Millennium™ RNA Markers (Invitrogen™)

The Ambion® RNA Millennium™ Markers are designed to provide accurate size determination of single-stranded RNA transcripts from 0.5 to 9 kb and can be used in any northern protocol. Sufficient markers are provided for 25 northern gel lanes. They are a mixture of 10 discrete RNA transcripts with easy-to-remember sizes: 0.5, 1, 1.5, 2, 2.5, 3, 4, 5, 6, and 9 kilobases. The markers may be stained with ethidium bromide either during or after electrophoresis. Because of their quantity and even spacing, these markers are ideal for constructing very accurate standard curves for mRNA size determination. They also provide a reference scale for easy verification and comparison of known bands. 1–2 µg (1–2 µg) of RNA Millennium™ Markers will generate 10 distinct bands on a 1% denaturing agarose gel with ethidium bromide staining. All marker lots undergo stringent nuclease testing and are shown to be stable overnight at 37°C.

phiX174 RF1 DNA (Thermo Scientific™)

Thermo Scientific phiX174 was the first DNA virus discovered to have a single-stranded, circular genome. phiX174 DNA is 5386 nt in length. The DNA strand packaged into the virion is termed the “plus” strand. After entering the cell, the Thermo Scientific phiX174 DNA is used as a template for minus-strand synthesis, producing double-stranded DNA. The conversion of plus DNA strands to double-stranded DNA does not require any of the phage genes to function.

The double-stranded DNA can then be transcribed, resulting in synthesis of phage-encoded proteins. Synthesis of single-stranded (plus) DNA requires phage-encoded gene A protein. DNA synthesis is initiated at ori (+) and proceeds in the direction indicated. Late in infection, the single-stranded circles are encapsidated into new virions. The cycle terminates by cellular lysis, mediated by phage gene E encoded protein.

RNA 6000 Ladder (Invitrogen™)

The Ambion® RNA 6000 Ladder is a set of six RNA transcripts with lengths of 0.2, 0.5, 1.0, 2.0, 4.0, and 6.0 kb. The 0.2 kb band is at a concentration of 20 ng/ µL (A260). The RNA 6000 Ladder is designed for use with Agilent's lab-on-a-chip. Three tubes containing 20 µL each are provided. 150 ng (1 µL) will generate 6 distinct bands when run on a RNA Labchip™ using the Total RNA or mRNA assay.

NoLimits 3000 bp DNA Fragment (Thermo Scientific™)

Thermo Scientific NoLimits is a collection of 36 highly-purified individual DNA fragments ranging from 10 to 20,000 bp. Unlike other vendors, where custom standards are produced by PCR, Thermo Scientific NoLimits DNA fragments are produced using specifically designed plasmids purified by patented technology.Plasmid DNA, digested with Thermo Scientific Restriction Enzymes, is chromatography-purified to prepare the individual DNA fragments from the digestion mixture. The results are extremely pure DNA fragments, free of any truncated or degraded molecules. These custom DNA fragments are the best custom standards for applications ranging from electrophoresis (both gel and capillary) to HPLC and beyond.

GeneScan™ 1000 ROX™ dye Size Standard (Applied Biosystems™)

GeneScan™ 1000 ROX™ Size Standard is an internal lane size standard developed for use with Applied Biosystems® fluorescence-based DNA electrophoresis systems. The use of an internal lane size standard during electrophoresis enables automated data analysis and precise DNA fragment size comparisons between electrophoresis runs. GeneScan™ 1000 ROX™ Size Standard is designed for sizing DNA fragments in the 47–946 bp range and provides labeled fragments of 47, 51, 55, 82, 85 93 99, 126, 136, 262, 293, 317, 439, 557, 692, 695 and 946 bases. Each of the DNA fragments is labeled with ROX™ fluorophore, which results in a single peak when run under non-denaturing conditions.

Note: Under denaturing conditions using POP-4® polymer, fragment lengths between 100 and 539 bp can be determined (the linear range of the separation with respect to length vs. scan number).

1 Kb Plus DNA Ladder (Invitrogen™)

Invitrogen 1 Kb Plus DNA Ladder is designed for sizing and approximate quantification of double-stranded DNA in the range of 100 bp to 15,000 bp. 1 Kb Plus DNA Ladder consists of 18 individual chromatography-purified DNA fragments and has a reference band at 1,500 bp for easy orientation.

1 Kb DNA Ladder is ideal for separation on 0.8–1% agarose gels.

Lambda DNA/EcoRI plus HindIII Marker (Thermo Scientific™)

Thermo Scientific Lambda DNA/EcoRI+HindIII Marker is recommended for sizing of linear double-stranded large DNA fragments in agarose gels. Lambda DNA is digested to completion with the appropriate Thermo Scientific restriction enzyme(s) and purified and dissolved in storage buffer. DNA fragments contain various sticky ends depending on the restriction enzyme used for preparation of the marker.

Lambda DNA/EcoRI+HindIII Marker can be labeled radioactively with T4 Polynucleotide Kinase. Alternatively, and it can be labeled radioactively or non-radioactively with Klenow fragment or Klenow Fragment, exo- with the fill-in reaction (see Resources for Protocols).

Note
The cohesive ends (the 12nt cos site of lambda DNA) of fragments indicated may anneal and form additional bands. These fragments can be separated by heating at 65°C for 5minutes and then cooling on ice for 3minutes.

The shortest fragments (oblique) are not visible in standard electrophoresis. Fragment lengths are predicted by computer analysis of respective DNA sequences.

GeneScan™ 2500 TAMRA™ dye Size Standard (Applied Biosystems™)

The GeneScan™ 2500 TAMRA™ Size Standard is an internal lane size standard developed for use with the Applied Biosystems fluorescencebased DNA electrophoresis systems. The use of an internal lane size standard during electrophoresis enables automated data analysis and precise DNA fragment size comparisons between electrophoresis runs. GeneScan™ 2500 TAMRA™ Size provides labeled fragments of 37, 94, 109, 116, 172, 186, 222, 233, 238, 269, 286, 361, 470, 490, and 536 bases as well as larger fragments. Each of the DNA fragments is labeled with the TAMRA™ fluorophore, which results in a single peak when run under denaturing or native conditions.
