Dynabeads™ CD34 Positive Isolation Kit (Invitrogen™)

Use this kit to isolate a high purity and yield of human CD34+ progenitor stem cells. Positively isolated cells are bead and antibody-free, phenotypically unaltered and suitable for any downstream applications including flow cytometry, functional studies, and cell culture to produce dendritic cells (DC's). The Dynal® CD34 Progenitor Cell Isolation System can also be used to isolate CD34+ cells from rhesus monkey (Macaca mulatta).

MagniSort™ Human pan-Monocyte Enrichment Kit (Invitrogen™)

The MagniSort™ Human Pan-Monocyte Enrichment Kit is designed for the magnetic separation of conventional CD14+ and unconventional CD16+CD14- monocytes by negative selection. It has been optimized for the isolation of human pan-monocytes from normal human peripheral blood mononuclear cells utilizing a biotinylated antibody cocktail and streptavidin-coated magnetic beads. Undesired cells, including CD16+ NK cells, are bound by antibody and then magnetic beads that, when placed in a magnetic field, leaving monocytes untouched and free in solution.

MagniSort™ Mouse CD11b Positive Selection Kit (Invitrogen™)

The MagniSort™ Mouse CD11b Positive Selection Kit is designed for the magnetic separation of CD11b+ cells by positive selection. It has been optimized for the isolation of CD11b+ cells from mouse bone marrow cells or splenocytes utilizing a biotinylated Anti-Mouse CD11b antibody and streptavidin-coated magnetic beads. Please refer to the MagniSort™ Mouse CD11b Positive Selection Protocol for specific optimal conditions for isolating CD11b+ cells from either of these two tissues. CD11b+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD11b+ cells by decanting.

CELLection™ Epithelial Enrich Dynabeads™ (Invitrogen™)

Use this kit to obtain up to 5-log enrichment of human epithelial tumor cells, directly from whole blood, bone marrow or PBMC. Tumor cell clumps can be enriched from bone marrow. Enriched cells are bead-free, viable and suitable for immunocytochemical staining or any other downstream application.

Methodology: Add CELLection™ Dynabeads® to the sample. The beads will bind the tumor cells after a short incubation. The bead-bound tumor cells are quickly separated on a magnet and subsequently released from the beads with the supplied Release Buffer. (CELLection™ Dynabeads® contains a DNA linker that is cleaved by the DNase I in the Release Buffer.)

Note: If you want to deplete or use the isolated tumor cells for molecular applications, please use the equivalent product Dynabeads® Epithelial Enrich (Cat. No. 161-02). This product contains the same antibody, but with no release of the cells from the beads after positive isolation.

Dynabeads™ FlowComp™ Human CD8 Kit (Invitrogen™)

This gentle and easy-to-use kit allows you to isolate flow-compatible human CD8+ T-cells from PBMC or directly from whole blood. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. In addition, your cells will not be exposed to the stress of being passed through a column. You can expect a purity level of >95% human CD8+ cells.

Dynabeads® CD14 is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet, it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. Starting samples can be whole blood, buffy coat, bone marrow, PBMC, or tissue digests.

Dynabeads® Mouse pan B (B220) are uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of mouse B cells from spleen or lymph nodes, although any single cell suspension may be used. Dynabeads® Mouse pan B beads are coated with a primary monoclonal antibody specific for the B220 membrane antigen. B220 (also known as CD45R) is expressed on all B cells throughout development, from the early pro-B stage of B cell differentiation. Dynabeads® Mouse pan B binds to B cells during a short incubation period in a tube, after which the bead-bound cells are separated from unbound cells using a magnet. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the isolated B cells.

Ideal for Both Depletion and Positive Isolation for Downstream Molecular Assays
Because of the powerful magnets used for separation, Dynabeads® Mouse pan B (B220) can be used in viscous samples such as single cell suspensions from tissue, whole blood and bone marrow, and enable the efficient depletion of mouse B cells in about 30 minutes. Dynabeads® Mouse pan B will also afford excellent recovery of high-purity, viable cells when used for positive isolation for downstream studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. The kit also includes a DETACHaBEAD® reagent that releases the Dynabeads® from the purified CD4+ cells. Dynabeads™ M-450 Epoxy (Invitrogen™)

Dynabeads® M-450 Epoxy beads are ideal for custom cell-isolation protocols upstream of molecular applications. Purified antibodies can be used in conjunction with these beads to create a magnetic cell separation media with the good antibody orientation.

Ideal for Cell Separation when you have a custom antibody:
Low background and covalent linking of antibodies to the bead surface make these beads an excellent choice for cell separation applications when a downstream release from the beads is not required.

Ultra rapid protocols:

Quickly isolate targeted cell populations upstream of molecular analyses

• Excellent for cell isolation procedures that require custom antibodies

Not recommended for Immunoprecipitation (IP):

Coupling procedure outline:
Any purified antibody can be covalently coupled to the bead surface in a easy-to-perform overnight reaction. Simply incubate the antibody you wish to use for your cell isolation procedure with these 4.5 µm Dynabeads®. Optimal coupling occurs at high pH (8.5-9.5) and at 37°C. For pH labile antibodies, coupling can be performed in an alternative buffer at pH 7.4.

About Dynabeads®:
Dynabeads® are non-porous monodisperse superparamagnetic beads. They are highly mobile in solution enabling antibodies coupled to the beads to continuously interact with the cell suspension. These 4.5 µm superparamagnetic beads experience a strong pull when placed in a magnetic field and gently pull targeted cells to the tube walls when the tube is transferred to a rack containing a strong magnetic field. With the beads and attached cells concentrated at the tube wall the supernatant liquid containing non-targeted cells is easily and quickly decanted or pipetted away. Washing steps are performed similarly.
