated with a monoclonal mouse IgM antibody recognizing the human CD15 membrane antigen. The CD15 antigen is predominantly expressed on human neutrophil and eosinophil granulocytes, and to a varying degree on monocytes.

Magnetic bead-;based separation offers easy handling
Dynabeads® CD15 is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet, it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. Starting samples can be whole blood, PBMC, or buffy coat.

Dynabeads™ FlowComp™ Human CD4 Kit (Invitrogen™)

This gentle and easy-to-use kit allows you to isolate flow-compatible human CD4+ T-cells from PBMC or directly from whole blood. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. In addition, your cells will not be exposed to the stress of being passed through a column.
You can expect a purity level of >95% and up to 75% recovery of human CD4+ cells.

The MagniSort™ Human CD117 (c-Kit) Positive Selection Kit is designed for the magnetic separation of CD117+ cells by positive selection. Dynabeads™ Pan Mouse IgG (Invitrogen™)

Dynabeads® Pan Mouse IgG binds monoclonal mouse IgGs of your choice for specific cell isolation or depletion. It will bind all subclasses of mouse IgG and is Fc-specific. Low amount of IgG is required for binding. All cells from all species except from mouse can be isolated with this product, depending only on the specific antibody of choice.

• Depletion: cells can be depleted from a sample
• Positive isolation: cells can be positively isolated from a sample for molecular applications
• Negative isolation: cells can be negatively isolated from a PBMC sample by adding an antibody cocktail of mouse IgGs towards the unwanted cells. Compatible with flow cytometry.

Simple method for T cell activation

The 4.5 µm diameter, inert, superparamagnetic beads are similar in size to antigen-presenting cells, and are covalently coupled to anti-CD3 and anti-CD28 antibodies. These two antibodies provide primary and co-stimulatory signals, optimized for efficient T cell activation and expansion. Expansion of the T cell population can be stimulated using recombinant IL-2, and after activation or expansion, the magnetic beads can be easily removed using a DynaMag™ magnet.

Dynabeads® CD2 bind to CD2+ cells during a short incubation period in a tube, after which the bead-bound cells are separated from unbound cells using a magnet. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the isolated CD2+ cells.

Ideal for Both Depletion and Positive Isolation for Downstream Molecular Assays
Because of the powerful magnets used for separation, Dynabeads® CD2 can be used in viscous samples such as whole blood and bone marrow, thereby providing a convenient method for the efficient depletion of CD2+ cells in about 30 minutes. Dynabeads® CD2 will also afford excellent recovery of high-purity, viable cells when used for positive isolation for downstream molecular studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. MagniSort™ Human CD4 T cell 2-Step Enrichment Kit (Invitrogen™)

The MagniSort™ Human CD4 2-Step Enrichment Kit is designed for the magnetic separation of CD4+ T cells by positive selection after first depleting monocytes. It has been optimized for the isolation of CD4+ T cells from human peripheral blood monocuclear cells, utilizing biotinylated Anti-Human CD14 and Anti-Human CD4 antibodies, as well as streptavidin-coated magnetic beads. In the first step, monocytes are bound by Anti-Human CD14 antibody and then magnetic beads. When placed in a magnetic field, the monocytes are held in the magnet while the remaining cells are decanted and used for the second step. In the second step, the monocyte-depleted cells are bound by Anti-Human CD4 antibody and then magnetic beads. Now, when placed in the magnetic field, the CD4+ T cells are held in the magnet while undesired cells can be decanted.

After enrichment, the purity of selected cells can be verified by staining with Anti-Human CD4, clones OKT4 or SK3.

Dynabeads™ Mouse DC (Dendritic Cell) Enrichment (Invitrogen™)

Use this kit to enrich viable mouse dendritic cells (DC's) from spleen/lymph node cells by negative isolation. The kit depletes T cells, mIgM+ B cells, NK cells, erythrocytes and most granulocytes. Highly enriched mouse DC's are bead and antibody-free, and can be used in any functional assay. The cells can be further sorted to obtain high purity of specific DC sub-populations.

Obtain high recovery (80-100%) of viable Lin-CD11c+ DC's of all sup-populations, including DC's positive for CD4, CD8α, B220 and CD19.

• Substantial reduction in sorting time and volume compared to sorting only, thus saving money in addition.
• Pool cells/tissue for easy sample scale -up

Methodology:
An antibody mix towards the non-dendritic cells is added to the sample and allowed to bind to the cells. Dynabeads® are added and will bind to the antibody-labeled cells during a short incubation. The bead-bound cells are quickly separated on a magnet and discarded. The remaining untouched mouse DC's can be directly sorted into the sup-population of choice or used in any other application.

The MagniSort™ Mouse CD335 (NKp46) Positive Selection Kit is designed for the magnetic separation of CD335+ cells by positive selection. It has been optimized for the isolation of CD335+ cells from mouse spleens or lymph nodes utilizing a biotinylated Anti-Mouse CD335+ antibody and streptavidin-coated magnetic beads. CD335+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD335+ cells by decanting.

After positive selection, the purity of selected cells can be verified by staining with Anti-Mouse CD49b (Integrin alpha 2), clone DX5, or Anti-Mouse NK1.1, clone PK136, in C57BL, FVB/N and NZB mice.
