MMP 3-Plex Human ProcartaPlex™ Panel 2 (Invitrogen™)

ProcartaPlex immunoassays are an ideal solution for assessing multiple protein biomarkers in a single sample. Luminex xMAP technology is used for the multi-analyte detection of secreted proteins including cytokines, chemokines, growth factors and other proteins. ProcartaPlex assays are subject to the same stringent and robust development and manufacturing as are our extensively validated Platinum and Instant ELISA kits. HCT 116 (+hIGF-1) Control Lysate (1mg/mL) (Thermo Scientific™)

Thermo Scientific HCT 116 (+hIGF-1) Control Lysate is a stimulated cell lysate that has been validated for the expression levels and phosphorylation of key targets in the RAS signaling pathway.

HCT 116 (+hIGF-1) Control Lysate features include:

The RAS family of GTPase proteins modulates two key signaling pathways involved in cell growth and proliferation: the MAPK and PI3K pathways. Aberrant activation of RAS proteins promotes constitutive activation of cell growth and proliferation that can result in cancer. Mutations in the NRAS, KRAS, and HRAS oncogenes are the most common mutations in human cancer. This lysate has been validated for expression levels of the three isoforms of RAS using mass spectrometry.

The HCT 116 cell line is a human colon cancer epithelial cell line and has been used for proliferation and tumorigenicity studies for colon cancer. The HCT 116 (+hIGF-1) cell lysate is provided as frozen lysate at a standard concentration of 1 mg/mL (500 mg per vial). Lysates are prepared in non-denaturing buffer without loading dyes or dissociating agents (i.e., no 2-mercaptoethanol or dithiothreitol has been added). For western blot analysis, heat lysate to 95°C for 5 minutes. Add reducing agent before heating if desired. The recommended loading volume per lane is 10–20 μL (10–20 µg).

The PluriTest Assay is a bioinformatics assay that compares the transcriptional profile of a sample to an extensive reference set of >450 cell/tissue types, including 223 hESC lines, 41 iPSC lines, somatic cells, and tissues. The online analysis tool confirms pluripotency marker expression via two separate scores: Pluripotency and Novelty. The Pluripotency Score informs the user how strongly a model-based pluripotency signature is expressed in the samples analyzed. The Novelty Score indicates the general model fit for a given sample.

RAGE Human ProcartaPlex™ Simplex Kit (Invitrogen™)

The Human RAGE Simplex ProcartaPlex kit measures RAGE protein and is designed to be combinable with other Simplex kits so that you can create your own multiplex panel that utilizes Luminex xMAP technology for protein detection/quantitation. When combining multiple Simplex kits (i.e., when you are not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed for each assay plate regardless of plex size.

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to 80 protein targets simultaneously when using the FLEXMAP 3D or Luminex 200 instrument and up to 50 protein targets when using the MAGPIX instrument. ProcartaPlex assays require as little as 25 µL of plasma or serum, or 50 µL of cell culture supernatant, and just four hours to obtain analyzed results.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a ProcartaPlex multiplex assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile up to 80 times more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

Variant Reporter™ v3 Upgrade from v2 (Applied Biosystems™)

This is the v2 to v3 upgrade for Variant Reporter Software. This software performs comparative sequencing, also known as direct sequencing, medical sequencing, PCR sequencing, and resequencing with DNA sequencing files. The software is designed for reference-based and non-reference-based analysis such as mutation detection and analysis, SNP discovery and validation, and sequence confirmation. The robust algorithms will call SNPs, mutations, insertions, deletions, and heterozygous insertions⁄deletions for data generated using the Applied Biosystems genetic analyzers.
