Human Spheroid-Qualified Hepatocytes (Gibco™)

Gibco Human Spheroid-Qualified Hepatocytes are off-the-shelf cryopreserved human cells for generation of hepatic spheroids in your own lab. Each vial contains a minimum of five million viable cells, sufficient for up to sixteen 96-well plates of hepatic spheroids.

Pharma KingFisher™ Flex 96 Standard Plates (Applied Biosystems™)

These standard plates are for use with the Pharma KingFisher Flex 96 Deep-Well Head Magnetic Particle Processor or MagMAX Express-96 Standard Magnetic Particle Processor in combination with PrepSEQ ResDNA kits and ProteinSEQ kits. The standard configuration differs from the deep-well configuration in that the volume of sample input is from 20–200 µL per well compared to 100–1000 µL per well for the deep-well configuration.

Optimize performance using KingFisher Flex System consumables designed specifically for use with these instruments. KingFisher Flex disposable plastics (tip combs and microplates) are made of polypropylene and are ideal for magnetic particle processing because of their low binding affinity for biomolecules. Their special design enables excellent recovery of magnetic beads.

Activated Human Myofibroblastic Hepatic Stellate Cells (MF-HSCs) (Gibco™)

Gibco Activated Human Myofibroblastic Hepatic Stellate Cells (MF-HSCs) are produced from isolated pure quiescent human stellate cells (q- HSCs). The cells are obtained from a single donor and provided in a cryopreserved format identical to single-donor plateable human hepatocytes.

Each vial contains a minimum of one million viable cells at passage 5, sufficient for at least four 96-well plates in 2D culture.

Simplified protocol
Activated MF-HSCs are suitable for building hepatic co-culture with primary hepatocytes to study in vitro disease models such as hepatic fibrosis. CyQUANT™ LDH Cytotoxicity Assay (Invitrogen™)

The CyQUANT LDH Cytotoxicity Assay is a colorimetric assay that provides a simple and reliable method for determining cellular cytotoxicity. Lactate dehydrogenase (LDH) is a cytosolic enzyme present in many different cell types that is released into the cell culture medium upon damage to the plasma membrane. The CyQUANT LDH Cytotoxicity Assay provides the reagents to accurately and quantitatively measure this extracellular LDH.

Note: The CyQUANT LDH Cytotoxicity Assay (Cat. Nos. C20300 and C20301) are direct replacements for the Pierce LDH Cytotoxicity Assay Kit (Cat. Nos. 88953 and 88954).

CyQUANT LDH Cytotoxicity Assay features include:

LDH is a cytosolic enzyme present in many different cell types and is a well-established and reliable indicator of cellular toxicity. Damage of the plasma membrane results in a release of LDH into the surrounding cell culture medium. This extracellular LDH can be quantified by a coupled enzymatic reaction in which LDH catalyzes the conversion of lactate to pyruvate via NAD+ reduction to NADH. Diaphorase then uses NADH to reduce a tetrazolium salt (INT) to a red formazan product that can be measured at 490 nm. The level of formazan formation is directly proportional to the amount of LDH released into the medium.

The CyQUANT LDH Cytotoxicity Assay provides the reagents needed for the simple, reliable colorimetric quantification of cellular cytotoxicity. The kit can be used with different cell types, including 3D cell models, to measure cytotoxicity mediated by chemical compounds as well as cell-mediated cytotoxicity. Since LDH in the medium is the indicator of cellular cytotoxicity, the assay can be used to monitor cytotoxicity from the same sample over time. To perform the assay, an aliquot of the cell culture medium is transferred to a new plate and the reaction mixture is added. After a 30-minute incubation, the reaction is stopped by adding Stop Solution and absorbance is measured using a microplate reader.

Pharma KingFisher™ Flex 96 Deep-Well Tip Combs (Applied Biosystems™)

These tip combs are for use with the Pharma KingFisher Flex 96 Deep-Well Head Magnetic Particle Processor or MagMAX Express-96 Deep-Well Magnetic Particle Processor in combination with PrepSEQ ResDNA kits and ProteinSEQ kits.

Optimize performance using KingFisher Flex System consumables designed specifically for use with these instruments. KingFisher Flex disposable plastics (tip combs and microplates) are made of polypropylene and are ideal for magnetic particle processing because of their low binding affinity for biomolecules. Phusion™ Hot Start II DNA Polymerase & dNTP Mix (10 mM each) (Thermo Scientific™)

This package includes Phusion Hot Start II High-Fidelity DNA Polymerase (4 x 500 U) and high quality dNTP Mix (10 mM each, 2 x 1 mL).

Phusion Hot Start II High-Fidelity DNA Polymerase
Thermo Scientific Phusion High-Fidelity DNA polymerases set a high standard for high performance PCR. Featuring an error rate 52-fold lower than that of Taq, and 6-fold lower than that of Pfu, they are an excellent choice for cloning and other applications requiring high fidelity. They offer robust performance with short protocol times, even in the presence of PCR inhibitors, and generate higher yields with lower enzyme amounts than other DNA polymerases.

Phusion Hot Start II High-Fidelity DNA Polymerase is immediately reactivated at high temperatures, so does not require a separate activation step in PCR protocols.

dNTP Mix
Thermo Scientific dNTP Mix contains pre-mixed aqueous solutions of dATP, dCTP, dGTP, and dTTP, each at a final concentration of 10 mM. The nucleotides have greater than 99% purity and are free of nuclease activities, as well as human and E. coli DNA. Mixes offer the possibility of reducing the number of pipetting steps and the risk of reaction setup errors.

Platinum™ Taq DNA Polymerase, DNA-free (Invitrogen™)

Invitrogen Platinum Taq DNA Polymerase, DNA-free, is manufactured using a novel, closed single-use system together with stringent quality-control testing to minimize the risk of contamination with bacterial and human DNA. This enzyme is an ideal choice for PCR-based applications requiring the highest sensitivity without false-positive results from reagent-borne contamination. This DNA-free enzyme provides the same high level of performance and lot-to-lot consistency as standard Platinum Taq DNA Polymerase.

DNA-free Platinum Taq DNA Polymerase offers:
• Certified low-DNA contamination level (≤0.01 copy of bacterial gDNA per enzyme unit)

Platinum Taq DNA Polymerase, DNA-free, is a recombinant Taq polymerase complexed with a proprietary antibody that blocks polymerase activity at ambient temperatures and dissociates after the initial denaturation step at 94°C. This automatic “hot start” provides increased sensitivity, specificity, and yield, while allowing convenient reaction assembly at room temperature. Just as with Taq DNA Polymerase, Platinum Taq DNA Polymerase, DNA-free, has a non-template–dependent terminal transferase activity that adds a 3′ deoxyadenosine to product ends, and has a 5′→3′ exonuclease activity.

It is designed to increase the dynamic range of the side-scatter detection without sacrificing resolution of the detector. The use of this 488/10 side scatter filter combined with ultra-filtering of the focusing fluid has been found to dramatically decrease background noise and enable discrimination of 100-nm particles.

Easy to use
This filter is easily swapped for the standard side-scatter filter. It cannot be installed the wrong way because the optical block is keyed to accept only one orientation of the filter blade. The filter also has a color-coded label to help identify the correct position.

Attune™ NxT Maintenance Kit (Invitrogen™)

Attune™ NxT Flow Cell Cleaning Solution (Invitrogen™)

The quality of your flow cytometry data can be affected by the cleanliness of the flow cell. Attune NxT Flow Cell Cleaning Solution is an alkaline liquid concentrate that, when mixed with water, removes contaminants without damaging the quartz glass used in the optical system of the Attune NxT Acoustic Focusing Cytometer.

Attune NxT Flow Cell Cleaning Solution has been formulated to minimize the contamination build-up that may occur in the system lines or flow cell after repeated usage. This solution is recommended for use during the SIP sanitize function just prior to the shutdown procedure. The recommended frequency of use depends upon the number of samples analyzed on the instrument. For high volume users, it is recommended as a daily preventive measure. For less frequent users, weekly use is recommended.
