PDCD1 Recombinant Human Protein, hIgG1-Fc.His Tag Thermo Scientific™

Programmed cell death 1 (PDCD1), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in protein studies such as protein structure analysis and protein-protein interactions. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant protein was expressed from a DNA sequence encoding the extracellular domain (Met 1-Gln 167) of human PDCD1 (NP_005009.2) fused to the polyhistidine-tagged Fc region of human IgG1 at the C-terminus.

Activity: Measured by its binding ability in a functional ELISA
. Immobilized recombinant human PDCD1LG2 at 1 µg/ml (100 µl/well) can bind human PDCD1-Fc chimera with a linear range of 7.8-1000 ng/ml

NGF Recombinant Human Protein Thermo Scientific™

Nerve growth factor (NGF) recombinant human protein is supplied as a lyophilized powder. It is suitable for use in analysis of protein structure and performing cell based assays. It can also be used as an immunogen, as a protein standard, or in other research applications.

This recombinant protein was expressed from a DNA sequence encoding the mature form of human NGF (NP_002497.2) (Ser 122-Arg 239).

N-terminal Sequence Analysis: Ser 122

Activity: Measured in a cell proliferation assay using TF-1 human erythroleukemic cells. The EC50 for this effect is typically 0.4-1.0 ng/mL.

Formulation: Lyophilized in 50 mM NaAc, 150 mM NaCl, pH 5.5, 5% mannitol, 5% trehalose, 0.02% Tween®-80.

Background
As the first one of a series of neurotrophic factors, nerve growth factor (NGF) is important for the development and maintenance of the sympathetic and sensory nervous systems. NGF was identified as a large complex consisting of three non-covalently linked subunits, α, β, and γ, among which, the β subunit, called β-NGF, was demonstrated to exhibits the growth stimulating activity of NGF. β-NGF is a target-derived secreted homodimeric protein of two 120-amino acid mature polypeptides after proteolytic removal of the signal and pro peptide. NGF acts via at least two receptors on the surface of cells, TrkA and p75 receptors to regulate neuronal survival, promote neurite outgrowth, and up-regulate certain neuronal functions such as mediation of pain and inflammation. In additon to its neurotropic activities, studies indicated that NGF may also have an important role in the regulation of the immune system. The synthesis of β-NGF in astrocytes is enhanced by various other cytokines including IL-1, TNF-α, PDGF and TGF-β .Defects in β-NGF are the cause of hereditary sensory and autonomic neuropathy type 5 (HSAN5) characterized by degeneration of dorsal root and autonomic ganglion cells, as well as sensory and/or autonomic abnormalities.

Background
Programmed Cell Death 1 is a type I transmembrane immunoreceptor and belongs to the CD28 immunoglobulin superfamily that mediates immune responses. PD1 is expressed on activated T-cells, B cells, myeloid cells and a subset of thymocytes. PD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN-γ by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. Furthermore, PD1 is shown to be a regulator of virus-specific CD8+ T cell survival in HIV infection.

Activity:

Background
Interleukin-6 (IL-6) is a multifunctional α-helical cytokine that regulates cell growth and differentiation of various tissues, and is known particularly for its role in the immune response and acute phase reactions. IL-6 is also involved in hematopoiesis, bone metabolism, and cancer progression, and has been defined an essential role in directing transition from innate to acquired immunity. IL-6 is secreted by a variety of cell types including T cells and macrophages as phosphorylated and variably glycosylated molecule with the molecular mass of 22 kDa-28 kDa. Three IL-6 signaling pathways (STAT1, STAT3, and Ras-dependent MAPK cascade) have been reported. Dysregulated production of IL-6 and IL6R are implicated in the pathogenesis of several inflammatory diseases and malignancies such as multiple myeloma, rheumatoid arthritis, or osteoporosis.

Background
Cluster of Differentiation 19 (CD19), is a single-pass type I membrane protein which contains two Ig-like C2-type (immunoglobulinlike) domains. CD19 forms a complex with CD21, CD81 and CD225 in the membrane of mature B cells. It interacts with VAV. It interacts with GRB2 and SOS when phosphorylated on Tyr-348 and/or Tyr-378. CD19 also interacts with PLCG2 when phosphorylated on Tyr-409. CD19 is an important pan B cell marker and co-stimulatory protein in humans and mice. CD19 is a hallmark differentiation antigen of the B cell lineage and positively regulates antigen receptor signal transduction in mature B cells. CD19 defines signaling thresholds for the Ag receptor as well as other cell surface receptors that regulate B lymphocyte selection, activation, and differentiation. Defects in CD19 are a cause of hypogammaglobulinemia.

Background
Tumor necrosis factor alpha (TNF-α), also known as cachectin and TNFSF1A, is the prototypic cytokine of the TNF superfamily, and is a multifunctional molecule involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF-α is produced mainly by macrophages, and large amounts of this cytokine are released in response to lipopolysaccharide, other bacterial products, and Interleukin-1 (IL-1). TNF-α is primarily produced as a type I I transmembrane protein arranged in stable homotrimers, and a soluble trimeric form of TNF-α is released via proteolytic cleavage by the metalloprotease TACE/ ADAM17. Two receptors mediating the TNF-α function have been identified, the ubiquitous TNF RI from which most signaling are derived, and the hematopoietic cell-restricted TNF RII. Upon the TNF-α trimers binding, TNF receptors also form trimers, undergo a conformational change, and enable the adaptor protein TRADD to bind to the death domain, and thus initiates the signal pathways such as the NF-kB, Jak/STAT, and the MAPK pathways, as well as the death signaling. As a major mediator of apoptosis, inflammation and immunity, TNF-α has been implicated in the pathogenesis of a variety of human diseases including autoimmune diseases, insulin resistance, and cancer.

Intercellular adhesion molecule 1 (ICAM1) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in performing cell based assays. It can also be used as a protein standard, or in other research applications.

This recombinant human protein is expressed from a DNA sequence encoding the extracellular domain of human ICAM1 (NP_000192.2) (Met 1 - Glu 480) fused to the polyhistidine-tagged Fc region of human IgG1 at the C-terminus.

Background
ICAM1 (Inter-Cellular Adhesion Molecule 1), also known as CD54, is a type of intercellular adhesion molecule constitutely present in low concentrations in the membranes of leukocytes and endothelial cells. As an endothelial and leukocyte-associated transmembrane protein, ICAM1 is well known for its importance in stabilizing cell-cell interactions and facilitating leukocyte endothelial transmigration.ICAM1 is a ligand for LFA-1(integrin). When activated, leukocytes bind to endothelial cells via ICAM-1 / LFA-1 interaction and then transmigrate into tissues. Presence with heavy glycosylation and other structural characteristics, ICAM1 possesses binding sites for a number of immune-associated ligands and serves as the binding site for entry of the major group of human Rhinovirus (HRV) into various cell types. ICAM1 also becomes known for its affinity for Plasmodium falciparum-infected erythrocytes (PFIE), providing more of a role in infectious disease.

FGF10 Recombinant Human Protein Thermo Scientific™

Fibroblast Growth Factor 10 (FGF10), recombinant human protein is supplied as a lyophilized powder. In general, recombinant proteins can be used as protein stucture analysis and in cell biology research applications.

This recombinant protein was expressed from a DNA sequence encoding the mature form of human FGF10 (NP_004456.1) (Gln 38-Ser 208).

Nerve growth factor (NGF), recombinant mouse protein is supplied as a lyophilized powder. It is suitable for use in performing cell based assays. It can also be used as a protein standard, or in other research applications.

This recombinant protein was expressed from a DNA sequence encoding the mature form of mouse NGF (NP_001106168.1), residues 122 to 241, with an initial Met at the N-terminus.

Activity: Measured in a cell proliferation assay using TF1 human erythroleukemic cells. The ED50 for this effec is 0.2-2.0 ng/ml.

Formulation: Lyophilized in 50 mM Na acetate buffer, pH 5.5, 150 mM NaCl, 5% Mannitol, 5% Trehalose, and 0.02% Tween 80.

Cluster of Differentiation 4 (CD4), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in analysis of protein structure, studying protein-protein interactions, and cell biology research applications. It can also be used as an immunogen, or as a protein standard.

This recombinant protein was expressed from a DNA sequence encoding the extracellular domain (Met 1-Trp 390) of human CD4 (NP_000607.1) fused to a polyhistidine tag at the C-terminus.

Activity:
1. Measured by its ability to bind towith HIV-1 gp120 in a functional ELISA.< BR> 2. Measured by the ability of the immobilized protein to support the adhesion of HeLa human cervical epithelial carcinoma cells . When 5 x 104 cells/well are added to CD4 coated plates (2.5 µg/mland 100 µl/well), approximately 60-80% will adhere specifically after 60 minutes at 37°C.

Background
The green fluorescent protein (GFP) is a protein that exhibit bright green fluorescence when exposed to blue light. The protein is in the shape of a cylinder, comprising 11 strands of β-sheet with an α-helix inside and short helical segments on the ends of the cylinder. Its ability to generate a highly visible, efficiently emitting internal fluorophore is both intrinsically fascinating and tremendously valuable. The green-fluorescent protein (GFP) of the jellyfish Aequorea victoria has long been used as a universal reporter in a broad range of heterologous living cells and organisms. GFP has become well established as a marker of gene expression and protein targeting in intact cells and organisms.

Activity:
1. Measured by its binding ability in a functional ELISA. Immobilized cynomolgus IL-13 at 4 µg/ml (100 µl/well) can bind human IL13RA1 with a linear ranger of 0.156-2.5 µg/ml.
2. Measured in a cell proliferation assay using TF-1 human erythroleukemic cells. The specific activity is 5-20 ng/ml.

Background
CD36, is integral membrane protein found on the surface of many cell types in vertebrate animals. It belongs to the CD36 family and is found on platelets, erythrocytes, monocytes, differentiated adipocytes, mammary epithelial cells, spleen cells and some skin microdermal endothelial cells. CD36 has also been implicated in hemostasis, thrombosis, malaria, inflammation, lipid metabolism and atherogenesis. CD36 seems to have numerous potential physiological functions. It binds to collagen, thrombospondin, anionic phospholipids and oxidized LDL. It directly mediates cytoadherence of Plasmodium falciparum parasitized erythrocytes and binds long chain fatty acids. CD36 may also function as a cell adhesion molecule and in the transport and/or as a regulator of fatty acid transport. Defects in CD36 are the cause of platelet glycoprotein IV deficiency also known as CD36 deficiency. Delta-like protein 4 (DLL4) is a type I membrane-bound Notch ligandcharacterized by an extracellular region containing several EGF-likedomains and a DSL domain required for receptor binding, and issuggested to play a key role in vascular development and tumorangiogenesis. The Notch pathway is an evolutionary conservedintercellular signaling pathway involved in numerous biological processesincluding cell fate determination, cellular differentiation, proliferation, survival, and apoptosis. In mammalian cells, five Notch ligands (Jagged1, 2, DLL1, 3, 4) and four Notch receptors (Notch1-4) have been identified, and ligand-receptor interactions results in proteolysis and translocation of the Notch intracellular domain. Background
Human epidermal growth factor (EGF) exerts actions on a wide variety of biological processes, and plays an important role in the regulation of cell growth, proliferation, and differentiation mediated by its high-affinity receptor EGFR on the cell surface. Triggering receptor expressed on myeloid cells 2 ( TREM2 ) is a single Ig domain receptor. It is expressed on macrophages and dendritic cells but not on granulocytes or monocytes.
