Anti-Clumping Agent (Gibco™)

GIBCO® Anti-Clumping Agent is a concentrated liquid reagent that reduces cell clumping to assist in attaining higher viable cell densities in suspension culture. Anti-Clumping Agent is a gentle, animal origin-free, chemically defined formulation that contains no proteins, enzymes, hydrolysates, or components of unknown composition.

Simple and Gentle Cell Dissociation
Cell lines adapted to suspension culture can revert to a more adherent or aggregated “clumping" state. When this occurs, culture condition can be greatly improved by adding a gentle reagent to reduce clumping and adhesion instead of the harsh enzymes often found in cell dissociation products. Anti-Clumping Agent is a simple, gentle, concentrated solution which, when added to culture media (usually titrated between a 1:100 and 1:1000 dilution), can reduce cell clumping significantly.

Reduced Clumping, Increased Viability, Improved Protein Expression
Reduced cell clumping achieved through use of Anti-Clumping Agent is illustrated below (Fig 1). See also the data demonstrating improved viable cell density (Fig 2). Higher viable cell density contributes to the stabilization of culture conditions for better protein yield.

For actual cell dissociation, we recommend our animal origin-free TrypLE™ products.

Use Anti-Clumping Agent after Transfection
Anti-Clumping Agent is not recommended in all suspension cell culture conditions. For example, when creating a cell line using lipid cationic transfection, Anti-Clumping Agent can interfere with the transfection process, so in this case we recommend that Anti-Clumping Agent be used after transfection.

Animal Origin-free and Chemically Defined Components
Because Anti-Clumping Agent is made with only animal origin free and chemically defined components, you can be assured that the risk of adding adventitious agents to your culture system is minimized. Avoiding additives in your system that are not well defined can lead to more robust results from batch to batch.

Convenient and Customizable Packaging
Previously, Anti-Clumping Agent was only available in a 20 mL bottle. However, it is now available in a more economical and convenient 100 mL bottle. Cell Dissociation Buffer, enzyme-free, PBS (Gibco™)

Gibco® Cell Dissociation Buffer is a membrane-filtered, isotonic, and enzyme-free solution of salts, chelating agents, and cell-conditioning agents in calcium-free and magnesium-free phosphate-buffered saline (PBS). Gibco® Cell Dissociation Buffer is suitable for the gentle dissociation of mammalian cells for studies such as ligand binding, flow cytometry, and immunohistochemistry, that require intact cell surface proteins. Gibco® Cell Dissociation Buffer works well with lightly adherent cell lines, such as HeLa and NIH 3T3, but is not recommended for the routine passaging of strongly adherent cells. Dispase II, powder (Gibco™)

Dispase II (neutral protease) is an amino-endo peptidase that hydrolyzes the N-terminal peptide bonds of non-polar amino acid residues. This sequence is found with high frequency in collagen. Dispase II has a mild proteolytic action that makes it useful for the isolation and routine passaging of primary cells. Dispase II is relatively gentle, dissociates well at physiological temperature and pH, and generally maintains cell membrane integrity.

Gibco® Dispase II is produced in Bacillus polymyxa and packaged as a lyophilized, non-sterile powder for research use in cell or tissue dissociation, and to prevent clumping in suspension cultures. Gibco® Dispase II activity is guaranteed to be greater than 0.5 units/mg.

Collagenase, Type I, powder (Gibco™)

Room-Temperature Stable
TrypLE™ Express is room-temperature stable and ready to use. TrypLE™ Express remains stable for 24 months at room temperature or 2–8°C, making storage and handling easy and convenient.

Easy to Use
TrypLE™ Express can be directly substituted for trypsin in existing protocols. In addition, dilution alone inactivates TrypLE™ Express, avoiding the need for trypsin inhibitors, such as FBS.

Animal Origin-Free (AOF)

StemPro™ Accutase™ Cell Dissociation Reagent (Gibco™)

StemPro® Accutase® Cell Dissociation Reagent is a cell detachment solution of proteolytic and collagenolytic enzymes. Accutase® does not contain mammalian or bacterial derived products and is useful for the routine detachment of cells from standard tissue culture plasticware and adhesion coated plasticware, including Geltrex™, CELLStart™ and polymers. Accutase® performs exceptionally well in detaching primary and stem cells and maintaining high cell viabilty compared to animal origin enzymes such as trypsin. Some downstream applications following Accutase® treatment include analysis of cell surface markers, virus growth assay, cell proliferation, tumor cell migration assays, routine cell passage, production scale-up (bioreactor), and flow cytometry. Cell lines tested for Accutase® application include human embryonic stem cells, human mesenchymal stem cells, human neural stem cells and primary cells including macrophages, fibroblasts, keratinocytes, vascular endothelial cells, hepatocytes, and established cell lines such as adherent CHO and BHK cells and 293 cells. CTS™ Versene

CTS Versene Solution is a cGMP-manufactured EDTA solution for use as a gentle non-enzymatic cell dissociation reagent. CTS Versene Solution (0.48 mM) is formulated as 0.2 g EDTA(Na4) per liter of phosphate buffered saline (PBS). It reduces the burden in qualifying reagents when transitioning from the research bench to the clinic.

Facilitates regulatory filings
CTS Versene Solution is manufactured at a site that uses methods and controls that conform with cGMP for medical devices, 21 CFR Part 820. Trypsin-EDTA (0.25%), phenol red (Gibco™)

This liquid formulation of trypsin contains EDTA and phenol red. Gibco Trypsin-EDTA is made from trypsin powder, an irradiated mixture of proteases derived from porcine pancreas. Due to its digestive strength, trypsin is widely used for cell dissociation, routine cell culture passaging, and primary tissue dissociation. Quality testing

Soybean Trypsin Inhibitor, powder (Gibco™)

Soybean Trypsin Inhibitor is one of several protease inhibitors found in soybeans. Trypsin is inhibited at a molar ratio of 1:1, while chymotrypsin and plasmin are inhibited to a lesser extent. Soybean trypsin inhibitor will also inhibit other serine proteases.

Gibco® Soybean Trypsin Inhibitor, powder has an activity greater than 7,000 BAEE units per mg. A 0.5 mg/mL Soybean Trypsin Inhibitor stock solution should be made by dissolving the non-sterile powder in 0.01M Borate Buffer or Dulbecco's Phosphate Buffered Saline (DPBS), without calcium and magnesium. Filter sterilize the stock solution and store for up to a week at 2 - 8°C.
