Gibco EfficientFeed A+ 3X Supplement is a single-part liquid supplement for fed batch addition to the culture of multiple cell lines to assist with process development and maximization of bioreactor utilization. Compared to original CHO CD EfficientFeed A AGT nutrient supplement, EfficientFeed A+ 3X Supplement has components that are super-concentrated from 66.4 g/L (1X AGT) to 199.2 g/L (3X), helping reduce product dilution or increase titers with additional feeding. The Applied Biosystems CTS PureQuant assays are a family of assays that accurately identify and quantify specific immune cell types in a mixed population of cells. These assays are TaqMan qPCR-based and measure genomic DNA methylation at specific sites that serve as a unique identifier. The assays include standards and controls and each 96-well plate can accommodate up to seven test samples. Results are provided as a percentage of target cell type out of total input cells using a free analysis tool.

The CTS PureQuant Treg Assay utilizes a specific demethylated site in the transcription factor FOXP3 gene to positively identify regulatory T-cells. This specific site allows for discrimination of regulatory T-cells from activated CD4+ FOXP3+ conventional T-cells (see https://onlinelibrary.wiley.com/doi/full/10.1002/eji.200737594).

The Applied Biosystems MicroAmp TriFlex 96-Well PCR Reaction Plate is a versatile polypropylene PCR plate designed for use with Applied Biosystems standard (0.2 mL) 96-well and ProFlex 3x32-well thermal blocks. CyQUANT™ Direct Red Cell Proliferation Assay (Invitrogen™)

The CyQUANT Direct Red Cell Proliferation Assay is a simple no-wash, add-and-read fluorescent microplate assay that can be used to assess cell growth, viability, or compound toxicity. The assay consists of a cell-permeant DNA-binding red fluorescent probe and a cell-impermeant background suppressor. The probe and suppressor are mixed and added to cells; the probe stains all cells, while the suppressor only enters dead cells or cells with compromised cell membranes, suppressing probe fluorescence in those locations. The result is that only healthy cells are detected.

CyQUANT Direct Red Cell Proliferation Assays features include:
Convenient—no washes, cell lysis, or temperature equilibration; simply add and read to detect changes in adherent and suspension cell proliferation
Accurate―based on amount of cellular DNA, which is highly regulated and independent of the metabolic state of the cells
Robust―optimized red fluorescent probe and background suppressor result in a highly sensitive assay with a large dynamic range

The CyQUANT Direct assay is a red fluorescence-based proliferation and cytotoxicity assay for microplate readers with a linear detection range from less than 100 to 20,000 cells per well for most cell types. The no-wash, add-mix-read assay can be completed in one hour. The red fluorescent excitation and emission wavelengths result in an assay that is ideal for multiplexing with green fluorescent dyes or proteins. Additionally, the fluorescent signal is stable for several hours, for additional workflow convenience.

Since the amount of DNA in cells is highly regulated, detection methods based on determining the amount of DNA are highly accurate. The CyQUANT Direct Red Cell Proliferation Assay consists of a red fluorescent cell-permeant DNA-binding probe and a background suppressor dye. The probe and suppressor are mixed, added to cells, incubated for one hour, and then the fluorescence measured. The probe enters all cells, but the suppressor only enters dead cells or cells with compromised membranes. Upon entering dead and dying cells the background suppressor dye suppresses the signal from the fluorescent probe, resulting in the detection of only healthy cells. Thus, the CyQUANT Direct Red assay can be used to measure changes in proliferation as well as cytotoxicity.

CTS™ PureQuant™ CD8+ T-Cell Assay (Applied Biosystems™)

The Applied Biosystems CTS PureQuant assays are a family of assays that accurately identify and quantify specific immune cell types in a mixed population of cells. These assays are TaqMan qPCR-based and measure genomic DNA methylation at specific sites that serve as a unique identifier. The assays include standards and controls and each 96-well plate can accommodate up to seven test samples. Results are provided as a percentage of target cell type out of total input cells using a free analysis tool.

The CTS PureQuant CD8+ T-Cell Assay utilizes the CD8B gene as an epigenetic marker for CD8+ T cells; the assay targets regulatory elements within its third intron (see http://stm.sciencemag.org/content/10/452/eaan3508).

Subjected to bisulfite conversion followed by copy number estimation via qPCR standard curve. Allows estimation of assay-specific bisulfite efficiency factor: the ratio between TpG/GAPDH copy numbers measured from the same plasmid.
Reference: checks assay performance. Consists of genomic DNA extracted from human blood of pooled healthy donors. The reference values represent the expected range of the target cell type. For example, CD8+ cells are generally more abundant than Treg or Th17 cells.
Standards: determine the unknown copy number. Consist of six dilutions spanning 4-log linear dynamic range (30-31250 copies). Absolute copy numbers can reliably be determined using the slope and intercept of the respective standard curves.

eBioscience™ Streptavidin Super Bright 645 Conjugate (Invitrogen™)

eBioscience Streptavidin Super Bright 645 is a streptavidin-fluorochrome conjugate for indirect staining protocols to detect biotinylated primary antibodies in flow cytometry. Super Bright dyes are polymer-based fluorophores that are excited by the violet laser (405 nm) and emit fluorescence at the wavelength indicated in their name. They are optimized for use in flow cytometry and allow for use of additional channels with violet excitation. Super Bright dyes are fully compatible with other commonly used fluorescent molecules, eBioscience buffers and fixatives, viability stains, and reagents.

Super Bright 645 is a tandem dye that can be excited with the violet laser line (405 nm) and emits at 645 nm. We recommend using a 660/20 bandpass filter or equivalent. Please make sure that your instrument is capable of detecting this fluorochrome.

Applications tested
Streptavidin Super Bright 645 Conjugate has been tested by flow cytometric analysis to detect biotinylated primary antibodies. The amount of this conjugate that will stain a cell sample in a final volume of 100 μL is less than or equal to 0.125 μg. Cell number should be determined empirically but can range from 105 to 108 cells/test. It is recommended that the streptavidin be carefully titrated for optimal performance in the assay of interest.

Samples can be stored in IC Fixation Buffer or 1-step Fix/Lyse Solution for up to three days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Clone specific performance after fixation should be determined empirically.

When using two or more Super Bright dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Complete Staining Buffer for more information.

The Connect platform provides scientists with a secure place to store, analyze, and share data. Applied Biosystems qPCR Analysis Modules within the platform are web browser-based software tools that are flexible, fast, and easy to use. These software tools support a variety of analyses including relative quantitation, standard curve, high resolution melt (HRM), genotyping, and presence/absence. They are compatible with Mac or PC as well as most Applied Biosystems instrument file types.

Format flexibility
The QuantStudio 6 Pro system accommodates the interchange of 96-well (0.2 mL), 96-well (0.1 mL), and 384-well blocks. eBioscience Streptavidin Super Bright 600 is a streptavidin-fluorochrome conjugate for indirect staining protocols to detect biotinylated primary antibodies in flow cytometry. Super Bright dyes are polymer-based fluorophores that are excited by the violet laser (405 nm) and emit fluorescence at the wavelength indicated in their name. They are optimized for use in flow cytometry and allow for use of additional channels with violet excitation. Super Bright dyes are fully compatible with other commonly used fluorescent molecules, eBioscience buffers and fixatives, viability stains, and reagents.

Super Bright 600 is a tandem dye that can be excited with the violet laser line (405 nm) and emits at 600 nm. We recommend using a 610/20 bandpass filter or equivalent. Please make sure that your instrument is capable of detecting this fluorochrome.

Applications tested
Streptavidin Super Bright 600 Conjugate has been tested by flow cytometric analysis to detect biotinylated primary antibodies. The amount of this conjugate that will stain a cell sample in a final volume of 100 μL is less than or equal to 0.125 μg. Cell number should be determined empirically but can range from 105 to 108 cells/test. It is recommended that the streptavidin be carefully titrated for optimal performance in the assay of interest.

Samples can be stored in IC Fixation Buffer or 1-step Fix/Lyse Solution for up to three days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Clone specific performance after fixation should be determined empirically.

When using two or more Super Bright dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Complete Staining Buffer for more information.

Optimized for the ex vivo activation and expansion of regulatory T cells (Tregs), CTS Dynabeads Treg Xpander provides more specific and higher fold expansion compared to our CTS Dynabeads CD3/CD28.
