RNA Century™-Plus Marker Templates (Invitrogen™)

Ambion® Century™-Plus Marker Templates contain mixtures of linearized plasmids ready for use as templates in in vitro transcription reactions for synthesis of labeled RNA molecular size standards. One tube containing 5 μg (0.5 mg⁄mL) is provided. When transcribed with T7 RNA polymerase, the templates produce 7 RNA transcripts with lengths of 100, 200, 300, 400, 500, 750, and 1,000 bases. They can be labeled with radioisotopes, biotin, or other nonisotopic modified nucleotides. They are ideal as size standards for ribonuclease protection assays and other applications where a low molecular weight RNA ladder is useful.

GeneRuler High Range DNA Ladder (Thermo Scientific™)

Thermo Scientific GeneRuler High Range DNA Ladder is recommended for fast sizing and approximate quantification of high molecular weight double-stranded DNA on agarose gels (in 1.5 hour, 0.4% agarose gel). The ladder is a mixture of chromatography-purified individual DNA fragments.

GeneRuler High Range DNA Ladder can be labeled radioactively with T4 Polynucleotide Kinase (see Protocol in the Resources.)

The ladder is supplied with 6X Orange DNA Loading Dye for sample DNA.

Millennium™ RNA Markers-Formamide (Invitrogen™)

The Ambion® Millennium™ Markers-Formamide are designed to provide accurate size determination of single-stranded RNA transcripts from 0.5 to 9 kb and can be used in any Northern protocol. Sufficient markers are provided for 25 Northern gel lanes. They are a mixture of 10 discrete RNA transcripts with easy-to-remember sizes: 0.5, 1, 1.5, 2, 2.5, 3, 4, 5, 6, and 9 kilobases. The markers are supplied in formamide and show increased stability and resistance to RNase compared to other large molecular weight RNA markers. They may be stained with ethidium bromide either during or after electrophoresis. Because of their quantity and even spacing, these markers are ideal for constructing very accurate standard curves for mRNA size determination. They also provide a reference scale for easy verification and comparison of known bands. 1-2 µg (1-2 µl) of RNA Millennium™ Markers will generate 10 distinct bands on a 1% denaturing agarose gel with ethidium bromide staining. All marker lots undergo stringent nuclease testing and are shown to be stable overnight at 37°C.

ZipRuler Express DNA Ladder Set, ready-to-use (Thermo Scientific™)

Thermo Scientific ZipRuler Express DNA Ladder Set contains two ladders: ZipRuler Express DNA Ladder 1 and ZipRuler Express DNA Ladder 2. Both are mixtures of chromatography-purified individual DNA fragments.The set is designed for fast and highly accurate sizing of a broad range (100-20,000 bp) of DNA fragments and their approximate quantification under a variety of electrophoresis conditions. The band pattern is not affected by the composition of electrophoresis buffer, voltage or gel percentage (see Table 1).

Features
• For fast separation, the ZipRuler Express DNA Ladder 1 and 2 are loaded into two different wells in the same gel to evaluate a wide range of DNA bands during short electrophoresis runs. For longer runs, equal amounts of both ladders are loaded into the same gel well. Three reference bands will then be formed during electrophoresis of the mixed ZipRuler ladder. Longer electrophoresis run times are needed to achieve full separation of complex ladder.

ZipRuler Express DNA Ladder 1 is premixed with the 6X Orange DNA Loading Dye (orange G and xylene cyanol), whereas ZipRuler Express DNA Ladder 2 is premixed with the 6X MassRuler DNA Loading Dye (bromophenol blue).

When both ladders are loaded in the same gel lane, the migration of DNA is monitored by three electrophoresis tracking dyes (bromophenol blue, xylene cyanol FF and orange G).

RiboRuler Low Range RNA Ladder, ready-to-use (Thermo Scientific™)

Thermo Scientific RiboRuler Low Range RNA Ladder 100-1000 bp, ready-to-use, is a mixture of 7 chromatography purified single-stranded RNA transcripts (100–1000 bp long), produced from specific templates that contain a fragment of the pTZ19R polylinker and Lambda phage fragments.

This ladder is premixed with loading dye.

RiboRuler Low Range RNA Ladder, ready-to-use, is composed of chromatography-purified transcripts what makes them free from NTPs and RNA degradation products. The concentration of each RNA transcript is determined spectrophotometrically. To minimize the possibility of contamination during the use the ladder is aliquotted in smaller volumes.

RiboRuler Low Range RNA Ladder is optimal for end-labeling using T4 Polynucleotide Kinase (see protocol in the Resources) and is ideal for Northern blots.

Lambda DNA/EcoRI plus HindIII Marker (Thermo Scientific™)

Thermo Scientific Lambda DNA/EcoRI+HindIII Marker is recommended for sizing of linear double-stranded large DNA fragments in agarose gels. Lambda DNA is digested to completion with the appropriate Thermo Scientific restriction enzyme(s) and purified and dissolved in storage buffer. DNA fragments contain various sticky ends depending on the restriction enzyme used for preparation of the marker.

Lambda DNA/EcoRI+HindIII Marker can be labeled radioactively with T4 Polynucleotide Kinase. Alternatively, and it can be labeled radioactively or non-radioactively with Klenow fragment or Klenow Fragment, exo- with the fill-in reaction (see Resources for Protocols).

Note
The cohesive ends (the 12nt cos site of lambda DNA) of fragments indicated may anneal and form additional bands. These fragments can be separated by heating at 65°C for 5minutes and then cooling on ice for 3minutes.

The shortest fragments (oblique) are not visible in standard electrophoresis. Fragment lengths are predicted by computer analysis of respective DNA sequences.
