Oncomine™ Myeloid Assay GX v2 Ion Torrent™

The Ion Torrent Oncomine Myeloid Assay GX v2 is a comprehensive targeted next-generation sequencing (NGS) assay designed for sensitive detection of myeloid disorder-associated DNA mutations and RNA fusion transcripts in blood and bone marrow samples. This assay is compatible with the Genexus Integrated Sequencer, which performs library preparation, sequencing, analysis, and reporting in an automated sample-to-result workflow. Depending on the workflow, results can be obtained in as little as a single day.

Oncomine Myeloid Assay GX v2 features include:

The Oncomine Myeloid Assay GX v2 is designed to assist in myeloid cancer research. The DNA panel is comprised of 45 key genes, while the RNA panel includes a broad fusion panel of 30 driver genes, covering the most relevant targets associated with major myeloid disorders, including acute myeloid leukemia (AML), myeloid dysplastic syndrome (MDS), myeloproliferative neoplasms (MPN), chronic myeloid leukemia (CML), chronic myelomonocytic leukemia (CMML), and juvenile myelomonocytic leukemia (JMML).

The multiplex primer design leverages Ion AmpliSeq technology to generate results from multiple samples in a single run. Sequencing results are automatically analyzed by Genexus software using an optimized assay-specific analysis workflow.

The assay kit includes three pools of Ion AmpliSeq oligonucleotide primers (a 2-pool DNA panel and 1-pool RNA panel) and Genexus Strips 1 & 2-AS library reagents, enough to sequence up to 32 samples on the Genexus Integrated Sequencer. One end-to-end sequencing run uses one lane on the GX5 Chip and accommodates up to eight samples. Additional Genexus sequencer reagents and supplies must be ordered separately.

Bigfoot™ Calibration Beads

Bigfoot Calibration Beads are a mix of three different calibration beads used in the automated QC of the Bigfoot Spectral Cell Sorter. The beads are retained within the Bigfoot cell sorter in a dedicated loader position and are used during the automated QC process to verify alignment and drop delay of the system. No pre-mixing or lot monitoring are required. The onboard QC beads allow the operator to perform automated QC at any time and assists with remote support. The Bigfoot QC bead bottle is automatically capped to minimize evaporation from the vial, and the loader location contains an integrated vortex to keep the beads suspended.

Ponceau S Staining Solution Thermo Scientific™

Ponceaus S Staining Solution is a ready-to-use membrane stain for evaluating the transfer efficiency of a western blot. This stain is compatible with nitrocellulose and PVDF membranes. Ponceau S staining is reversible and can be removed with a short incubation in 0.1% NaOH.

Ponceau S membrane staining is a nondestructive, reversible method to stain and detect proteins on nitrocellulose and PVDF membranes. The stain has minimal nonspecific interactions with the membrane surface and provides a reliable method for visualization of protein transfer to a membrane. The staining protocol is simple and quick and results in reddish-pink bands that are easily photographed. The stain can be reversed easily in less than five minutes. Subsequent western blot detection is unaffected as the stain does not alter the membrane-bound protein.

EVOS™ Light Cube, Cy7 2.0 Invitrogen™

Our second-generation EVOS LED light cubes provide superior illumination uniformity across the field of view, resulting in exceptional tile-stitch image quality. At the same time, optimized spectral fidelity across channels significantly improves data quality by minimizing undesired bleed-through. Like the original design, they combine condensing optics and hard-coated filters into one remarkable light engine that delivers digitally controlled fluorophore excitation. These LED light cubes are backwards compatible with older EVOS and Countess systems and can be used with first-generation light cubes. Like the original light cubes, these LED cubes are user-changeable and automatically recognized by the instrument. The EVOS Light Cube, Cy7 2.0, is ideal for use with Cy7 and Alexa Fluor 750, and other fluorophores with excitation and emission maxima near 716 and 794 nm.

Efficient light engines
Our second-generation light cube design dramatically improves on the original. A larger aperture increases illumination uniformity, which affords superb tile-stitched image quality and consistency. In addition, new LED light and filter combinations maximize the signal-to-background ratio so critical for dim samples, improving data quality irrespective of signal strength for both EVOS and Countess systems. By placing the LED light cube as close as possible to the objective turret, the number of optical elements in the light path is minimized. High-intensity illumination over a short light path increases the efficiency of fluorophore excitation, providing better detection of weak fluorescent signals.

Hard-coated filter sets for higher transmission efficiencies
Hard-coated filters are more expensive but have sharper edges and significantly higher transmission efficiencies that typically result in >25% higher light transmission compared to soft-coated filters. LED bulbs and filters optimized to maximize the signal from target fluorophores, while minimizing that from other dyes (i.e., bleed-through), will help you obtain brighter fluorescence, higher transmission efficiencies and ability to detect faint fluorescence signals, and better signal-to-noise ratios. These factors raise both image and data qualities.

Continuous light intensity
Mercury arc lamps can decrease in intensity by 50% in the first 100 hours of operation—plus, images acquired in different sessions cannot be quantitatively compared using mercury illumination without complicated calibrations. EVOS cubes retain continuous light cube intensity that affords consistent illumination from day-to-day and experiment-to-experiment and help you to directly compare quantitative results from images acquired on different days. Each light cube contains a precisely matched set of optical components to optimize the position, evenness, and intensity of the light beam. These digitally controlled light sources make it easy to adjust light intensity, exposure time, and camera gain that can reduce phototoxicity and photobleaching.

Less expensive to own and maintain
The LED bulbs on the EVOS systems are rated for >50,000 hours (∼17 years), compared to 300 hours for a typical mercury bulb and 1,500 hours for a metal halide bulb. This translates to savings in the overall upkeep of your instrument.

Confidently identify microbes
The MicroSEQ ID sequence libraries consist of bacterial libraries of 500-bp 16S rDNA sequences and over 9500 validated entries, with extensive coverage of gram-negative non-fermenters, Bacillus, coryneforms, mycobacteria, and Staphylococcus, as well as a fungal library containing more than 1700 entries. To help ensure accurate identification of the species of the sequences, all library entries have been rigorously validated. And unlike other widely available databases, MicroSEQ libraries contain only correctly identified organisms and quality-controlled sequences derived from type strains of preeminent culture collections. The libraries also support sequence polymorphisms and include bacterial Category A biothreat agents.

Automatically identify unknown specimens
Reviewing each specimen in a project file to make a manual identification call can be tedious and time consuming. The Auto-ID feature in MicroSEQ ID Software uses a set of editable parameters to automatically identify unknown specimens. The results table is displayed in the report, and an editable comments field allows additional information or manual identifications to be entered into the same table.
