The next-generation Thermo Scientific Labtainer Pro BioProcess Container (BPC) offers a versatile solution for liquid-handling applications when used in conjunction with our comprehensive single-use portfolio. The Labtainer Pro BPC is an ideal solution for use in a variety of applications, including sample preparation, cell culture processes, storage and transport, as well as in custom configurations to fit into your unique workflow. Labtainer Pro BPCs provide flexibility and assurance—without compromise. They are offered in a 2D pillow style with available sizes ranging from 50 mL to 20 L in a variety of porting configurations. Labtainer Pro BPC features include:

MicroAmp™ Optical 96-Well GPLE Reaction Plates with Barcode Applied Biosystems™

MIG (CXCL9) Mouse ProcartaPlex™ Simplex Kit Invitrogen™

The Mouse MIG (CXCL9) Simplex ProcartaPlex Kit measures MIG (CXCL9) protein and is designed to be combinable with other Simplex kits so that you can create your own multiplex panel that utilizes Luminex xMAP technology for protein detection/quantitation. When combining multiple Simplex kits (i.e., when you are not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed for each assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate all protein targets simultaneously using a Luminex instrument. ProcartaPlex multiplex assays require as little as 25 µL of plasma or serum, or 50 µL of cell culture supernatant, and just four hours to obtain analyzed results.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for the simultaneous detection and quantitation of up to 65 protein targets in a single 25–50 µL sample from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create spectrally unique signatures that can be identified by the Luminex xMAP detection systems (e.g. Luminex 200, FLEXMAP 3D, and MAGPIX). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a multiplexed assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

The improved port design features tapered sides that facilitate a structurally stronger seal. The manufacturing process for the Labtainer Pro BPC incorporates impulse heat sealing for port insertion utilizing Labtainer Automated Manufacturing (LAM) technology. This fully automated method of sealing enables a higher level of control over the amount of heat applied, resulting in a consistent and strong seal throughout the finished product. With no change in materials or components, users of CX5-14- and Aegis5-14-based BPCs will be able to adopt Labtainer Pro BPCs with minimal to no qualification.

Quality: Labtainer Pro BPCs are inspected after manufacturing, prior to being packaged and shipped. A 100% helium integrity test is also conducted, helping ensure that the most integral product is delivered to the customer. In addition, lot-based bacterial endotoxin testing (BET) and particulate analysis testing (PAT) of Labtainer Pro BPC products has been implemented, to established industry standards. Plus, the outer packaging of the Labtainer Pro BPC was developed to prevent damage during unpacking from sharps or other tools, as well as to reduce outer packaging material by approximately 25%.

Ease of use: The top handle featured on the Labtainer Pro BPC has been improved for more ergonomic handling. The addition of a lower handle on the 2, 5, 10, and 20-L units allows for more ergonomic manipulation. The shape of the BPC chamber has also been redesigned to allow for easy handling of larger BPCs. The Labtainer Pro BPC provides optimal drainage through low profile ports that do not overly extend into the chamber and features chamber geometry that tapers to the ports, reducing the holdup volume at the bottom of the BPC.

TrueMark™ MSI Assay Applied Biosystems™

The Applied Biosystems TrueMark MSI Assay is a low-input, single-amplification solution for the rapid identification of microsatellite instability (MSI) in formalin-fixed paraffin-embedded (FFPE) samples from multiple tumor tissue types for research applications. The TrueMark MSI Assay leverages an expanded 6-dye set to incorporate additional markers and does not require the use of a tumor-normal match, thereby cutting in half the number of wells required for each sample, as well as reducing the sample handling burden. The assay is compatible with the Applied Biosystems SeqStudio and 3500 Series genetic analyzers.

MSI analysis software
The associated TrueMark MSI Analysis Software (included at no additional charge) is designed specifically for the TrueMark MSI Assay to enable automated calling of markers and samples, offering greater ease of analysis by reducing the manual QC, analysis, and review required to determine sample status. The automated calling functions without the use of a tumor-normal match, but it is also compatible with tumor-normal pairs.

Diploid Growth Serum-Reduced Medium (SRM) Gibco™

The Gibco Diploid Serum-Reduced Medium (SRM) system is designed to support vaccine manufacture under serum-reduced or serum-free conditions for cell types traditionally cultured in classical media with 10% serum. With this system, human diploid cells, such as MRC-5, Wi38, and KMB17, can be grown with just 1–2% serum. Additionally, it supports growth of chicken embryo fibroblasts under serum-free conditions. Virus production can then be conducted under serum-free conditions without the addition of albumin or other supplements.

The Diploid SRM system is composed of two different kits:
1. Diploid Growth Serum-Reduced Medium (SRM) kit (this page): A basal medium and 100X growth supplement designed for cell expansion
2. Diploid Production Serum-Free Medium (SFM) kit: A basal medium and 100X production supplement designed for virus production

Human cells have traditionally been used to manufacture a variety of vaccines, such as the varicella zoster, polio, smallpox, hepatitis A, measles, mumps, and rubella vaccines. Traditionally, the only way to culture these cells has been to use classical media with 10% serum for growth and 1–5% human albumin (human sourced or recombinant) for virus production. These are two of the highest cost components in vaccine production. The Diploid SRM system helps reduce or eliminate both components while maintaining high virus titers.

Cost savings
Reduction or elimination of serum can significantly reduce cost for a vaccine manufacturing process. Premium FBS is expensive with high price volatility. Given the typical cost pressures on vaccine production, both economic drivers make vaccine manufacturing to a target price extremely challenging. Diploid Growth SRM reduces the impact of this volatility in price and offers a level of cost consistency not available with processes that use serum in high volumes.

In addition, serum requires frozen storage. A 10% serum concentration in growth media can result in high frozen storage requirements for a vaccine process. A process requiring 1,000 L of growth medium would require the frozen storage of 100 L of serum. The Diploid Growth SRM kit comes with a 100X frozen supplement, enabling an 80% reduction in frozen storage requirements. 1,000 L of Diploid Growth SRM would only need 20 L of frozen storage: 10 L for FBS and 10 L for the 100X supplement.

Albumin is commonly added to virus production media. With the Diploid Production SFM, viruses can be produced at comparable titers without any additional supplementation.

No adaptation required
The Diploid SRM system was developed with existing cell banks in mind. Transitioning a vaccine process to a new medium can be challenging due to the need to adapt the cells to the new medium. For human cells such as MRC-5, there is the added risk of reaching the Hayflick limit due to the added passages necessary during cell adaptation and subsequent re-banking of the cells. The transition to the Diploid SRM system does not require adaptation or re-banking of cells when using the Diploid Growth SRM kit. Existing cGMP cells banked with serum may be thawed directly in Diploid Growth SRM supplemented with 1% FBS, allowing a simple transition to this medium.

Mitigate serum supply risk
Obtaining a steady supply of serum can be challenging, especially if the serum required is from a BSE/TSE safe region such as New Zealand or Australia. Serum supply can fluctuate based on several environmental (drought, cattle disease) and economic (feed cost, dairy pricing) factors. With Diploid Growth SRM this dependence is substantially reduced due to the reduced serum requirements.
