FCGR3A (CD16a) Recombinant Human Protein, 176 Val, His Tag

Fc gamma receptor III-a (FCGR3A/CD16a), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in analysis of protein structure, studying protein-protein interactions, and cell biology research applications. It can also be used as an immunogen, or as a protein standard.

This recombinant protein was expressed from a DNA sequence encoding the extracellular domain (Met 1-Gln 208) of human FCGR3A (P08637-1), containing the mutation 176 Phe/Val, precursor fused to a polyhistidine tag at the C-terminus.

Activity: Measured using the Octet RED System, the affinity constant (Kd) of FCGR3A bound to human IgG1 was 180nM.

Formulation: Lyophilized in PBS, pH 7.4, 5% Mannitol, 5% Trehalose, and 0.02% Tween 80.

Reconstitution: Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and be used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

EGF Recombinant Human Protein

Epidermal growth factor (EGF) recombinant human protein is supplied as a lyophilized powder. It is suitable for use in analysis of protein structure and performing cell based assays. It can also be used as an immunogen, as a protein standard, or in other research applications.

This recombinant protein was expressed from a DNA sequence encoding the mature form of human EGF (NP_001954.2) (Asn 971-Arg 1023) retaining the initial Met at the N-terminus.

N-terminal Sequence Analysis: Met 1

Activity: Measured in a cell proliferation assay using BALB/c 3T3 mouse embryonic fibroblasts (Rubin, J.S. et al. 1991, Proc. Natl. Acad. Sci. USA 88:415.) . The EC50 for this effect is typically 20-100 pg/mL.

Background
Human epidermal growth factor (EGF) exerts actions on a wide variety of biological processes, and plays an important role in the regulation of cell growth, proliferation, and differentiation mediated by its high-affinity receptor EGFR on the cell surface. It induces differentiation of specific cells, promotes angiogenesis, and inhibits gastric acid secretion in vivo, while serves as a potent mitogenic actor for a variety of cultured cells of both ectodermal and mesodermal origin. Human EGF was originally isolated from urine based on its inhibitory effect on gastric secretion and named urogastrone accordingly. EGF is the founding member of the EGF-family of proteins which are characterized by the presence of at least one EGF structural unit (a conserved 6 cysteine motif that forms three disulfide bonds). The EGF precursor is believed to exist as a membrane-bound molecule containing 9 EGF units, and is proteolyticallycleaved to generate the 53-amino acid peptide hormone.

CEACAM-5 Recombinant Human Protein, His Tag

IL6 Recombinant Human Protein

Interleukin 6 (IL-6) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in analysis of protein structure and performing cell based assays. It can also be used as an immunogen, as a protein standard, or in other research applications.

This recombinant protein is expressed from a DNA sequence encoding the mature form of human IL6 (NP_000591.1) (Val 30-Met 212) retaining the initial Met at the N-terminus.

N-terminal Sequence Analysis: Met.

Activity:
1. Measured by its binding ability in a functional ELISA. Immobilized recombinant human IL6 (100 µL/well of a 8 µg/mL solution) has been shown to bind recombinant human IL6R with a linear range of 1.25-20.0 ng/mL.
2. Measured in a cell proliferation assay using TF1 human erythroleukemic cells. The EC50 for this effect is typically 0.2-0.8 ng/mL.

Background
Interleukin-6 (IL-6) is a multifunctional α-helical cytokine that regulates cell growth and differentiation of various tissues, and is known particularly for its role in the immune response and acute phase reactions. IL-6 is also involved in hematopoiesis, bone metabolism, and cancer progression, and has been defined an essential role in directing transition from innate to acquired immunity. IL-6 is secreted by a variety of cell types including T cells and macrophages as phosphorylated and variably glycosylated molecule with the molecular mass of 22 kDa-28 kDa. Three IL-6 signaling pathways (STAT1, STAT3, and Ras-dependent MAPK cascade) have been reported. Dysregulated production of IL-6 and IL6R are implicated in the pathogenesis of several inflammatory diseases and malignancies such as multiple myeloma, rheumatoid arthritis, or osteoporosis.

MET (c-MET) Recombinant Human Protein (without Catalytic Activity Domain), His Tag

Met proto-oncogene/hepatocyte growth factor receptor (MET/c-MET), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in protein studies such as protein structure analysis and protein-protein interactions. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

Since this product is a truncated protein and does not contain intracellular catalytic activity domain, it is not suitable for use in enzyme activity studies..

This recombinant protein was expressed from a DNA sequence encoding the extracellular domain (Met 1-Thr 932) of human MET (NP_000236) fused to a polyhistidine tag at the C-terminus.

Activity: Measured by its binding ability in a functional ELISA. Immobilized recombinant human HGF at 10 µg/ml (100 µl/well) can bind biotinylated MET. The ED50 of biotinylated Met is 5.28 µg/ml.

Immunoglobulin G (IgG) is synthesized and secreted by plasma B cells, and constitutes 75% of serum immunoglobulins in humans. IgG antibodies protect the body against the pathogens by agglutination and immobilization, complement activation, toxin neutralization, as well as the antibody-dependent cell-mediated cytotoxicity (ADCC). Interleukin 1 Beta

Background
As the first one of a series of neurotrophic factors, nerve growth factor (NGF) is important for the development and maintenance of the sympathetic and sensory nervous systems. NGF was identified as a large complex consisting of three non-covalently linked subunits, α, β, and γ, among which, the β subunit, called β-NGF, was demonstrated to exhibits the growth stimulating activity of NGF. β-NGF is a target-derived secreted homodimeric protein of two 120-amino acid mature polypeptides after proteolytic removal of the signal and pro peptide. NGF acts via at least two receptors on the surface of cells, TrkA and p75 receptors to regulate neuronal survival, promote neurite outgrowth, and up-regulate certain neuronal functions such as mediation of pain and inflammation. In additon to its neurotropic activities, studies indicated that NGF may also have an important role in the regulation of the immune system. The synthesis of β-NGF in astrocytes is enhanced by various other cytokines including IL-1, TNF-α, PDGF and TGF-β .Defects in β-NGF are the cause of hereditary sensory and autonomic neuropathy type 5 (HSAN5) characterized by degeneration of dorsal root and autonomic ganglion cells, as well as sensory and/or autonomic abnormalities.

DLL4 Recombinant Human Protein, hIgG1-Fc Tag

Delta-like 4 (Drosophila) (DLL4) recombinant human protein is supplied as a lyophilized powder. Background
Delta-like protein 4 (DLL4) is a type I membrane-bound Notch ligandcharacterized by an extracellular region containing several EGF-likedomains and a DSL domain required for receptor binding, and issuggested to play a key role in vascular development and tumorangiogenesis. The Notch pathway is an evolutionary conservedintercellular signaling pathway involved in numerous biological processesincluding cell fate determination, cellular differentiation, proliferation, survival, and apoptosis. In mammalian cells, five Notch ligands (Jagged1, 2, DLL1, 3, 4) and four Notch receptors (Notch1-4) have been identified, and ligand-receptor interactions results in proteolysis and translocation of the Notch intracellular domain. As a ligand for Notch1 and Notch 4, DLL4 is selectively expressed in the developing endothelium and in some tumor endothelium, and is induced by vascular endothelial growth factor (VEGF)-A and hypoxia. In recent studies, it has been revealed that this DLL4 inhibition may paradoxically lead to increased angiogenesis but poor tumor growth because of the non-functional neovascularization, and constitutive expression of DLL4 may leads to a lethal lymphoproliferative disease. Accordingly, this may provide a new therapeutic approach for certain carcinomas.

Background
Triggering receptor expressed on myeloid cells 2 ( TREM2 ) is a single Ig domain receptor. It is expressed on macrophages and dendritic cells but not on granulocytes or monocytes. TREM2 may play a role in chronic inflammations and may stimulate production of constitutive rather than inflammatory chemokines and cytokines. TREM2 forms a receptor signaling complex with TYROBP and triggers activation of the immune responses in macrophages and dendritic cells. It also associates with the signal adapter protein, DAP12, which has a cytoplasmic ITAM, leading to the subsequent activation of cytoplasmic tyrosine kinases. TREM2 is both required and sufficient for competent uptake of apoptotic neuronal cells. TREM2 and TREM2-L form a receptor-ligand pair connecting microglia with apoptotic neurons, directing removal of damaged cells to allow repair. Deficiency of the adapter protein DAP12 or its associated receptor TREM2 is associated with abnormal osteoclast development in humans. Defects in TREM2 are causes of PLOSL, also known as NHD. In addition, TREM2 signaling is also an important pathway to promote healing.
