Dynabeads™ Pan Mouse IgG (Invitrogen™)

Dynabeads® Pan Mouse IgG binds monoclonal mouse IgGs of your choice for specific cell isolation or depletion. It will bind all subclasses of mouse IgG and is Fc-specific. Low amount of IgG is required for binding. All cells from all species except from mouse can be isolated with this product, depending only on the specific antibody of choice.

• Depletion: cells can be depleted from a sample
• Positive isolation: cells can be positively isolated from a sample for molecular applications
• Negative isolation: cells can be negatively isolated from a PBMC sample by adding an antibody cocktail of mouse IgGs towards the unwanted cells. Compatible with flow cytometry.

Note: For cellular assays we recommend positive isolation followed by release of beads from the cells. Recommended product for bead-free cells: Dynal® CELLection™ Pan Mouse IgG (Cat. no 115-31D) in combination with your mouse IgG.

MagniSort™ Mouse CD45.2 Positive Selection Kit (Invitrogen™)

The MagniSort™ Mouse CD45.2 Positive Selection Kit is designed for the magnetic separation of CD45.2+ cells by positive selection. It has been optimized for the isolation of CD45.2+ cells from mouse spleens or lymph nodes utilizing a biotinylated Anti-Mouse CD45.2 antibody and streptavidin-coated magnetic beads. CD45.2+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD45.2+ cells by decanting.

After positive selection, the purity of selected cells can be verified by staining with Anti-Mouse CD45.2, clone 104.

Dynabeads™ Mouse CD8 (Lyt 2) (Invitrogen™)

Dynabeads® Mouse CD8 (Lyt 2) are magnetic beads that provide a robust and versatile tool to isolate or deplete mouse CD8+ T-cells directly from any sample, including tissue digests such as spleen or lymph nodes, whole blood, bone marrow, and mononuclear cells (MNC).

Excellent Yield, Purity, and Viability of Recovered Cells
Dynabeads® Mouse CD8 (Lyt 2) are uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of mouse CD8+ cells from spleen or lymph nodes, although any single-cell suspension may be used. Dynabeads® Mouse CD8 beads are coated with a primary monoclonal antibody specific for the Lyt 2 membrane antigen mainly expressed on cytotoxic/suppressor subpopulation of mature T-cells of all common mouse strains. Dynabeads® Mouse CD8 binds to CD8+ cells during a short incubation period in a tube, after which the bead-bound cells are separated from unbound cells using a magnet. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the isolated CD8+ cells.

Ideal for Both Depletion and Positive Isolation for Downstream Molecular Assays
Because of the powerful magnets used for separation, Dynabeads® Mouse CD8 (Lyt 2) can be used in viscous samples such as single cell suspensions from tissue, whole blood and bone marrow, and enable efficient depletion of mouse CD8+ cells in about 30 minutes. Dynabeads® Mouse CD8 (Lyt 2) will also afford excellent recovery of high-purity, viable cells when used for positive isolation for downstream studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. Note that intact cells will not be released from these beads, so if you want to isolate mouse CD8+ cells for cell-based applications, or you need to check your samples with flow cytometry, you should use Dynabeads® FlowComp Mouse CD8 Cells (for bead-free cells).

Dynabeads® Rat anti-Mouse IgM binds mouse IgM of your choice for specific cell isolation or depletion. All cells from all species except from mouse can be isolated with this product, depending only on the specific antibody of choice.

The Mouse Depletion Dynabeads® suspension contains uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of cells from any single cell suspension. This reagent is therefore especially well-suited for enriching tissue digests such as mouse spleen and lymph node cells. The Mouse Depletion Dynabeads® are coated with a secondary polyclonal antibody that binds rat IgGs. The Antibody Mix contains a cocktail of rat IgGs that bind mouse B cells, NK cells, monocytes/macrophages, dendritic cells, erythrocytes and granulocytes. The Antibody Mix is first added to the sample in a tube to bind the unwanted non-T cells. The cells are washed to remove excess antibodies, and Mouse Depletion Dynabeads® are then added. After a short incubation, the bead-bound cells are separated within 1 – 2 minutes using a magnet. The purified target mouse T-cells in the supernatant can be transferred to a new tube for use in any downstream application. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the untouched T cells. The protocol can easily be scaled up according to your sample size.

Dynabeads™ FlowComp™ Human CD8 Kit (Invitrogen™)

This gentle and easy-to-use kit allows you to isolate flow-compatible human CD8+ T-cells from PBMC or directly from whole blood. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. In addition, your cells will not be exposed to the stress of being passed through a column. You can expect a purity level of >95% human CD8+ cells.

It has been optimized for the isolation of CD45R+ cells from mouse spleens, bone marrow, or lymph nodes utilizing a biotinylated Anti-Mouse CD45R antibody and streptavidin-coated magnetic beads. CD45R+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD45R+ cells by decanting.

After positive selection, the purity of selected cells can be verified by staining with Anti-Mouse CD19, clone eBio1D3 (1D3).

Dynabeads™ Sheep Anti-Rat IgG (Invitrogen™)

Dynabeads® Sheep Anti-Rat IgG are 4.5 µm superparamagnetic beads with affinity purified polyclonal sheep anti-rat IgG covalently bound to the bead surface. These beads can be used to isolate any cell from any species (except rat), depending on the specificity of the monoclonal rat IgG employed. Dynabeads® Sheep Anti-Rat IgG are especially well suited for cell isolation from mouse samples (cross-reactivity to mouse antibodies is high and cross-reactivity to human antibodies is minimal).

Magnetic bead-based separation offers easy handling
The rat primary antibody may be added to the cell sample (indirect technique) or pre-coated onto the Dynabeads® Sheep Anti-Rat IgG (direct technique). Regardless of which of these techniques is used, when the cell sample and the Dynabeads® Sheep Anti-Rat IgG are mixed, the beads bind to the target cells. Placing the sample on a magnet separates the bead-bound cells from the rest of the sample. One or more rat primary antibodies may be used to either deplete the sample of unwanted cells (negative isolation) or to capture specific cells for molecular downstream experiments (positive isolation). For positive isolation of bead-free cells (e.g., for flow cytometry), we recommend using your rat antibody in combination with the Dynabeads® FlowComp™ Flexi Kit.

Magnetic bead–based separation offers easy handling
Dynabeads® CD3 is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet, it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. Starting samples can be whole blood, buffy coat, bone marrow, PBMC, or tissue digests.
