Dynabeads™ FlowComp™ Mouse CD8 Kit (Invitrogen™)

This gentle and easy-to-use kit allows you to isolate flow-compatible mouse CD8+ T cells. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. In addition, your cells will not be exposed to the stress of being passed through a column.

You can expect a purity level of >85% and viability of >85% of mouse CD8+ cells.

About Dynabeads® CD19 Pan B
Dynabeads® CD19 pan B are uniform (4.5 µm diameter) superparamagnetic beads coated with a primary monoclonal antibody specific for the CD19 membrane antigen mainly expressed on human B cells. Because of the bead size, Dynabeads® CD19 pan B can easily and efficiently isolate or deplete cells from viscous samples such as whole blood and bone marrow in about 30 minutes. Dynabeads® CD19 pan B will also afford excellent recovery of high-purity, viable cells for use in downstream molecular studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. Note that intact cells will not be released from these beads, so if you want to isolate CD19+ cells for cell-based applications, or you need to check your samples with flow cytometry, you can combine with DETACHaBEAD® CD19 (purchased separately to get bead- and antibody-free cells) or use Dynabeads® Untouched™ Human B Cells to obtain untouched human B cells.

Magnetic bead-based separation offers easy handling
Dynabeads® CD19 Pan B is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet, it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. Starting samples can be whole blood, buffy coat, bone marrow, PBMC, or tissue digests.

About Dynabeads® Human T-Activator CD3/CD28
Dynabeads® Human T-Activator CD3/CD28 offer a simple method for activation and expansion of T cells that does not require feeder cells (antigen-presenting cells) or antigen. The uniform 4.5 µm diameter, inert, superparamagnetic beads are similar in size to antigen-presenting cells, and are covalently coupled to anti-CD3 and anti-CD28 antibodies. These two antibodies provide primary and co-stimulatory signals, optimized for efficient T cell activation and expansion. Expansion of the T cell population can be stimulated using recombinant IL-2, and after activation or expansion, the magnetic beads can be easily removed using a DynaMag™ magnet. For expansion of antigen-specific T cells from T cell clones or T cell lines, we recommend using Dynabeads® Human T-Activator CD3/CD28/CD137.

Starting samples
Start with mononuclear cells (MNCs)/peripheral blood mononuclear cells (PBMCs) from either whole blood or buffy coat, or T cell subsets including CD3+ T cells, CD4+ or CD8+ T cells, or regulatory T cells.

Downstream applications
The activated T cells can be analyzed shortly after activation (for transfection/transduction or to study TCR signaling, proteomics, gene expression, etc.). T cells can be left in culture to differentiate into T helper cell subsets or expansion of polyclonal/antigen-specific T cells.

CTS™ Dynabeads™ Treg Xpander (Gibco™)

Optimized for the ex vivo activation and expansion of regulatory T cells (Tregs), CTS Dynabeads Treg Xpander provides more specific and higher fold expansion compared to our CTS Dynabeads CD3/CD28. Tregs activated and expanded with this product show suppressive activity and display the characteristic Treg cytokine profile and phenotype. Furthermore, high stability of the expanded Tregs has been demonstrated through analysis of the Treg-specific demethylated region.

These beads can be used to isolate any cell from any species (except rat), depending on the specificity of the monoclonal rat IgG employed. Dynabeads® Sheep Anti-Rat IgG are especially well suited for cell isolation from mouse samples (cross-reactivity to mouse antibodies is high and cross-reactivity to human antibodies is minimal).

Magnetic bead-based separation offers easy handling
The rat primary antibody may be added to the cell sample (indirect technique) or pre-coated onto the Dynabeads® Sheep Anti-Rat IgG (direct technique). Regardless of which of these techniques is used, when the cell sample and the Dynabeads® Sheep Anti-Rat IgG are mixed, the beads bind to the target cells. Placing the sample on a magnet separates the bead-bound cells from the rest of the sample. One or more rat primary antibodies may be used to either deplete the sample of unwanted cells (negative isolation) or to capture specific cells for molecular downstream experiments (positive isolation). For positive isolation of bead-free cells (e.g., for flow cytometry), we recommend using your rat antibody in combination with the Dynabeads® FlowComp™ Flexi Kit.

Dynabeads™ M-450 Epoxy (Invitrogen™)

Dynabeads® M-450 Epoxy beads are ideal for custom cell-isolation protocols upstream of molecular applications. Purified antibodies can be used in conjunction with these beads to create a magnetic cell separation media with the good antibody orientation.

Ideal for Cell Separation when you have a custom antibody:
Low background and covalent linking of antibodies to the bead surface make these beads an excellent choice for cell separation applications when a downstream release from the beads is not required.

Quickly isolate targeted cell populations upstream of molecular analyses

Not recommended for Immunoprecipitation (IP):

Coupling procedure outline:
Any purified antibody can be covalently coupled to the bead surface in a easy-to-perform overnight reaction. Simply incubate the antibody you wish to use for your cell isolation procedure with these 4.5 µm Dynabeads®. Optimal coupling occurs at high pH (8.5-9.5) and at 37°C. For pH labile antibodies, coupling can be performed in an alternative buffer at pH 7.4.

About Dynabeads®:
Dynabeads® are non-porous monodisperse superparamagnetic beads. They are highly mobile in solution enabling antibodies coupled to the beads to continuously interact with the cell suspension. These 4.5 µm superparamagnetic beads experience a strong pull when placed in a magnetic field and gently pull targeted cells to the tube walls when the tube is transferred to a rack containing a strong magnetic field. With the beads and attached cells concentrated at the tube wall the supernatant liquid containing non-targeted cells is easily and quickly decanted or pipetted away. Washing steps are performed similarly.

CTS™ Dynabeads™ CD3/CD28 are intended for ex vivo isolation, activation, and expansion of human T-cells in translational research. The technology—previously known as Xcyte™ Dynabeads™ or Dynabeads™ ClinExVivo™ CD3/CD28—has been used in a number of clinical studies. By combining anti-CD3 and anti-CD28 antibodies on Dynabeads, the beads provide both the primary and co-stimulatory signals that are required for activation and expansion of T cells. CTS Dynabeads CD3/CD28 are manufactured and controlled according to ISO 9001:2000 and ISO 13485:2012 requirements. A Device Master File is held with the FDA for cross-referencing in IND applications, and a CoA is available on request. In the US, CTS Dynabeads CD3/CD28 are available for clinical use only under an approved Investigational New Drug (IND) application. Dynabeads™ Human T-Expander CD3/CD28 (Cat. No. 11141D) is the research grade version of CTS Dynabeads CD3/CD28, containing the same amount and ratio of antibodies from the same clones as CTS Dynabeads CD3/CD28, and intended for small scale pre-clinical research.

Applications:

Dynabeads™ Mouse DC (Dendritic Cell) Enrichment (Invitrogen™)

Use this kit to enrich viable mouse dendritic cells (DC's) from spleen/lymph node cells by negative isolation. The kit depletes T cells, mIgM+ B cells, NK cells, erythrocytes and most granulocytes. Highly enriched mouse DC's are bead and antibody-free, and can be used in any functional assay. The cells can be further sorted to obtain high purity of specific DC sub-populations.

Obtain high recovery (80-100%) of viable Lin-CD11c+ DC's of all sup-populations, including DC's positive for CD4, CD8α, B220 and CD19.

Methodology:
An antibody mix towards the non-dendritic cells is added to the sample and allowed to bind to the cells. Dynabeads® are added and will bind to the antibody-labeled cells during a short incubation. The bead-bound cells are quickly separated on a magnet and discarded. The remaining untouched mouse DC's can be directly sorted into the sup-population of choice or used in any other application.

MagniSort™ Mouse CD45 Positive Selection Kit (Invitrogen™)

The MagniSort™ Mouse CD45 Positive Selection Kit is designed for the magnetic separation of CD45+ cells by positive selection. It has been optimized for the isolation of CD45+ cells from mouse spleens, bone marrow or digested lung tissue utilizing a biotinylated Anti-Mouse CD45 antibody and streptavidin-coated magnetic beads. CD45+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD45+ cells by decanting.

After positive selection, the purity of selected cells can be verified by staining with Anti-Mouse CD45, clone 30-F11.

Use this kit with any biotinylated antibody for the positive isolation of a cell population including mouse cells. Biotinylated antibodies are attached to CELLection™ Dynabeads® via a DNA linker; this linker provides a cleavable site to remove the beads from the cells after isolation. The MagniSort™ Mouse CD4 Positive Selection Kit is designed for the magnetic separation of CD4+ cells by positive selection. It has been optimized for the isolation of CD4+ cells from mouse spleens or lymph nodes utilizing a biotinylated Anti-Mouse CD4 antibody and streptavidin-coated magnetic beads. CD4+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD4+ cells by decanting.

After positive selection, the purity of selected cells can be verified by staining with Anti-Mouse CD4, clones RM4-5 or GK1.5.

Dynabeads™ Sheep-Anti Mouse IgG (Invitrogen™)

Dynabeads® Sheep anti-Mouse IgG binds monoclonal mouse IgG of your choice for specific cell isolation or depletion. All cells from all species except from mouse can be isolated with this product, depending only on the specific antibody of choice.

• Depletion: cells can be depleted from a sample
• Positive isolation: cells can be positively isolated from a sample for molecular applications
• Negative isolation: cells can be negatively isolated from a PBMC sample by adding an antibody cocktail of mouse IgGs towards the unwanted cells. Compatible with flow cytometry.

Note: For a more universal binding of your mouse IgG of any subclass, please use Dynabeads® Pan Mouse Ig (Cat.no. 110-41D or 110-42D).

CD45+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the CD45- cells can be separated from CD45+ cells by decanting.

After depletion, the purity of selected cells can be verified by staining with Anti-Human CD45, clone 2D1.

Dynabeads™ Untouched™ Human B Cells Kit (Invitrogen™)

Use this kit to isolate pure and viable untouched B cells from PBMC by negative isolation. The kit depletes T cells, NK cells, monocytes, platelets, dendritic cells, granulocytes and erythrocytes. The negatively isolated human B cells are left in the sample and have not been in contact with the Dynabeads. The bead and antibody-free B cells are truly untouched, and in perfect shape for any functional assay and application.

Dynabeads® Mouse pan T (Thy 1.2) are uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of mouse T-cells from spleen or lymph nodes, although any single-cell suspension may be used. Dynabeads® Mouse pan T beads are coated with a primary monoclonal antibody specific for the Thy 1.2 membrane antigen expressed on peripheral T cells, thymocytes and intraepithelial T lymphocytes of all common mouse strains. Dynabeads® Mouse pan T binds to T-cells during a short incubation period in a tube, after which the bead-bound cells are separated from unbound cells using a magnet. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the isolated T cells.

Ideal for Both Depletion and Positive Isolation for Downstream Molecular Assays
Because of the powerful magnets used for separation, Dynabeads® Mouse pan T (Thy 1.2) can be used in viscous samples such as single cell suspensions from tissue, whole blood and bone marrow, and enable efficient depletion of mouse T-cells in about 30 minutes. About Dynabeads® CD4 Positive Isolation Kit
