Use this kit to isolate a high purity and yield of human CD34+ progenitor stem cells. Dynabeads® Human T-Expander CD3/CD28 is intended for separation, activation and expansion of human T cells. Dynabeads® Human T-Expander CD3/CD28 is the research grade version of Dynabeads® CD3/CD28 CTS™. Containing the same amount and ratio of antibodies from the same clones as Dynabeads® CD3/CD28 CTS™, Dynabeads® Human T-Expander CD3/CD28 are intended for small scale pre-clinical research. Dynabeads® Human T-Expander CD3/CD28 offers a simple method for activation and expansion of T-cells that does not require antigen presenting cells or antigen. Applications:

Dynabeads™ FlowComp™ Mouse CD4 Kit (Invitrogen™)

This gentle and easy-to-use kit allows you to isolate flow-compatible mouse CD4+ T cells. Immediately after positive isolation, the beads are released and removed from your cell sample. In this way, you avoid exposing your cells to potentially cytotoxic or immunogenic foreign substances like iron oxides or dextrans, a potential problem when working with biodegradable particles. In addition, your cells will not be exposed to the stress of being passed through a column.

You can expect a purity level of >85% and viability of >85% of mouse CD4+ cells.

CELLection™ Epithelial Enrich Dynabeads™ (Invitrogen™)

Use this kit to obtain up to 5-log enrichment of human epithelial tumor cells, directly from whole blood, bone marrow or PBMC. Tumor cell clumps can be enriched from bone marrow. Enriched cells are bead-free, viable and suitable for immunocytochemical staining or any other downstream application.

Methodology: Add CELLection™ Dynabeads® to the sample. The beads will bind the tumor cells after a short incubation. The bead-bound tumor cells are quickly separated on a magnet and subsequently released from the beads with the supplied Release Buffer. (CELLection™ Dynabeads® contains a DNA linker that is cleaved by the DNase I in the Release Buffer.)

Note: If you want to deplete or use the isolated tumor cells for molecular applications, please use the equivalent product Dynabeads® Epithelial Enrich (Cat. No. 161-02). This product contains the same antibody, but with no release of the cells from the beads after positive isolation.

Excellent Yield, Purity, and Viability of Recovered Cells
Dynabeads® Mouse pan B (B220) are uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of mouse B cells from spleen or lymph nodes, although any single cell suspension may be used. Dynabeads® Mouse pan B beads are coated with a primary monoclonal antibody specific for the B220 membrane antigen. B220 (also known as CD45R) is expressed on all B cells throughout development, from the early pro-B stage of B cell differentiation. Dynabeads® Mouse pan B binds to B cells during a short incubation period in a tube, after which the bead-bound cells are separated from unbound cells using a magnet. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the isolated B cells.

Ideal for Both Depletion and Positive Isolation for Downstream Molecular Assays
Because of the powerful magnets used for separation, Dynabeads® Mouse pan B (B220) can be used in viscous samples such as single cell suspensions from tissue, whole blood and bone marrow, and enable the efficient depletion of mouse B cells in about 30 minutes. Dynabeads® Mouse pan B will also afford excellent recovery of high-purity, viable cells when used for positive isolation for downstream studies; for example, those in which cells are to be lysed while still attached to beads, and nucleic acids or proteins further purified. Note that viable cells will not be released from these beads, so if you want to isolate mouse B cells for cell-based applications, or you need to check your samples with flow cytometry, you should use Dynabeads® Mouse CD43 (Untouched™ B cells).

The Mouse Depletion Dynabeads® suspension contains uniform, superparamagnetic beads (4.5 µm diameter) that enable easy isolation of cells from any single cell suspension. This reagent is therefore especially well-suited for enriching tissue digests such as mouse spleen and lymph node cells. The Mouse Depletion Dynabeads® are coated with a secondary polyclonal antibody that binds rat IgGs. The Antibody Mix contains a cocktail of rat IgGs that binds mouse CD8+ T cells, B cells, NK cells, monocytes/macrophages, dendritic cells, erythrocytes and granulocytes. The Antibody Mix is first added to the sample in a tube to bind the unwanted non-CD4+ cells. The cells are then washed to remove excess antibodies, and Mouse Depletion Dynabeads® are added. After a short incubation, the bead-bound cells are then separated from unbound cells in 1 – 2 minutes using a magnet. The purified target mouse CD4+ cells in the supernatant can be transferred to a new tube for use in any downstream application. This fast and gentle isolation method does not require the use of columns, and helps ensure high purity, recovery and viability of the untouched CD4+ cells. The protocol can easily be scaled up according to your sample size.

Magnetic bead–based separation offers easy handling
Dynabeads® CD8 is added to the sample under continuous mixing to optimize the binding of the Dynabeads® to the target cells. By placing the sample on a magnet, it separates the bead-bound target cells from the rest of the sample in just 1–2 minutes. For depletion, remove the supernatant to a new tube for further studies and discard the bead-bound cells. For positive isolation for molecular studies, remove the supernatant and wash the bead-bound cells 2–3 times in buffer to get optimal purity. The cells can be lysed while still attached to the beads and the supernatant transferred to a new tube for downstream molecular analysis. Starting samples can be whole blood, buffy coat, bone marrow, PBMC, or tissue digests.

How Dynabeads® Mouse T-Activator CD3/CD28 works:
Dynabeads® Mouse T-Activator CD3/CD28 offers a simple method for activation and expansion of T-cells that does not require feeder cells (antigen-presenting cells) or antigen. The 4.5 µm diameter, inert, superparamagnetic beads are similar in size to antigen-presenting cells, and are covalently coupled to anti-CD3 and anti-CD28 antibodies. These two antibodies provide primary and co-stimulatory signals, optimized for efficient T-cell activation and expansion. Expansion of the T-cell population can be stimulated using recombinant IL-2, and after activation or expansion, the magnetic beads can be easily removed using a DynaMag™ magnet.

For expansion of antigen specific T-cells from T-cell clones or T-cell lines, we recommend using Dynabeads® Mouse T-Activator CD3/CD28/CD137.

Starting samples
Start with single cell suspensions from spleen or lymph nodes, mononuclear cells (MNCs), whole blood, or T-cell subsets including CD3+ T cells, CD4+ or CD8+ T cells, or regulatory T cells. Get an overview of the different Dynabeads® available for cell isolation.

Downstream applications
The activated T-cells can be analyzed shortly after activation (for transfection/transduction or to study TCR signaling, proteomics, gene expression, etc.). T-cells can be left in culture to differentiate into T helper cell subsets or expansion of polyclonal/antigen-specific T cells.

CELLection™ Pan Mouse IgG Kit (Invitrogen™)

Use this kit with any mouse IgG for the positive isolation of a cell population from all species except mouse. Antibodies are attached to the surface of the CELLection™ Dynabeads® via a DNA linker; this linker provides a cleavable site to release and remove the beads from the cells after isolation. Bead-free cells are viable and can be used in any downstream application.

The MagniSort™ Mouse CD90.2 Positive Selection Kit is designed for the magnetic separation of CD90.2+ cells by positive selection. It has been optimized for the isolation of CD90.2+ cells from mouse spleens or lymph nodes utilizing a biotinylated Anti-Mouse CD90.2 antibody and streptavidin-coated magnetic beads. CD90.2+ cells are bound by antibody and then magnetic beads. When placed in a magnetic field, the undesired cells can be separated from CD90.2+ cells by decanting.

After positive selection, the purity of selected cells can be verified by staining with Anti-Mouse CD90.2, clone 53-2.1.

Dynabeads™ Human Treg Expander (Gibco™)

Dynabeads® Human Treg Expander is intended for activation and expansion of human regulatory T-cells (Treg cells) after isolation with the Dynabeads® Regulatory CD4+CD25+ T Cell Kit (Cat. No. 11363D).

Dynabeads® Human Treg Expander offers a simple method for activation and expansion of Treg cells that does not require antigen presenting cells or antigen. Dynabeads® Human Treg Expander are uniform 4.5 µm, superparamagnetic beads coupled with an optimized mixture of anti-CD3 and anti-CD28 monoclonal antibodies. By combining anti-CD3 and anti-CD28 antibodies on Dynabeads®, the beads will provide both the primary and co-stimulatory signals that are required for activation and expansion of the Treg cells.

Dynabeads™ Regulatory CD4+/CD25+ T Cell Kit (Invitrogen™)

Use this kit to isolate highly pure CD4+ CD25+ regulatory T-cells (Treg) that express the intracellular transcription factor Foxp3. The CD4+ CD25- cell fraction can be used as effector cells in downstream inhibitory assays. You can expect >95% purity (CD4+ CD25+ expression) and >80% Foxp3 expression.

Isolate bead- and antibody-free human CD4+ CD25+ Foxp3+ Treg cells

Treg cells can be expanded with Dynabeads® CD3/CD28 Treg Expander and remain
Foxp3+.

Treg cells can be used in any downstream applications (e.g., suppression assays)

Methodology

The isolation procedure includes the following steps:

• Negative isolation of untouched human CD4+ T-cells from PBMC.
• Positive isolation of CD25+ T-cells from the untouched CD4+ T-cell subset.
• Removal of beads from the CD4+CD25+ Treg cells with DETACHaBEAD®.

MagniSort™ Human CD4 T cell 2-Step Enrichment Kit (Invitrogen™)

The MagniSort™ Human CD4 2-Step Enrichment Kit is designed for the magnetic separation of CD4+ T cells by positive selection after first depleting monocytes. It has been optimized for the isolation of CD4+ T cells from human peripheral blood monocuclear cells, utilizing biotinylated Anti-Human CD14 and Anti-Human CD4 antibodies, as well as streptavidin-coated magnetic beads. In the first step, monocytes are bound by Anti-Human CD14 antibody and then magnetic beads. When placed in a magnetic field, the monocytes are held in the magnet while the remaining cells are decanted and used for the second step. In the second step, the monocyte-depleted cells are bound by Anti-Human CD4 antibody and then magnetic beads. Now, when placed in the magnetic field, the CD4+ T cells are held in the magnet while undesired cells can be decanted.

After enrichment, the purity of selected cells can be verified by staining with Anti-Human CD4, clones OKT4 or SK3.
